City Center Regeneration to Drive Economic Competitiveness? The Case Study of Liverpool One

This paper discusses how urban regeneration can be used to drive economic competitiveness using the case study of Liverpool One, in Liverpool, United Kingdom. With a process which began in 1999, Liverpool One resulted in over 42 hectares of land being redeveloped as part of a wholesale rejuvenation of Liverpool City Centre. This contributed to Liverpool becoming the fastest growing UK region outside of London. In discussing Liverpool One, the paper outlines some of the planning challenges faced during its construction and what this successful regeneration project now means for those seeking to develop within the city centre, whilst drawing out some of the lessons international planners and developers might learn from Liverpool.

Weed management options for adzuki-bean growers in Ontario, Canada are limited due to few herbicide registrations. Four field trials were conducted at three locations in south-western Ontario in 2007 and 2008 to determine the tolerance of adzuki bean to several preplant-incorporated (PPI), pre-emergence (PRE), and post-emergence (POST) herbicides. All the herbicides were applied at the doses registered for use in soybean. The application of pendimethalin, cloransulam-methyl, and halosulfuron-methyl (PPI), flumetsulam, cloransulam-methyl, and halosulfuron-methyl (PRE), and acifluorfen and fomesafen (POST)caused ${\leq}15%$ crop injury; however, the injury was transient and did not reduce the adzuki bean yield. The POST application of cloransulam-methyl and imazethapyr caused ${\leq}23%$ crop injury and reduced the biomass by ${\leq}50%$, but did not reduce the plant height or crop yield. Metribuzin, flumetsulam, atrazine, and pyroxasulfone (PPI), metribuzin, linuron, pyroxasulfone, and atrazine (PRE), and bentazon, imazethapyr plus bentazon, halosulfuron-methyl, and thifensulfuron-methyl (POST) caused ${\leq}61%$ crop injury. These treatments reduced the biomass, plant height, and crop yield. Based on these results, pendimethalin, cloransulam-methyl, and halosulfuron-methyl applied PPI, flumetsulam, cloransulam-methyl, and halosulfuron-methyl applied PRE, and acifluorfen and fomesafen applied POST might be potential weed management options for weed management in adzuki bean. Cloransulam-methyl and imazethapyr applied POST will need further evaluation due to phytotoxicity concerns. Metribuzin, flumetsulam, atrazine, and pyroxasulfone applied PPI, metribuzin, linuron, atrazine, and pyroxasulfone applied PRE, and bentazon, imazethapyr plus bentazon, halosulfuron-methyl, and thifensulfuron-methyl applied POST did not have an adequate margin of safety.

<P>The fission yeast Schizosaccharomyces pombe is a model organism used widely to study various aspects of eukaryotic biology. A collection of heterozygous diploid strains containing individual deletions in nearly all S. pombe genes has been created using a PCR based strategy. However, deletion of some genes has not been possible using this methodology. Here we use an efficient knockout strategy based on plasmids that contain large regions homologous to the target gene to delete an additional 29 genes. The collection of deletion mutants now covers 99% of the fission yeast open reading frames.</P>
The CCR4-NOT Complex Is Implicated in the Viability of Aneuploid Yeasts

<P>To identify the genes required to sustain aneuploid viability, we screened a deletion library of non-essential genes in the fission yeast <I>Schizosaccharomyces pombe</I>, in which most types of aneuploidy are eventually lethal to the cell. Aneuploids remain viable for a period of time and can form colonies by reducing the extent of the aneuploidy. We hypothesized that a reduction in colony formation efficiency could be used to screen for gene deletions that compromise aneuploid viability. Deletion mutants were used to measure the effects on the viability of spores derived from triploid meiosis and from a chromosome instability mutant. We found that the CCR4-NOT complex, an evolutionarily conserved general regulator of mRNA turnover, and other related factors, including poly(A)-specific nuclease for mRNA decay, are involved in aneuploid viability. Defective mutations in CCR4-NOT complex components in the distantly related yeast <I>Saccharomyces cerevisiae</I> also affected the viability of spores produced from triploid cells, suggesting that this complex has a conserved role in aneuploids. In addition, our findings suggest that the genes required for homologous recombination repair are important for aneuploid viability.</P><P><B>Author Summary</B></P> <P>Aneuploidy is a major cause of abortive development and is implicated in tumorigenesis in humans. Recent studies revealed that the increased need for protein degradation might account for the detrimental effects of aneuploidy on a cell. Here, we investigated the genetic systems responsible for aneuploid viability. Using a collection of gene deletions in fission yeast, we isolated mutants that affect aneuploid viability. We found that an evolutionarily conserved transcription regulator, the CCR4-NOT complex, and its related factors are required for aneuploid viability, suggesting that regulation of mRNA turnover is required to tolerate aneuploidy. In addition, homologous recombination repair is important for aneuploid viability.</P>
