We have fabricated galactosylated chitosan-incorporated polyurethane foams (GC-PU) by dipping polyurethane foam (PUF) in an aqueous solution containing galactosylated chitosan and by subsequent reaction with glutaraldehyde after freeze-drying. It was found from the hepatocyte adhesion experiment that the amount of hepatocytes seeded on GC-PU was higher than those on PUF and chitosan incorporated PUF (C-PU). The cells inoculated in GC-PU showed higher ammonia removal ability and albumin secretion than those on collagen monolayered dish. Albumin secretion by hepatocytes seeded on GC-PU increased by the presence of serum. The results obtained from 3-(3,4-dimethylthiazol-2yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay indicated that GC-PU can provide a better microenvironment for hepatocyte culture along with nutrition and metabolite transfer through the pores of the matrices.
