    생명공학기술과 함께 많은 발전을 이루고 있는 바이오 의약품인 치료용 재조합 단백질 (therapeutic recombinant proteins)과 단클론항체 (monoclonal antibodies)의 생산에는 Chinese Hamster Ovary (CHO) 세포주가 널리 사용되고 있다. 생성물은이 당그룹이 연결되어 있는 복잡한 단백질이기 때문에 단백질 합성뿐만 아니라 번역 후 수정(post-translational modification)을 잘 수행할 수 있는 안전한 동물세포를 이용하는 것이 제품의 품질면에 있어서 중요하기 때문이다. 그리고 CHO 세포주에서 단백질의 높은 생산성을 위해서, 특정 세포주와 생산되는 재조합 단백질의 특성에 따라서 동물유래성분을 포함하지 않은, 화학적으로 성분이 명확한 화학 조성 배지 (Chemically Defined Medium, CDM)를 개발하는 것은 높은 생산성과 고품질의 생산 공정 설계에 있어 중요하다. 본 연구에서는 CDM 조성 및 농도를 최적화하기 위해서 Design of Experiment (DoE)을 사용하여 자체 화학 조성 배지 내 vitamins, lipids, polyamines, nucleoside, Pluronic F68, 아미노산에 대하여 세포 성장 및 생산성에 대한 영향을 평가하였다. screening DoE로 PBD (Plackett-Burman design)를 사용했으며, Characterization을 위해 Optimization 단계를 거쳐 Sequential DoE를 진행하였다. 유의미한 요인들의 농도를 재설정하는 단계를 거쳐 배지를 최적화하였다. 결과적으로, 기존의 자체화학조성 배지와 비교하여 세포 농도와 생산량을 1.5배 이상 향상 시키는 결과를 얻을 수 있었다.

    Recombinant therapeutic proteins produced in CHO cells represent a major class of biopharmaceuticals. The reason for this wide use of CHO cells is mainly their ability to perform post-translational modifications and correctly fold recombinant proteins, as well as their proven track record of being safe hosts. To produce high recombinant protein yields in CHO cells, media development is an integral step for this optimal growth environment for cells and a fundamental part in the design of upstream cell culture processes. Additionally, many industrial-scale production techniques now use Chemically Defined Media (CDM) to facilitate downstream processing and reduce the risk of adventitious infectious agents and contaminants. This study focuses on developing the CDM for recombinant CHO cells using Design of Experiments (DoE) regarding Peak Viable Cell Density (PVCD) and production (Titer). Identify the impact of vitamins and lipids, polyamines, nucleosides, Pluronic F68 were selected using Plackett-Burman Design (PBD). Subsequently, a Fractional Factorial Design (FFD) and Central Composite Design (CCD) were used to identify optimal levels and interaction among effective groups of medium components. In the optimal condition, the cells successfully reached PVCD and productivity was 1.5-fold improvement over the control culture. Our findings contribute to more efficient way for optimizing the effective fomulation and to attain higher growth and productivity to develop CDM in CHO cells.
