        In vitro DNA damage, protein oxidation protective activity and antioxidant potentials of almond fruit (Amygdalus trichamygdalus) parts (hull and drupe) using soxhlet ethanol extraction

        Almond is a nut that is consumed abundantly all over the world. In Turkey, almonds are grown in most parts of the Turkey and the consumption of nuts has become an essential part of healthy life. Almond contain powerful nutrients, which may be related to their fatty acid composition. Therefore, the aim of present study was to investigate the antioxidant potentials and the protective activity of ethanol extraction of Amygdalus trichamygdalus fruit parts (Hull and Drupe) against the lipid peroxidation, oxidative DNA damage and protein oxidation using soxhlet ethanol extraction. To the best of our knowledge, our study can be evaluated as the first report. The total phenolic content was found to be equivalent to 28.16 ± 9.990 μg/g gallic acid extract and the total flavonoid content was equivalent to 8.866 ± 0.208 μg/g quercetin extract. The present study shows that A. trichamygdalus extract have good protective activity against lipid peroxidation in the FeCl2– H2O2 system. The extract also showed a strong DPPH radical scavenging ability in a concentration-dependent manner. At 2000 μg/ml, the DPPH radical scavenging activity was found to be 84.47 ± 0.147%. The protein oxidation was also inhibited by A. trichamygdalus extract. At 2000 μg/ml, A. trichamygdalus extract protected protein oxidation by 76.27 ± 1.14%. In addition, the DNA oxidation damage against OH radicals resulting from H2O2 photolysis was inhibited concentration dependently and DNA damage was suppressed by 50.26 ± 2.6% at 1000 μg/ml concentration. The results suggest that A. trichamygdalus fruit extract have antioxidant activity and protect lipid, protein and DNA against oxidative damage. Therefore, the extract may be useful in the food industry as natural antioxidants.

        The Evaluation of Oxidative Stress in the Serum and Tissue Specimens of Patients With Chronic Otitis Media

        Objectives. To underline the effect of oxidative stress in chronic otitis media with and without cholesteatoma and to compare the oxidative stress values in the serum and tissue specimens in these two forms. Methods. The study included a total of 75 individuals, 35 cases with chronic otitis media (COM; 16 females and 19 males) and a healthy control group of 40 cases (20 females and 20 males). The COM patient group was comprised of 18 patients with cholesteatoma and 17 patients without cholesteatoma. All patients underwent mastoidectomy. Serum specimens were taken prior to surgery and diseased tissue specimens from the ear were obtained during surgery from all patients. Only serum specimens were taken from the healthy control cases. The malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GHPx) were measured in the serum and tissue samples of the patient group and in the serum specimens of the control group. Results. The age ranged from 14 to 48 years in the patient group (mean age, 20.4±12.2 years) and from 19 to 40 years in the control group (mean age, 26.4±4.64 years). When the serum values of all COM patients were compared with those of the control group, in the patient group MDA, which reflects lipid peroxidation, was found to be significantly higher (P<0.01) whereas the antioxidant enzymes SOD, CAT, and GHPx were found to be significantly lower (P<0.01). When the serum and tissue MDA, SOD, CAT, and GHPx values in patients with and without cholesteatoma were compared, no significant difference was found these parameters (P>0.01). Conclusion. Although oxidative stress plays a role in the pathogenesis of COM with or without cholesteatoma, it may not reflect the severity of the disease. In patients with COM, the evaluation of only serum oxidative stress values without tissue evaluation may be sufficient for assessing oxidative stress.

      • Serum Levels of Trace Elements in Patients with Prostate Cancer

        Background: Trace elements are major components of biological structures; however, excessive levels of these elements can be toxic. Materials and Methods: In the present study, serum levels of trace elements were measured in 30 patients with newly diagnosed as prostate cancer and 32 healthy volunteer by using furnace atomic absorption spectrophotometry. Results: It was found that there was an increase in serum levels of Co, Cu, Mg and Pb (p<0.05), whereas a decrease in serum levels of Fe, Mn, and Zn levels in patients with prostate cancer (p<0.05). Conclusions: These changes may be important in the pathogenesis of prostate cancers; however, further prospective studies are needed to identify relationships between prostate cancer and trace elements.

      • Levels of Serum Trace Elements in Renal Cell Carcinoma Cases

        Trace elements which are essential components of biological structures may also be toxic when present at levels above the amounts required for biological function. In our study, trace element levels were measured with furnace atomic absorption spectrophotometry in 33 newly diagnosed renal cell carcinoma cases (preoperative) and 32 healthy controls. When compared with the control group, it was found that the levels of cadmium (Cd), lead (Pb) were higher and the levels of zinc (Zn), iron (Fe) and manganese (Mn) were lower in the patient group. These changes may be important in the formation of renal cell carcinoma, a question which should be explored with postoperative comparative studies.
