        Ultra Small, mono dispersed green synthesized silver nanoparticles using aqueous extract of <i>Sida cordifolia</i> plant and investigation of antibacterial activity

        <P><B>Abstract</B></P> <P>The present study is focused on the synthesis of silver nano particles (Ag NPs) using an aqueous extract of the whole plant of <I>Sida cordifolia</I> as a potential bio-reducing agent and assessment of their antibacterial activity. UV–Vis spectroscopy of composed silver colloidal solution displayed surface Plasmon resonance peak at 420 nm. XRD and TEM analysis revealed the morphology as ultra-small, monodispersed spherical nanoparticles with face-centered cubic structure and mean particle size of 3–6 nm. This ultra-small nano size might owe to the slow reaction time and phytochemicals existing in the <I>S. cordifolia</I> extract. The Ag NPs are trailed for antibacterial activity against 5 fish (<I>Aeromonas hydrophila</I>, <I>Pseudomonas fluorescence</I>, <I>Flavobacterium branchiophilum, Edwardsiella tarda</I> and <I>Yersinia rukeri</I>) and 4 human (<I>Escherichia coli</I>, <I>Klebsiella pneumonia</I>, <I>Bacillus subtilis</I> and <I>Staphyloccocus aureus</I>) bacterial pathogens. In all the cases, Ag NPs from <I>Sida cordifolia</I> plant extract manifested noteworthy antibacterial effects on par with positive control i.e.; Gentamicin.</P> <P><B>Highlights</B></P> <P> <UL> <LI> First report on the biosynthesis of Ag NPs using Sida Cordifolia plant extract. </LI> <LI> Ultra Small, Monodispersed Ag NPs using green Synthesis. </LI> <LI> Act as a good antibacterial agent against various human and fish bacterial pathogens. </LI> <LI> Ag NPs were characterized with UV–Vis, XRD, SEM and TEM. </LI> </UL> </P>

        Ecobiotechnology of Marine Sponges and Their Symbionts - Review and Present Status

        Marine sponges are a rich source of highly diversified bioactive compounds. These medicinally valuable molecules represent extreme physiological and ecological functions in sponges, more presumably involving in the resistance to the feeding by other marine organisms like fish and fouling by barnacles, bacteria, fungi, etc. This feature of attaining resistance made sponges as successful poriferans that possess an impressive array of biological properties ranging from antimicrobial to anticancerous activities. The diversified bioactive principle of sponges might be due to their spacio-temporal distribution and although, the gateway for exploiting the sponges for isolating these distinct, potential molecules is open, suitable technical and methodological approaches are yet to be implemented in order to bring the sponges as successful pharmaceutical leads in the field of marine biotechnology. Despite of the identified difficulties of marine sponge research from past few decades, one should concentrate not only on the basic and applied technical/methodological considerations, but also on the novel strategies like in vitro sponge cell, fragment and whole sponge culture; sponge symbiont cell culture; in situ and ex situ sponge cultivation; and sponge bioreactors and metagenomic approaches, for the successful exploitation of marine sponges towards the novelty in sponge biotechnology. The present review narrates the pros and cons of the nowadays-marine sponge research by focusing on the suggestive ecobiotechnological approaches, based on the latest studies for feasible ecological exploitation and biotechnological application of sponges from the sea.

        An amperometric nanobiosensor using a biocompatible conjugate for early detection of metastatic cancer cells in biological fluid

        Metastasis is the major cause of cancer-associated death in humans, and its early diagnosis will help clinicians to develop suitable therapeutic strategies which may save life of cancer patients. In this direction, we designed an amperometric biosensor using a biocompatible conjugate to diagnose cancer metastasis by detecting epithelial cell adhesion molecule expressing metastatic cancer cells (Ep-MCCs). The sensor probe is fabricated by immobilizing monoclonal capture antibody (CapAnti) on the gold nanoparticles (AuNPs)/conducting polymer composite layer. The detection relies on a sandwich-type approach using a bioconjugate composed of reporter antibody (RepAnti), nanostructured collagen (nCOL), AuNPs, and hydrazine (Hyd) which served as a nonenzymatic electrocatalyst for the reduction of H<SUB>2</SUB>O<SUB>2</SUB>. The binding of Ep-MCCs with the sensor probe was confirmed using electrochemical impedance spectroscopy, cyclic voltammetry, and chronoamperometry. A dynamic range for the Ep-MCCs detection is determined between 45 and 100,000 Ep-MCCs/mL with the detection limit of 28+/-3 Ep-MCCs/mL. The proposed immunosensor is successfully applied to detect Ep-MCCs in serum and mixed cell samples and interferences due to nontarget cells and molecules present in the real sample matrix are also examined. The early stage of Ep-MCCs was examined by fluorescence-activated cell sorting assay, which confirms that the developed biosensor has detected Ep-MCCs in its early stage.

      • Synthesis and evaluation of the cytotoxic activities of some isatin derivatives.

        <P>A series of isatin derivatives, 1-butyl-5/7-chloro/fluoro-3-((4-methoxybenzyl)imino)indolin-2-ones (3a-d), 6-butyl-chloro/fluoro-6H-indolo[2,3-b]quinoxalines (4a-h), and 5/7-chloro/fluoro-3-((4-methoxybenzyl)imino)indolin-2-ones (5a-h) were synthesized and characterized by using Fourier transform (FT)-IR, (1)H- and (13)C-NMR spectroscopy, mass spectrometric and elemental analysis. The substances were further subjected to in vitro cytotoxicity evaluation against HeLa, SK-BR-3, and MCF-7 cells. The results showed that quinoxalines 4d, 4e, and 4g; and indolin-2-one 5f display significant in vitro cytotoxic activities against HeLa cells and further the compound 4d has resulted in highest cytotoxicity in the entire series studied. In addition, 5f was shown to display substantial activity against all the three cell lines used in the current study.</P>
