      • 異種 Plasmid에 의한 Saccharomyces cerevisiae의 同時形質 轉換

        The yeast S. cerevisiae DBY747 was transformed with E. C - S. C shuttle vector YIp5, YEp13 and YRp7 by the method of spheroplast. The transformation frequency of YEp13 and YRp7 in S. cerevisiae DBY747 was 1.2×10^(3) and 1.0×1.0^(2) per 10㎍ of DNA, respectively. The transformants with YIp5 plasmid incapable of autonomous replication in S. cerevisiae were not detected in the condition of this experiment, but YIp5 plasmid expressed the gene carried on it when cotransformed with a helper plasmid such as YEp13 or YRp7 : autonomously replicating plasmid. When plasmids were used in covalently closed circular form, contransformation frequency of YIp5 - YEp13 and YIp5 - YRp7 was 210 and 95 per 10㎍ of DNA, respectively. In contransformation of linear plasmids, transformation frequency of the same cohesive ends was similar to that of noncomplementary cohesive ends. Transformants by the cotransformation with circular plasmids have been shown much higher frequency than with linear plasmids in S. cerevisiae DBY 747. The mitotic segregation stability test suggested that the cotransformant of YIp5-YEp13 was more stable than that of YIp5-YRp7.

      • Aninetobacter calcoaceticus C10에서 Cyclohexanol Dehydrogenase의 생합성 억제

        The growth of A. calcoaceticus C10 in CL medium was not increased by the addition of 0.5% E-caprolactone or succinate, but increased by 0.2% adipate, xylose or even glucose which was not metabolized as a carbon source. The addition of 0.2% glucose after culture in CL medium for 6 hours increased the growth of A. calcoaceticus C10 twice as much as that in CL medium after culture for further 10 hours. Biosynthesis of cyclohexanol dehydrogenase in A. calcoaceticus C10 was not repressed by ε - caprolactone, succinate, xylose and glucose, but repressed by adipate which is endproduct of cyclohexanol metabolism. The induction of dehydrogenase by cyclohexanol in CL medium was not repressed completely by 0.1% adipate, but repressed almost completely by 0.2% adipate in A. calcoaceticus C10.

      • 優良室素固定菌의 探索分離 및 生理的 特性

        This experiment was conducted to search for Rhizobia with good nitrogen fixation abilities and to investigate their physiological characteristics isolated from 12 soybean cultivars and the affinities of root nodule bacteria with soybeans. The results obtained were as follows ; Based on colors, Rhizobia grown on YMA medium were divided into 3 groups, i. e., white, translucent and transparent, amounting to 60, 30 and 10%, respectively. In litmus milk reation, the strains which produced alkali, acid serum, alkaline serum and acid reached to 51, 29, 9 and 11%, respectively. Strains, S022, and S096 were slow-growers and produced alkaili, while strains, S080, S090, and S118 were fastgrowers and produced acid. The growth of root nodule bacteria on YMA medium was favorable between the initial pHs of 6.0~7.0. Glutamine, asparagine and allantoin as nitrogen sources enhanced the growth of root nodule bacteria. All the strains tested formed nodules on the soybean roots, and the strains with good symbiotic nitrogen fixation abilities that had white color, small colony, nitrate reduction abilities and no nitrite reduction abilities showed comparatively high nitrogen fixing activities. Some strains varied in nitrogen fixing activities according to soybean cultivars, and a few strains formed ineffective nodules which showed no nitrogen fixing activity.
