    도라지는 일본, 한국, 중국, 몽골과 같은 동아시아 국가들에서 2000년 이상 기침, 가래, 목 아픔, 폐 농양, 가슴 답답함 등의 호흡기 질환 증상에 전통적인 치료 요법으로 사용해 왔다. 도라지의 대표적인 생리활성물질로는 트리테르페노이드 사포닌 계열인 플라티코사이드가 알려져 있다. 도라지 내의 플라티코사이드는 재배 지역, 수확 시기, 재배 기간에 따라 함량이 다르며, 현재까지 70여 종의 플라티코사이드가 분리, 동정되었다. 플라티코사이드는 5환성의 올레난 구조의 3번 탄소와 28번 탄소에 glucose, laminaribiose, gentiobiose, arabinose, rhamnose, xylose, apiose 등의 당이 결합된 다양한 형태로 존재하며, 이에 따라 플라티코사이드의 생리활성 또는 장내 흡수율이 다르다. Platycodin D는 도라지의 플라티코사이드 중 많은 양을 차지하며 도라지의 생리활성을 나타내는 대표적인 물질로 한국에서 품질 관리의 지표 물질로 설정되어 있다. Platycodin D의 생리활성으로 항염증, 항종양, 항당뇨, 항비만, 항고지혈증, C형 간염 바이러스 복제 저해, 알레르기 관련 질병들의 저감화 등의 효과가 알려져 있다.
    사포닌의 당 잔기는 생물전환을 이용하여 제거할 수 있으며, 당 잔기를 제거함으로 원하는 생리활성물질로 전환할 수 있다. 서로 다른 종류나 개수의 당 잔기를 가지는 동일한 아글리콘 골격의 사포닌 물질들은 서로 다른 생리활성이나 생체이용률을 나타낸다. 또한 사포닌 아글리콘 골격의 작용기의 변화에 따라서도 그 물질의 생리활성이나 생체이용률, 장내 미생물에 의한 대사가 변화한다. 인삼의 진세노사이드의 경우 이러한 당이나 작용기의 전환에 대한 연구가 많이 보고되어 있다. 하지만 도라지의 플라티코사이드의 경우 이러한 연구가 거의 되어있지 않은 상태이다. 따라서 본 연구에서는 식용이 가능한 미생물 균주들을 이용하여 도라지 내의 사포닌 계열의 생리활성물질인 플라티코사이드의 당 잔기 혹은 작용기를 전환하여 생리활성이나 생체이용률이 높은 대사체를 만들고, 그 물질을 효율적으로 생산할 수 있는 방법을 탐색하며, 생산한 대사체를 분리하고 구조를 확인하고자 하였다.
    연구 1에서는 생물전환을 이용하여 도라지 내의 주요 생리활성물질인 platycodin D의 양을 증가시키기 위하여 platycodin D의 전구체인 platycoside E와 platycodin D3를 platycodin D로의 전환에 필요한 β-glucosidase 활성이 높은 식용 미생물을 탐색하고 생물전환 조건을 최적화하고자 하였다. 11개의 식용 미생물을 대상으로 실험한 결과, Aspergillus usamii의 β-glucosidase 활성이 가장 높았으며, 실제 기질인 platycoside E와 platycodin D3의 β-1, 6-glucosidic 결합을 분해하여 platycodin D로 전환하였다. 전환 효율을 높이기 위하여 조건을 최적화한 결과, 탄소원으로 cellobiose를 사용하였을 때 A. usamii의 β-glucosidase 활성이 4배 이상 높아졌으며, 그 조효소액을 사용하였을 때 platycoside E와 platycodin D3를 2시간 안에 99.9% 이상 platycodin D로 전환하여 platycodin D의 양이 2배 이상 증가하였다.
    연구 2에서는 platycodin D의 생리활성이나 생체이용률을 높이기 위하여 식용 미생물 균주의 효소 활성을 이용하여 platycodin D의 당 잔기를 제거한 대사체들을 만들고 그 구조를 분석하고자 하였다. 이를 위하여 여러 식용이 가능한 미생물을 탐색하여 platycodin D의 생물전환에 필요한 β-glucosidase, β-xylosidase, α-rhamnosidase, esterase 활성이 높은 A. niger와 A. usamii를 선발하였다. 이들 균에 대하여 각각의 효소 활성을 선택적으로 높일 수 있는 탄소원을 탐색하여 cellobiose, xylose, rhamnose를 선정하였으며, 이 당을 첨가하여 배양한 각각의 조효소액을 이용하여 platycodin D를 전환하였다. HPLC와 electrospray ionization triple quadrupole LC/MS를 이용하여 분석한 결과, 전환한 대사체는 deapiosyl-xylosylated platycodin D, deapiosyl-xylosyl-rhamnosylated platycodin D, prosapogenin D, deglucosylated platycodin D, deglucosyl-apioxyl-xylosylated platycodin D, deglucosyl-apioxyl-xylosyl-rhamnosylated platycodin D, platycodigenin임을 확인하였다.
    연구 3에서는 platycodin D를 A. niger의 조효소액으로 전환하여 생산한 신규 물질의 구조를 분석하고 그 물질의 생리활성을 platycodin D와 비교·분석하였다. Platycodin D의 당 잔기의 전환에 필요한 효소 활성을 증가시키기 위하여 A. niger의 배양 시 도라지의 사포닌 분획을 첨가한 경우 산화효소 활성이 유도됨을 발견하였다. 이 조효소액을 이용하면 platycodin D가 기존의 당이 제거된 platycodin D 대사체가 아니라 신규 물질로 전환되는 것을 HPLC로 확인하였다. LC/MS와 NMR을 이용하여 구조를 확인한 결과 이 신규 물질은 platycodin D의 히드록시기가 산화되어 케톤기로 변화된 16-oxo-platycodin D인 것을 알 수 있었다. A. niger의 배양 시 도라지 사포닌 분획을 첨가한 조효소액으로 platycodin D를 전환하였을 때, platycodin D는 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, 16-oxo-prosapogenin D 순으로 전환되었다. 온도와 pH에 대한 전환조건 최적화로 platycodin D를 16-oxo-platycodin D로 2시간 내에 99.9% 이상 전환할 수 있었다. Platycodin D와 16-oxo-platycodin D의 구조적인 변화에 따른 생리활성의 변화를 확인하기 위해서 이들의 세포 증식 저해 활성을 확인한 결과 16-oxo-platycodin D는 정상 세포주인 NCTC 1469와 FHC에서 platycodin D와 비교하여 세포 증식 저해 활성이 8배 이상 줄어들었다.
    본 연구에서는 도라지에 함유되어 있는 플라티코사이드를 생물전환하여 생리활성과 생체이용률이 높은 대사체를 만들기 위하여 식용 미생물들을 탐색하여 A. niger와 A. usamii를 선발하고 생물전환 조건을 최적화하였다. 그 결과 도라지 내의 platycoside E와 platycodin D3를 A. usamii의 β-glucosidase 활성을 이용하여 전환하여 도라지의 생리활성을 나타내는 platycodin D의 함량을 2배 이상 증가시킬 수 있었다. 또한 A. niger와 A. usamii의 배양 시 탄소원의 변화를 주어 β-glucosidase, β-xylosidase, α-rhamnosidase 활성을 선택적으로 높이고 이를 이용하여 platycodin D의 당 잔기가 전환된 7가지의 대사체를 생산하였다. 한편, A. niger의 배양 시 도라지의 사포닌 분획의 첨가로 산화효소 활성을 유도하였으며, 이를 이용하여 platycodin D를 신규 물질인 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, 16-oxo-prosapogenin D로 전환하였다. 전환한 16-oxo-platycodin D는 platycodin D에 비해서 낮은 세포 증식 저해 활성을 나타내었다. 본 연구를 통하여 선발한 균주를 이용한 도라지 플라티코사이드의 생물전환 대사체 생산 기술은 도라지 내의 기능성 물질의 생리활성 및 생체이용률을 높여 도라지의 기능성 식품 소재로의 활용에 적용될 수 있을 것이다. 향후 각 대사체의 효율적인 생산과 이를 이용한 생리활성 및 생체이용률에 대한 추가적인 연구가 필요하다.

    Platycodi Radix has been used as a traditional herbal medicine to prevent and treat respiratory diseases such as cough, excessive phlegm, sore throat, bronchitis, lung abscess, and chest pain in China, Japan, Korea, and Mongolia for thousands of years. Bioactive chemical compounds in Platycodi Radix are known to be platycosides, one of triterpenoid saponin family. Total platycosides content of PR may be dependent on Platycodon grandiflorum’s soil, weather, cultivation method, and years of growth. About 70 varieties of platycosides have been identified to date. Platycosides consist of an olenane type triterpenoid backbone with two side chains linked with various sugar moieties such as glucose, laminaribiose, arabinose, rhamnose, xylose, and apiose at C-3 and C-28. Depending on the type of sugar moieties, platycosides have different bioactivity and bioavailability. Among these platycosides, platycodin D is a major and representative triterpenoid saponin which shows bioactivities in Platycodi Radix and is the indicator substance for quality management of Platycodi Radix in Korea. Platycodin D has been reported to have diverse pharmacological activities including anti-inflammatory, anti-diabetic, anti-cancer, anti-obesity, and anti-hyperlipidemic effects, inhibition of hepatitis C virus replication, and reduction of allergy-related diseases.
    Saponin can be converted into desired bioactive substances by removing sugar residues. Saponins with the same backbone with different types or numbers of sugar moieties exhibit different bioactivities or bioavailability. Also, saponin’s bioactivity, bioavailability, and metabolisms by intestinal bacteria can be different according to changes in the functional groups of the saponin backbone.
    In the case of ginsenosides of ginseng, many studies have been reported on the conversion of these sugars or functional groups of ginsenosides. However, transformations of sugar moieties and functional groups of platycosides have not been studied. Therefore, we tried to investigate effective methods for bioconversion of platycosides changing their sugar moieties or functional groups into new metabolites which may have increased bioactivities or bioavailability than the precursors using food-grade microorganisms, and identify the transformed molecules.
    The purpose of Study 1 was to increase the amount of platycodin D, a major bioactive substance in Platycodi Radix, using biotransformation. Food grade microorganisms with high β-glucosidase activity required for bioconversion of platycoside E and platycodin D3, two major platycodin D precursors, into platycodin D were screened, and the biotransformation condition was optimized. Among 11 microorganism strains tested, Aspergillus usamii had the highest β-D-glucosidase activity and active properties on bioconversion of platycoside E and platycodin D3 into platycodin D, hydrolyzing β-1, 6-glucosidic bond. Also, as a result of optimizing the culture condition to increase conversion efficiency, β-glucosidase activity of A. usamii was more than 4 times when cellobiose was used as a carbon source. More than 99% of platycoside E and platycodin D3 were converted into platycodin D when treated with the crude enzyme extract with increased β-glucosidase activity of A. usamii for 2 h. As the platycodin D precursors were converted, platycodin D content increased more than twice.
    The purpose of Study 2 was to biotransform platycodin D into metabolites, which have less sugar moieties, using enzyme activities of food grade microorganisms to increase bioactivities or bioavailability, and identification of those metabolites. A. niger and A. usamii were selected for platycodin D transformation due to their relatively high enzyme activities in β-glucosidase, β-xylosidase, α-rhamnosidase, and esterase when food grade microorganisms were tested. To get each of the transformed molecules from platycodin D, appropriate enzymes were induced via different carbon sources such as xylose, rhamnose, and cellobiose in the culture media. Using various crude enzyme extracts, platycodin D was transformed into 7 kinds of metabolites. Deapiosyl-xylosylated platycodin D, deapiosyl-xylosyl-rhamnosylated platycodin D, prosapogenin D, deglucosylated platycodin D, deglucosyl-apioxyl-xylosylated platycodin D, deglucosyl apioxyl-xylosyl-rhamnosylated platycodin D, and platycodigenin transformed from platycodin D were identified as platycodin D metabolites by using HPLC and LC/MS.
    The purpose of Study 3 was to analyze structure of a new metabolite from platycodin D produced by crude enzyme extract of A. niger and to compare the bioactivity of the metabolite with that of platycodin D. Platycosides fraction of Platycodi Radix was added in a culture medium of A. niger as an inducer to increase the enzyme activities for hydrolyzing apiose, xylose, rhamnose, arabinose, and glucose of platycosides D. New metabolites were produced by the crude enzyme extract of A. niger cultured with saponin fraction of Platycodi Radix. One of the new metabolites was identified to be 16-oxo-platycodin D, which was converted into a ketone group by oxidation of hydroxyl group of platycodin D, using HPLC, LC/MS, and NMR. Platycodin D was converted into 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, and 16-oxo-prosapogenin D in order over time by the crude enzyme extract of A. niger. The transformation condition such as pH, temperature, and heat stability were optimized so that 99% of platycodin D was transformed into 16-oxo-platycodin D in 2 h. Inhibitory activity of platycodin D and 16-oxo-platycodin D on proliferation of normal cell lines, NCTC 1469 and FHC from mouse and human, respectively, was tested if the physiological activity was changed by changing the hydroxyl group of platycodin D to the ketone group. Compared to platycodin D, 16-oxo-platycodin D showed over 10 times lower inhibitory ability on proliferation of the NCTC 1469 and FHC cell lines.
    In this study platycosides from Platycodi Radix were transformed using edible bacterial and fungal strains into metabolites which ccould have higher bioactivities or bioavailability. Platycosides were converted using crude enzyme extracts of A. niger and A. usamii, which were the most suitable microorganisms for the conversion of platycosides since they had β-apiosidase, β-xylosidase, α-rhamnosidase, β-glucosdise, and esterase activities. Content of platycodin D increased more than twice after the conversion of platycodin D precursors using the β-glucosidase activity of A. usamii. Activities of β-glucosidase, β-xylosidase, and α-rhamnosidase, which were required to hydrolyze glucose, apiose, xylose, and rhamnose of platycodin D, were induced with different carbon sources (cellobiose, xylose, and rhamnose) in the culture media of A. niger and A. usamii. Using various induced crude enzyme extracts, platycodin D was transformed into 7 kinds of metabolites. In addition, oxidase activity of crude enzyme extract of A. niger was induced by platycosides of Platycodi Radix during cultivation. Platycodin D was transformed into 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, and 16-oxo-prosapogenin D by the induced enzyme extract. 16-Oxo-platycodin D showed lower inhibition activity on proliferation of normal cells than platycodin D. These new metabolites from platycodin D produced by our methods may have new bioactivities or higher bioavailability. These results can be a new way to improve the quality of Platycodi Radix as a functional food. In the next study, the investigation on the change of bioactivities and bioavailability of platycodin D metabolites by their structural change compared to platycodin D is needed.

    Platycodi Radix has been used as a traditional herbal medicine to prevent and treat respiratory diseases such as cough, excessive phlegm, sore throat, bronchitis, lung abscess, and chest pain in China, Japan, Korea, and Mongolia for thousands of years. Bioactive chemical compounds in Platycodi Radix are known to be platycosides, one of triterpenoid saponin family. Total platycosides content of PR may be dependent on Platycodon grandiflorum’s soil, weather, cultivation method, and years of growth. About 70 varieties of platycosides have been identified to date. Platycosides consist of an olenane type triterpenoid backbone with two side chains linked with various sugar moieties such as glucose, laminaribiose, arabinose, rhamnose, xylose, and apiose at C-3 and C-28. Depending on the type of sugar moieties, platycosides have different bioactivity and bioavailability. Among these platycosides, platycodin D is a major and representative triterpenoid saponin which shows bioactivities in Platycodi Radix and is the indicator substance for quality management of Platycodi Radix in Korea. Platycodin D has been reported to have diverse pharmacological activities including anti-inflammatory, anti-diabetic, anti-cancer, anti-obesity, and anti-hyperlipidemic effects, inhibition of hepatitis C virus replication, and reduction of allergy-related diseases.
    Saponin can be converted into desired bioactive substances by removing sugar residues. Saponins with the same backbone with different types or numbers of sugar moieties exhibit different bioactivities or bioavailability. Also, saponin’s bioactivity, bioavailability, and metabolisms by intestinal bacteria can be different according to changes in the functional groups of the saponin backbone.
    In the case of ginsenosides of ginseng, many studies have been reported on the conversion of these sugars or functional groups of ginsenosides. However, transformations of sugar moieties and functional groups of platycosides have not been studied. Therefore, we tried to investigate effective methods for bioconversion of platycosides changing their sugar moieties or functional groups into new metabolites which may have increased bioactivities or bioavailability than the precursors using food-grade microorganisms, and identify the transformed molecules.
    The purpose of Study 1 was to increase the amount of platycodin D, a major bioactive substance in Platycodi Radix, using biotransformation. Food grade microorganisms with high β-glucosidase activity required for bioconversion of platycoside E and platycodin D3, two major platycodin D precursors, into platycodin D were screened, and the biotransformation condition was optimized. Among 11 microorganism strains tested, Aspergillus usamii had the highest β-D-glucosidase activity and active properties on bioconversion of platycoside E and platycodin D3 into platycodin D, hydrolyzing β-1, 6-glucosidic bond. Also, as a result of optimizing the culture condition to increase conversion efficiency, β-glucosidase activity of A. usamii was more than 4 times when cellobiose was used as a carbon source. More than 99% of platycoside E and platycodin D3 were converted into platycodin D when treated with the crude enzyme extract with increased β-glucosidase activity of A. usamii for 2 h. As the platycodin D precursors were converted, platycodin D content increased more than twice.
    The purpose of Study 2 was to biotransform platycodin D into metabolites, which have less sugar moieties, using enzyme activities of food grade microorganisms to increase bioactivities or bioavailability, and identification of those metabolites. A. niger and A. usamii were selected for platycodin D transformation due to their relatively high enzyme activities in β-glucosidase, β-xylosidase, α-rhamnosidase, and esterase when food grade microorganisms were tested. To get each of the transformed molecules from platycodin D, appropriate enzymes were induced via different carbon sources such as xylose, rhamnose, and cellobiose in the culture media. Using various crude enzyme extracts, platycodin D was transformed into 7 kinds of metabolites. Deapiosyl-xylosylated platycodin D, deapiosyl-xylosyl-rhamnosylated platycodin D, prosapogenin D, deglucosylated platycodin D, deglucosyl-apioxyl-xylosylated platycodin D, deglucosyl apioxyl-xylosyl-rhamnosylated platycodin D, and platycodigenin transformed from platycodin D were identified as platycodin D metabolites by using HPLC and LC/MS.
    The purpose of Study 3 was to analyze structure of a new metabolite from platycodin D produced by crude enzyme extract of A. niger and to compare the bioactivity of the metabolite with that of platycodin D. Platycosides fraction of Platycodi Radix was added in a culture medium of A. niger as an inducer to increase the enzyme activities for hydrolyzing apiose, xylose, rhamnose, arabinose, and glucose of platycosides D. New metabolites were produced by the crude enzyme extract of A. niger cultured with saponin fraction of Platycodi Radix. One of the new metabolites was identified to be 16-oxo-platycodin D, which was converted into a ketone group by oxidation of hydroxyl group of platycodin D, using HPLC, LC/MS, and NMR. Platycodin D was converted into 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, and 16-oxo-prosapogenin D in order over time by the crude enzyme extract of A. niger. The transformation condition such as pH, temperature, and heat stability were optimized so that 99% of platycodin D was transformed into 16-oxo-platycodin D in 2 h. Inhibitory activity of platycodin D and 16-oxo-platycodin D on proliferation of normal cell lines, NCTC 1469 and FHC from mouse and human, respectively, was tested if the physiological activity was changed by changing the hydroxyl group of platycodin D to the ketone group. Compared to platycodin D, 16-oxo-platycodin D showed over 10 times lower inhibitory ability on proliferation of the NCTC 1469 and FHC cell lines.
    In this study platycosides from Platycodi Radix were transformed using edible bacterial and fungal strains into metabolites which ccould have higher bioactivities or bioavailability. Platycosides were converted using crude enzyme extracts of A. niger and A. usamii, which were the most suitable microorganisms for the conversion of platycosides since they had β-apiosidase, β-xylosidase, α-rhamnosidase, β-glucosdise, and esterase activities. Content of platycodin D increased more than twice after the conversion of platycodin D precursors using the β-glucosidase activity of A. usamii. Activities of β-glucosidase, β-xylosidase, and α-rhamnosidase, which were required to hydrolyze glucose, apiose, xylose, and rhamnose of platycodin D, were induced with different carbon sources (cellobiose, xylose, and rhamnose) in the culture media of A. niger and A. usamii. Using various induced crude enzyme extracts, platycodin D was transformed into 7 kinds of metabolites. In addition, oxidase activity of crude enzyme extract of A. niger was induced by platycosides of Platycodi Radix during cultivation. Platycodin D was transformed into 16-oxo-platycodin D, 16-oxo-deapiosylated platycodin D, and 16-oxo-prosapogenin D by the induced enzyme extract. 16-Oxo-platycodin D showed lower inhibition activity on proliferation of normal cells than platycodin D. These new metabolites from platycodin D produced by our methods may have new bioactivities or higher bioavailability. These results can be a new way to improve the quality of Platycodi Radix as a functional food. In the next study, the investigation on the change of bioactivities and bioavailability of platycodin D metabolites by their structural change compared to platycodin D is needed.
