        Time-series transcriptomic analysis of the kelp grouper Epinephelus moara in response to low salinity stress

        The Kelp grouper Epinephelus moara is one of the most widely consumed and economically important marine fish in China. The species can tolerate a wide range of salinity, but genomic resources are not available, and the molecular mechanisms underlying adaptation to salinity at the transcriptomic level remain largely unclear. In this study, the transcriptomic responses of the liver of E. moara under low salinity were investigated using the Illumina digital gene expression system. After de novo assembly, 499,356 transcripts were generated and contributed 445,068 unigenes. A total of 14, 19, 33 and 3101 genes were differentially expressed following exposure to low salinity stress for 2, 6, 24 and 48 h, respectively. Only two genes were differentially expressed in all groups. Four genes related to metabolism and ambient salinity adaption were randomly selected to validate the differentially expressed genes (DEGs) by real-time PCR. Gene Ontology (GO) and Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway enrichment analysis were used to analyse the functional significance of DEGs, including those responding to salinity through diverse biological processes, cellular components, molecular functions, and pathways associated with metabolic and osmotic responses. This work provides new insight into the response to salinity challenges in E. moara, and the findings expand our knowledge of the molecular basis of metabolic regulation mechanisms in this species. Additionally, the transcriptional data provide a valuable resource for future molecular and genetic studies on E. moara.

        Immunomodulatory effects of Clostridium butyricum on human enterocyte-like HT-29 cells

        Oral administration of Clostridium butyricum as probiotics is increasingly gaining importance in the treatment of intestinal inflammations and improvement of animal performance. The mechanisms of host cell receptor recognition of C. butyricum and downstream immune signaling pathways leading to this beneficial event, however, remain unclear. In this study, C. butyricum was investigated for its capability to influence the innate immune response of HT-29 cells to explore its mechanism of action. Our data showed that C. butyricum was able to stimulate toll-like receptor 2 (TLR2) production at mRNA level; however, TLR4, TLR5, TLR9, and myeloid differentiation primary response protein 88 (MyD88) transcription levels remain unaltered. The nuclear factor kB (NF-kB), interleukin-8 (IL-8), and tumor necrosis factor alpha (TNF-a) levels in response to C. butyricum were significantly increased, indicating that HT-29 cells were sensitised by C. butyricum. It is the first time that our findings have showed the specific signaling pathways in HT-29 cells stimulated by C. butyricum, which, at least in part, can help to explain the beneficial properties of C. butyricum.

        By using the generalized Riccati transformation and the in-equality technique, we establish some new oscillation criterion for the second-order nonlinear delay dynamic equations [수식]on a time scale T, here r ≥1 is the ratio of two positive odd integers with a and q real-valued positive right-dense continuous functions defined on T. Our results not only extend and improve some known results, but also unify the oscillation of the second-order nonlinear delay differential equation and the second-order nonlinear delay difference equation.

        This paper is concerned with oscillation property of solutions of a class of second order nonlinear differential equations with perturbation. Four new theorems of oscillation property are established. These results develop and generalize the known results. Among these theorems, two the-orems in the front develop the results by Yan J(Proc Amer Math Soc, 1986,98: 276-282), and the last two theorems in this paper are completely new for the second order linear differential equations.

        Induction of apoptosis in HT-29 cells by quercetin through mitochondria-mediated apoptotic pathway

        With increasing use of natural plant-derived components, exploring the anti-proliferative effects of phytochemicals is increasingly gaining importance in designing anticancer drugs. Quercetin, a natural constituent abundantly present in food products, is capable of inducing apoptosis in tumor cells. However, little is known about its biological effect on colon cancer cells and molecular mechanism leading to this event. The aim of this study was to investigate the effect of quercetin on the apoptotic pathway in human colon carcinoma cells (HT-29 cells). The results indicated that quercetin induced suppression of cell viability and apoptosis in HT-29 cells in a dose-dependent manner. This involved characteristic changes in nuclear morphology, activation of caspases-3 and caspases-9, collapse of Mitochondrial membrane potential (DCm), upregulation of pro-apoptotic Bax, and downregulation of antiapoptotic Bcl-2 and Bcl-XL. Quercetin, which exerts anti-proliferative effect though different signaling pathways, is a classic candidate for anti-colon cancer drug design.

        Localization of α-, γ-, and δ-tubulin in the hypotrich ciliate Stylonychia pustulata (Hyportrichida, Ciliophora)

        Ciliature microtubular organelles, which organize into ciliary units, are highly complex microtubular systems. Toinvestigate the formation of ciliature microtubular organelles and the role that different tubulins play in this process,we examined the distribution and function of α-, γ-, and δ-tubulins in the hypotrich ciliate Stylonychia pustulata. Immunofluorescence analysis demonstrated that α-tubulin localized to all microtubular organelles, while γ- and δ-tubulinscolocalized at basal bodies. Immunogold labeling revealed that γ- and δ-tubulins colocalize to the proximalpart of basal bodies, suggesting that γ- and δ-tubulins probably interact and may function together. In addition, thepermanent localization of γ-tubulin to the adoral zone of membranelles suggests that γ-tubulin play a role in themaintenance of oral ciliature base-associated microtubules, but does not play a role in the biogenesis of theseciliature. We observe that δ-tubulin is restricted to the basal body and the connective fibers of the basal body,suggesting this protein may be required for the extension and stability of the microtubules in these structures. Dramatically different concentrations of these three tubulins were observed in the cytoskeleton and the insoluble andsoluble fractions of the cell, suggesting that these highly related proteins perform different functions in S. pustulata.
