프로바이오틱스 기반 구강 유해균 억제 및 치주염 치료전략 탐색 김태민 지도교수: 이건호 글로벌바이오융합학과 조선대학교 대학원 만성 치주염(chronic periodontitis)은 치아 지지 조직의 비가역적 파괴를 초래할 뿐만 아니라, 지속적인 감염원으로 작용하여 전신적인 염증 부담을 가중시키는 심각한 구강 질환이다. 구강 내 주요 유해균 중 Porphyromonas gingivalis가 치주염 발병을 주도하는 핵심 유해균(keystone pathogen)으로 작용한다면, Enterococcus faecalis는 복합 미생물 감염(polymicrobial infection) 환경에서 전체 미생물 군집의 병원성을 악화시키는 유해균(driver pathogen)으로 질환을 심화시킨다. 그러나 기존의 항생제는 추가적인 미생물 불균형과 항생제 내성을 유발하므로, 유해균만을 선택적으로 제어하는 정밀 항균(precision antimicrobial) 전략의 필요성이 대두되고 있다. 이에 본 연구 에서는 594개의 프로바이오틱스 라이브러리를 스크리닝하여, 치주염 핵심 유 해균들을 표적으로 하는 맞춤형 항균 물질을 발굴 및 규명하였다. 따라서, 그람 양성균인 E. faecalis에 대해 특이적으로 강력한 억제 활성 을 보이는 Enterococcus faecium 균주를 분리하였다. 생화학적 특성 분석 및 LC-MS/MS 분석을 통해, 이 활성 인자(active effector)가 43개의 아미노 산으로 구성된 새로운 박테리오신(bacteriocin)임을 규명하였다. 새로운 박테 리오신은 뚜렷한 알파 나선(alpha-helical) 구조를 형성하는 것으로 예측되었 으며, 구강 유래 E. faecalis 균주들에 대해 기존에 사용되고 있는 항생제인 미노사이클린(minocycline)보다 우수한 항균 효능을 보였다. 그러나 좁은 항 균 스펙트럼을 가진 이 박테리오신은 P. gingivalis를 포함한 그람 음성 유해 균에 대해서는 억제 효능을 나타내지 않았다. 따라서, 치주염을 유발하는 그람 음성 유해균인 P. gingivalis를 직접적으 로 제어하기 위해 추가적인 스크리닝을 수행한 결과, P. gingivalis를 억제할 수 있는 Limosilactobacillus reuteri 균주를 발굴하였다. 생화학적 분석 결과, L. reuteri가 분비하는 항균 인자는 열에 안정적이고 단백질 분해 효소에 저 항성(protease-resistant)을 지닌 비단백질성 분자(non-proteinaceous molecule)임이 확인되었다. 본 연구는 이러한 비단백질성 분자가 만성 치주 염 관련 주요 유해균인 P. gingivalis를 효과적으로 제어할 수 있음을 입증하 였다. 종합적으로 본 연구의 결과는 발굴된 프로바이오틱스 유래 물질들이 치주 염 관련 주요 유해균들에 대해 효과적인 항균 활성을 지니고 있음을 입증하 였다. 이러한 박테리오신 및 비단백질성 분자의 발굴은 주요 유해균을 개별 적으로 정밀 타겟팅하는 프로바이오틱스 기반 치주염 치료 전략에 대한 강력 한 in vitro 토대를 제공한다. 또한, 궁극적으로 이러한 표적 항균 물질들은 새로운 치주염 치료제 개발을 위한 유망한 선도 후보 물질(lead candidates) 로서, 향후 정확한 물질 규명 및 효능 검증을 위한 in vivo 연구의 필요성을 제시한다.

Probiotic-Based Approaches for the Control of Oral Pathogenic Bacteria and Treatment Tae-Min Kim Advisor: Prof. Kun Ho Lee Department of Integrative Biological Sciences Graduate School of Chosun University Chronic periodontitis is a severe oral disease that not only causes irreversible destruction of tooth-supporting tissues but also acts as a continuous reservoir of infection, exacerbating the systemic inflammatory burden. Among the major oral pathogens, Porphyromonas gingivalis acts as a keystone pathogen that initiates the pathogenesis of periodontitis, while Enterococcus faecalis functions as a driver pathogen that amplifies the virulence of the entire microbial community within a polymicrobial infection environment. However, since conventional antibiotics induce further dysbiosis and antimicrobial resistance, there is an emerging need for precision antimicrobial strategies that selectively control only these pathogenic bacteria. Accordingly, this study screened a library of 594 probiotics to discover and characterize tailored antimicrobial substances targeting these key periodontal pathogens. As a result, an Enterococcus faecium strain exhibiting specifically potent inhibitory activity against the Gram-positive E. faecalis was isolated. Through biochemical characterization and LC-MS/MS analysis, this active effector was identified as a novel bacteriocin composed of 43 amino acids. The novel bacteriocin was predicted to form a distinct alpha-helical structure and demonstrated superior antimicrobial efficacy against oral-derived E. faecalis strains compared to minocycline, a conventionally used antibiotic. However, due to its narrow antimicrobial spectrum, this bacteriocin did not exhibit inhibitory efficacy against Gram-negative pathogens, including P. gingivalis. Therefore, to directly control the Gram-negative periodontopathogen P. gingivalis, an additional screening was performed, resulting in the discovery of a Limosilactobacillus reuteri strain capable of inhibiting P. gingivalis. Biochemical analysis revealed that the antimicrobial factor secreted by L. reuteri is a heat-stable, protease-resistant, non-proteinaceous molecule. This study demonstrated that this non-proteinaceous molecule can effectively control P. gingivalis, a major pathogen associated with chronic periodontitis. In conclusion, the results of this study demonstrate that the discovered probiotic-derived substances possess effective antimicrobial activity against major periodontitis-associated pathogens. The discovery of this bacteriocin and non-proteinaceous molecule provides a robust in vitro foundation for probiotic-based therapeutic strategies for periodontitis that individually and precisely target key pathogens. Ultimately, these targeted antimicrobial substances serve as promising lead candidates for the development of novel periodontitis treatments, highlighting the need for future in vivo studies to determine their precise structures and validate their clinical efficacy.
