        Influence of Temperature and Relative Humidity on the Rearing Performance and Disease Incidence in CSR Hybrid Silkworms, Bombyx mori L.

        Fifth instar larvae of the new bivoltine hybrid (CSR2 ${\times}$ CSR5) were reared under different temperature and humidity viz., $20^{\circ}C$ and 85 ${pm}$ 5% R.H (T1), $25^{\circ}C$ and 70 ${pm}$ 5% R.H (T2-Optimum), $30^{\circ}C$ and 80% ${pm}$ 5 R.H (T3) and $35^{\circ}C$and 50 ${pm}$ 5% R.H (T4). The cocoon yield, cocoon characters and disease incidence were studied in normal (non infectious source, i.e control) rearing as well as in 1% infectious source of rearing. The results indicated that V instar larval duration was prolonged and cocoon weight was improved in T1. ERR and shell ratio were significantly improved and disease incidence was minimised in T2. Further significant difference was observed among the treatments with regard to spread of diseases.

      • Effect of Phytohormones on Multiple Shoot Bud Induction in cv. NARI-6 of Safflower (Carthamus tinctorius L.)

        In the present study, in vitro multiple shoot induction was achieved from cotyledonary node and stem nodal explants of cv. NARI-6 of safflower (Carthamus tinctorius L.). Among various growth regulators tested, MS salts and B5 vitamins supplemented with BA (6-Benzy-laminopurine) $17.76\;{\mu}M$ and KN (Kinetin) $6.96\;{\mu}M$ phytohormonal combination was found to be the most effective in initiating numerous shoot buds after 30 days of culture than BA ($4.44-44.39\;{\mu}M$) or KN ($2.32-46.40\;{\mu}M$) alone in the medium. In addition, 0.8% (w/v) agar (Hi-media) and 3.0% sucrose (w/v) was the optimum level for the formation of adventitious shoots. Further results showed the maximum shoot elongation occurred on MS medium with BA ($8.88\;{\mu}M$) and $GA_3$($11.56\;{\mu}M$) combinations. Efficient rooting occurred on quarter strength MS medium with NAA $10.74\;{\mu}M$. The regenerated plantlets were acclimatized and successfully transferred to the field.

        Acceleration of Mounting in Self Mounting Method and Its Effect on Cocooning, Cocoon Characters and Reeling Parameters in Silkworm Bombyx mori L.

        Rotary mountages are the best mountages among all the mountages that are used in sericultural areas. But to use this mountage more space, separate mounting hall & requires more labour to pickup the matured worms. to over come these problems, self mounting method is adopted to save the time Er labour, But the mounting rate is generally less. To accelerate the mounting rate different repellents viz; saw dust, phytoecdysone, 1% cresol with paddy husk, lime, kaolin, gormalin chaff were used in the present study. The results indicated that highest number of larvae climbed the mountage in 1% cresol with paddy husk, followed by phytoecdysone and lime. It is observed that the use of repellents at wandering stage accilerates mounting rate and did not affect the quality of the cocoons and reeling characters.

        Heterotic Expression in Silk Productivity of Newly Evolved Thermo-tolerant Bivoltine Hybrids of Silkworm Bombyx mori L.

        Studies on heterotic expression in the silk productivity of 66 thermo-tolerant bivoltine hybrids reared under high temperature $(36{\times}1^{\circ}C)$ and low humidity (60${\times}$5%) conditions revealed that manifestation of heterosis was highly significant for majority of the quantitative traits contributing to more cocoon productivity. Observations were made on 8 economically important traits such as fecundity, pupation rate, cocoon yield, cocoon weight, cocoon shell weight, cocoon shell ratio, cocoon filament length and raw silk. Analysis of variance results revealed great deal of variability indicating significant (P < 0.001) variations. Out of 66 hybrids, 7 hybrids exhibited positive heterosis for seven traits and 4 hybrids expressed significant hetero-beltiosis for 6 traits, out of 8 traits evaluated. Based on results, the hybrids ${SR_1} {\times} {SR_5}and {CSR_2} {\times} {SR_5}$ are adjudged as most promising ones and can be exploited commercially in tropical climates to increase the cocoon productivity.

        Line${\times}$Tester Analysis of Certain Quantitative Traits in Silkworm Bombyx mori L. under Optimum and Stress Rearing Conditions

        The combining abilities in the 5 newly evolved thermo tolerant breeds viz., SR6, SR7, SR8 SR9 and SR10 of silkworm Bombyx mori L. and their 15 hybrids were made in a line${\times}$tester crossing programme. Data were analysed for seven quantitative traits i.e., pupation rate, cocoon yield, cocoon weight, cocoon shell weight, cocoon shell ratio, filament length and raw silk percentage under optimum room temperature $(25{\pm}{1^{\circ}C})$ conditions (In case of high temperature $(36{\pm}{1^{\circ}C})$ stress conditions five economic traits except filament length and raw silk percentage) with 3 widely adapted testers i.e., KA, CSR2 and CC1 as lines (females) and testers (males) respectively. The performance at high temperature and low humidity conditions aye only taken into consideration for selecting the best lines/hybrids. Among the lines SR6 exhibited positive General combining ability (GCA) effects for pupation rate, cocoon yield, cocoon weight and cocoon shell ratio traits, followed by SR7 for pupation rate, cocoon yield and cocoon shell weight and cocoon shell ratio. Among testers, KA exhibited positive GCA effects for two quantitative traits cocoon yield, cocoon weight and CSR2 for cocoon shell weight and cocoon shell ratio under adverse temperature conditions. The hybrid SR6${\times}$CC1 and SR7${\times}$CSR2 exhibited significant positive Specific combining ability (SCA) effects for majority of the traits in high temperature stress conditions of rearing. The better parent value of heterosis(Heterobeltiosis) was exhibited by the hybrid SR6${\times}$CC1 for pupation rate, cocoon yield, cocoon weight and cocoon shell weight and SR7${\times}$CSR2 for all the trails evaluated under high temperature conditions. Based on the results, the lines SR6 and SR7 was judged as best combiners and the hybrids SR6${\times}$CC1 and SR7${\times}$CSR2 can be selected for commercial exploitation in tropical climate.

        Lipid Raft Integrity Is Required for Survival of Triple Negative Breast Cancer Cells

        Purpose: Lipid rafts are cholesterol enriched microdomains that colocalize signaling pathways involved in cell proliferation, metastasis, and angiogenesis. We examined the effect of methyl-β- cyclodextrin (MβCD)-mediated cholesterol extraction on the proliferation, adhesion, invasion, and angiogenesis of triple negative breast cancer (TNBC) cells. Methods: We measured cholesterol and estimated cell toxicity. Detergent resistant membrane (DRM) and non-DRM fractions were separated using the OptiPrep gradient method. Cell cycles stages were analyzed by flow cytometry, apoptosis was assessed using the TdT-mediated dUTP nick end-labeling assay, and metastasis was determined using a Matrigel invasion assay. Neo-vessel pattern and levels of angiogenic modulators were determined using an in vitro angiogenesis assay and an angiogenesis array, respectively. Results: The present study found that the cholesterol-depleting agent MβCD, efficiently depleted membrane cholesterol and caused concentration dependent (0.1–0.5 mM) cytotoxicity compared to nystatin and filipin III in TNBC cell lines, MDA-MB 231 and MDA-MB 468. A reduced proportion of caveolin-1 found in DRM fractions indicated a cholesterol extraction-induced disruption of lipid raft integrity. MβCD inhibited 52% of MDA-MB 231 cell adhesion on fibronectin and 56% of MDA-MB 468 cell adhesion on vitronectin, while invasiveness of these cells was decreased by 48% and 52% respectively, following MβCD treatment (48 hours). MβCD also caused cell cycle arrest at the G2M phase and apoptosis in MDA-MB 231 cells (25% and 58% cells, respectively) and in MDA-MB 468 cells (30% and 38% cells, respectively). We found that MβCD treated cells caused a 52% and 58% depletion of neovessel formation in both MDA-MB 231 and MDA-MB 468 cell lines, respectively. This study also demonstrated that MβCD treatment caused a respective 2.6- and 2.5-fold depletion of tyrosine protein kinase receptor (TEK) receptor tyrosine kinase levels in both TNBC cell lines. Conclusion: MβCD-induced cholesterol removal enhances alterations in lipid raft integrity, which reduces TNBC cell survival.
