        보리 無葉耳 形質의 遺傳과 關與 遺傳子 作用 效果

        The study was conducted to investigate the inheritance patterns of liguleless and its associated genetic effects on other agronomic characteristics in barley. From 1993 to 1994 the parents, F₁, and F₂s obtained from reciprocal cross of OUM 267, OUM 386 with liguleless and Olbori, Chalbori, Suwon #259 with normal leaf characteristics were planted in the field of Barley and Wheat Research Institute (Suwon). The OUM 267 and OUM 386 varieties showed a liguleless and an auricleless phenotype and contained higher chlorophyll content and light transmission ratio than varieties with normal leaf characteristics, while leaf area index (LAI) was lower than the latter population. In the plotting culture (5×5㎝), the liguleless barley of the F₂ generations showed more an improved light-absorving capacity and net assimilation ratio than variety having characterisitics of normal leaf. The liguleless characteristics were inherited by a simple recessive gene and appeared to be controlled by one set of allele. The genes of liguleless and two-rowed vs sixrowed spike in barley located in the Ⅱ linkage group, and the recombination values between gene of liguleless and two, six-rowed were 37.0~47.7%. The gene governing liguleless characteristics showed some pleiotropic effects on heading and growing characteristics to the direction of short stature, delaying of heading. The OUM 267 combination of two-rowed spike with liguleless gene, also, exhibited pleiopropic effects to the direction of decreasing 1,000-grain weight. The OUM 386 combination of six-rowed spike with liguleless gene was no significant genotypic and phenotypic correlation between leaf blade angle and 1,000 grain weight. But cross lines expected that these can be used to breed a new variety of high-yielding barley due to potential productivity as well as improved light-absorbing capability.

        Isolation and Characterization of a Ds-tagged liguleless Mutant in Rice (Oryza sativa. L)

        A liguleless mutant, which showed complete loss of lamina joint region at the junction between leaf blade and leaf sheath, was isolated from a Ds insertional mutants derived from the source cultivar, Dongjin. This mutant could not affect other developmental patterns like phyllotaxis. Southern blot analysis, using GUS as a probe, revealed that the liguleless mutant contained three Ds copies transposed in the rice genome. Among the four genomic sequences flanking the Ds, one was mapped in the intergenic region (31661640 - 31661759), and the other two predicted a protein kinase domain (12098980 - 12098667) as an original insertion site within a starter line used for massive production of Ds insertional mutant lines. Another predicted and inserted in first exon of liguleless 1 protein (OsLG1) that was mapped in coding region (LOC_Os04g56170) of chromosome 4. RT-PCR revealed that the OsLG1 gene was not expressed liguleless mutants. Structure analysis of OsLG1 protein revealed that it predicted transcription factor with a highly conserved SBP domain consisting of 79 amino acids that overlapped a nuclear localization signal (NLS). RT-PCR revealed that OsLG1 is mainly expressed in vegetative organs.
