Comparison of In vivo and In vitro Techniques for Methane Production from Ruminant Diets

This study was conducted to compare the methane ($CH_4$) production estimated by in vivo (sulfur hexafluoride tracer technique ($SF_6$)) with that of two in vitro rumen simulation (RUSITEC) and gas production (IVGPT)) techniques. Four adult dry Holstein cows, aged $7.4{\pm}3.0$ years and weighing $697{\pm}70$ kg, were used for measuring methane production from five diets by the $SF_6$ technique. The experimental diets were alfalfa hay ($D_1$), corn silage + soybean meal (SBM) (910: 90, $D_2$), Italian rye grass hay +SBM (920: 80, $D_3$), rice straw +SBM (910: 90, $D_4$) and Sudan grass hay +SBM (920: 80, $D_5$). Each diet was individually fed to all 4 cows and 5 feeding studies of 17 d each were conducted to measure the methane production. In the RUSITEC, methane production was measured from triplicate vessels for each diet .In vitro gas production was measured for each of the diets in triplicate syringes. The gas produced after 24 and 48 h was recorded and gas samples were collected in vacuum vials and the methane production was calculated after correction for standard temperature and pressure (STP). Compared to the $SF_6$ technique, estimates of methane production using the RUSITEC were lower for all diets. Methane production estimated from 24 h in vitro gas production was higher (p<0.001) on $D_1$ as compared to that measured by $SF_6$, whereas on $D_2$ to $D_5$ it was lower. Compared to $SF_6$, methane production estimated from 48 h in vitro gas production was higher on all diets. However, methane estimated from the mean of the two measurement intervals (24+48 h/2) in IVGPT was very close to that of $SF_6$ (correlation 0.98), except on $D_1$. The results of our study confirmed that IVGPT is reflective of in vivo conditions, so that it could be used to generate a database on methane production potential of various ruminant diets and to examine strategies to modify methane emissions by ruminants.
Nutrition of Range Goats in a Shrubland of Western India

An investigation was made to evaluate the native range of western India in terms of nutrient supply during different physiological stages in goats. One hundred female and 5 male Kutchi adult goats were grazed on a 35 ha plot of native range. They were maintained on sole grazing. Four experiments, one in a defined physiological stage was carried out. Stage I, when maximum number of females were bred; stage II, when bucks were used for breeding purpose; stage III, 3-months old (weaner) kids were allowed to graze on ranges and stage IV, when the goats were in early stage of lactation. During oestrous period, diet contained 15.2% crude protein (CP), 54.0% neutral detergent fibre (NDF), 31.5% acid detergent fibre (ADF) and 12.6% acid detergent lignin (ADL). Digestibility coefficient of dry matter (DM), CP, NDF and ADF was 0.586,0.531, 0.431 and 0.239, respectively. DM, digestible crude protein (DCP) and metabolizable energy (ME) intakes were $82.7,\;6.71g\;kg^{-1}\;W^{0.75}$ and $0.99MJ\;kg^{-1}\;W^{0.75}$. Diet of buck was constituted by $230g\;kg^{-1}$ DM of grasses and forb and $770g\;kg^{-1}$ DM of tree foliage. CP, NDF, ADF and ADL contents of the diet were 13.6, 57.8, 43.5 and 20.0%, respectively. DM, DCP and ME intakes were $57.9,\;4.13g\;kg^{-1}\;W^{0.75}$ and $0.82MJ\;kg^{-1}\;W^{0.75}$. Digestibility coefficient of CP, NDF and ADF were 0.496, 0.432 and 0.346, respectively. Diet of kids was constituted by Prosopis cineraria ($900g\;kg^{-1}DM$) and Zizyphus nummularia ($100g\;kg^{-1}DM$) leaves. Digestibility of CP, NDF and ADF were 0.456, 0.415 and 0.201, respectively. DM, DCP and ME intakes were $85.8,\;6.44g\;kg^{-1}\;W^{0.75}$ and $1.22MJ\;kg^{-1}\;W^{0.75}$. Lactation phase of goat fell between October-December and tree foliage formed the major portion of the diet. Diet of lactating goat contained only 6.8% CP and 16.5% ADL. Digestibility of DM, CP, NDF and ADF was 0.567, 0.221, 0.307 and 0.233, respectively. DM, DCP and ME intakes were $108.2,\;1.62g\;kg^{-1}\;W^{0.75}$ and $1.50MJ\;kg^{-1}\;W^{0.75}$, respectively. Present study establishes that goats on semiarid range with sufficient number of fodder trees were able to meet the nutrient requirement of ewes during oestrous and bucks during breeding season. However, during lactation and active growth phase, only DM requirement is met. Thus, it is recommended to supplement concentrate mixture to goats grazing on range during lactation and growth phase.

Sheanut cake (SNC), expeller (SNE) and solvent extractions (SNSE) samples were evaluated to determine their suitability in animal feeding. The CP content was highest in SNSE (16.2%) followed by SNE (14.7%) and SNC (11.6%). However, metabolizable energy (ME, MJ/kg) was maximum in SNC (8.2) followed by SNE (7.9) and SNSE (7.0). The tannin phenol content was about 7.0 per cent and mostly in the form of hydrolyzable tannin (HT), whereas condensed tannin (CT) was less than one per cent. The in vitro gas production profiles indicated similar y max (maximum potential of gas production) among the 3 by-products. However, the rate of degradation (k) was maximum in SNC followed by SNE and SNSE. The $t^{1/2}$ (time taken for reaching half asymptote) was lowest in SNC (14.4 h) followed by SNE (18.7 h) and SNSE (21.9 h). The increment in the in vitro gas volume (ml/200 mg DM) with PEG (polyethylene glycol)-6000 (as a tannin binder) addition was 12.0 in SNC, 9.6 in SNE and 11.0 in SNSE, respectively. The highest ratio of $CH_4$ (ml) reduction per ml of the total gas, an indicator of the potential of tannin, was recorded in SNE (0.482) followed by SNC (0.301) and SNSE (0.261). There was significant (p<0.05) reduction in entodinia population and total protozoa population. Differential protozoa counts revealed that Entodinia populations increased to a greater extent than Holotricha when PEG was added. This is the first report on the antimethanogenic property of sheanut byproducts. It could be concluded that all the three forms of SN byproducts are medium source of protein and energy for ruminants. There is a great potential for SN by-products to be incorporated in ruminant feeding not only as a source of energy and protein, but also to protect the protein from rumen degradation and suppress enteric methanogenesis.

Breeding programme was initiated during November, 1995 with the main objective to breed productive multivoltine breeds/ hybrids with suitable genetic constitution to suit the fluctuating tropical climate prevailed in India. Two multivoltine breeds viz., BL-24 and BL-27 selected were as breeding resource materials from the silkworm germplasm maintained at Central Sericul-tural Research and Training Institute, Mysore. By adopting hybridization, backcrossing inbreeding and selection, a new multivoltine breed namely BL-67. This breed spins light greenish yellow cocoons and cocoon shape is oval with medium to coarse grains. The evolved breed was crossed with five tropical bivoltine breeds viz., NB4D2, CSR2, CSR5, CSR18 and CSR101 to study the combining ability, and identified a superior hybrid, BL67 ${\times}$ CSR101, named as CAUVERY, The hybrid is characterized by high pupation rate (>95%), high shell weight (> 35 cg), high cocoon shell ratio (> 20%), longer filament length (> 900 m) and high neatness (93) with a renditta of 6.5 producing 2A-3A grade silk. The hybrid is selected for Race Autho-rization test of Central Silk Board.

A breeding programme was initiated during 2001 to introduce sex-limited larval markings to a productive multivoltine breed - BL67 from an inbred sex-limited line, MY1 (SL) maintained at Central Sericultural Research and Training Institute, Mysore. Introgressive hybridization, recurrent backcrossing for six generations followed by sib-mating resulted in synthesis of a new multivoltine silkworm breed BL67 (SL) with sex-limited larval markings. The new breed was studied for combining ability by crossing with eight bivoltine breeds viz., $NB_4D_2,\;CSR_2,\;CSR_2 (SL),\;CSR_2,\;CSR_4,\;CSR_8,\;CSR_{18}\;and\;CSR_{19}$. General combining ability effects of the new breed showed its superiority over the popular Pure Mysore by expressing significant GCA effects for six out of twelve characters whereas the results are on par with the original multivoltine breed. The hybrid $BL67(SL){\times}CSR2(SL)$ excelled in several quantitative characters such as pupation rate (90.2%), cocoon weight (1.97 g), cocoon shell weight (40 cg), cocoon shell ratio (20.3%), filament length (918 m), denier (2.96), raw silk percentage (14.96%) and neatness (90 p). Studies on cocoon size variability revealed that the cocoons of $BL67(SL){\times}CSR2(SL)$ were found comparatively uniform showing less standard deviation of 6.55 and co-efficient of variation of 3.91 %. The suitability of newly developed breed for easy grain age operation and commercial exploitation with promising hybrid have been discussed.

Variation in the size of the silk filament will determine the uniformity and quality of the silk reeled. In the present study, an attempt has been made to study the filament size variation in 6 multivoltine parental breeds and 9 multivoltine ${\times}$ bivoltine hybrids in all three seasons of a year. All multivoltine breeds and multivoltine ${\times}$ bivoltine hybrids showed variation in filament size throughout its length from the outer layer to inner layer. Results of the present study indicated that the size of the filament decreased from outer to inner layer. The decrease in filament size was sudden in some breeds/hybrids whereas it was gradual in other. Relationship between filament length to that of slope, average filament size to slope, and maximum filament size to slope was determined based on regression analysis. Regression analysis revealed significant positive correlation between slope vs average filament length (r=0.92$^{**}$) in multivoltine${\times}$bivoltine hybrids. Among parental breeds, 96C showed lowest slope of the curve (b-value: -0.00428) and 96A showed highest slope of the curve (b-value: -0.00269). Among 9 hybrids, PM${\times}$N $B_4$ $D_2$ recorded lowest slope value (b-value: -0.00328) and BL24${\times}$N $B_4$ $D_2$ showed highest value for slope of the curve (b-value: -0.00234). The breed 96C, which showed lowest slope value can be utilized for future breeding programmes to breed strains with less size deviation. Three multivoltine${\times}$bivoltine hybrids viz., PM${\times}$N $B_4$ $D_2$, 96E${\times}$CSR19 and BL67${\times}$CS $R_{101}$ , which showed less slope values (b-values: -0.00328, -0.00300 and -0.00297 respectively) can be utilized for commercial exploitation to produce uniform silk.k.

Data from an experiment involving six polyvoltine and three bivoltine silkworm breeds have been analysed to know the level of hybrid rigour and cocoon shape variation in Fl hybrids among silkworm breeds with lows medium and high cocoon weight. Results indicated significantly positive heterosis for several quantitative characters with maximum total heterosis over mid parent value in hybrids between low and medium followed by low and low, medium and medium, low and high and high and low cocoon-weighed breeds, respectively. Study on cocoon shape measurement revelled that among Fl hybrids, cocoons of the three combinations such as GNP ${\times}$ CSR2, GNP ${\times}$ J2 and PM ${\times}$ NB4 D2 were comparatively uniform in their cocoon shape. Importance of this study to know the level of heterosis and cocoon shape variability in different silkworm crosses hale been discussed.
