용암해수를 이용한 해조류-식물 복합추출물의 숙취해소 효과

본 연구에서는 복합추출물을 이용하여 ADH 및 ALDH효소 활성에 미치는 영향과 항산화 능력을 in vitro에서 분석하였고, 이를 알코올과 함께 마우스에 투여하였을 때 나타나는 활동성의 변화를 관찰하고 혈중 aldehyde 농도를 정량하여 알코올 섭취 후 나타나는 복합 추출물의 숙취해소 효능을 규명하였다. 복합추출물의 free radical 소거활성능력을 측정한 결과, 농도 의존적으로 DPPH 라디칼 소거능이 증가하였으며, 특히, 120 μg/mL의 농도에서 복합추출물은 59.48%의 free radical 소거활성을 나타내어 항산화 효능이 있음을 확인하였다. ADH 및 ALDH 효소 활성을 측정한 결과, 복합추출물 농도 의존적으로 ADH 및 ALDH 효소 활성을 증가시켰으나, 유의성 있는 변화는 관찰되지 않았다. Locomotor activity test를 실시한 결과, 마우스의 활동성은 복합추출물-물을 투여한 군의 경우 투여후 120분까지의 활동성이 알코올-물 투여군에 비하여 현저히 증가됨을 관찰할 수 있었다. 복합추출물을 경구 투여하여 120분이 경과한 후 혈중 alcohol과 acetaldehyde 정량결과, 알코올 투여 후 복합추출물군을 투여군에서 양성대조군인(알코올-물)군에 비하여 유의성 있게 감소된(p<0.0001)혈중 alcohol 농도(33.48±0.59 μg/mL)와 aldehyde 농도(1.54±0.13 μg/mL)를 나타내었다. 이상의 결과에서 복합추출물이 ADH와 ALDH의 활성을 유의적으로 증가시키지는 않았으나, 복합추출물에 의하여 증가된 마우스의 활동성은 혈중alcohol과 acetaldehyde의 감소된 농도에 기인함을 알 수 있었다. This study was conducted to evaluate the hangover relieving effect of algae and plant complex extract. The complex extract showed 1,1-diphenyl-2-picrylhydrazyl radical scavenging effect in a concentration-dependent manner and high antioxidant capacity (59.48%) when administered at 120 μg/mL. In addition, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activities were increased according to the concentration of the complex extract. A locomotor activity test with an alcohol-complex extract mouse group showed improved motor activities over that of the alcohol-water group at 120 min post-administration. Blood was harvested from each mouse at 120 min post-administration then concentrations of alcohol and aldehyde were measured. The alcohol-extract treated group showed significantly lower (p<0.0001) alcohol (33.48 ± 0.59 μg/mL) and aldehyde (1.54 ± 0.13 μg/mL) concentrations than the alcohol-water treated group. These results suggest that the hangover relieving effects of complex extract results in increased motor activity of mice, which reduces the alcohol and aldehyde concentration in the blood.

Objectives: Adipogenesis was defined as a differentiation process of preadipocytes into the adipocytes. Thus, to control of this process can be one of the most important strategies to prevent obesity. Korean ginseng(Panax ginseng C.A. Meyer) is one of the most widely used medicinal herbs. Although multiple biological activities of Korean ginseng, particularly ginsenosides, have been known, the anti-adipogenic role and function of polysaccharides from Korean ginseng are still unclear. In this study, we examined anti-adipogenic activity of polysaccharides and its molecular basis mechanisms are further investigated. Methods : The cytotoxicity of KGP in 3T3-L1 was evaluated by MTT assay. Anti-adipogenic effect of KGP was examined by Oil Red O (ORO) staining and microscopy observation in 3T3-L1 mature adipocytes. The mRNA expression levels of adipogenic transcriptional factors were analyzed by reverse transcription-polymer chain reaction (RT-PCR). To elucidate the adipogenic molecular mechanism of KGT, SB431542 (TGF-β specific inhibitor) was used. Results : We found that polysaccharides showed no effect on the viability of 3T3-L1 preadipocytes. Dose dependent inhibitory effect of polysaccharides on 3T3-L1 adipogenesis was observed as judged by ORO staining and microscopic image analysis. To obtain further mechanistic insight into anti-adipogenic function of polysaccharides, we then tested the effect of polysaccharides treatment on the adipogenic marker genes. The mRNA expressions level of C/EBPα, PPARγ, C/EBPβ, and fatty acid synthase (FAS) were dose-dependently inhibited by KGP treatment in 3T3-L1 mature adipocytes. Conclusions: In conclusion, these findings suggest that the KGP could be used in treatment of obesity and over-weight related diseases.
