      • Esophageal/Gastric Cancer Screening in High-risk Populations in Henan Province, China

        Objective: To summarize the endoscopic screening findings in high-risk population of esophageal and gastric carcinoma and analyze influential factors related to screening. Methods: In seven selected cities and counties with high incidences of esophageal carcinoma, people at age of 40-69 were set as the target population. Those with gastroscopy contradictions were excluded, and all who were voluntary and willing to comply with the medical requirements were subjected to endoscopic screening and histological examination for esophageal, gastric cardia and gastric carcinoma in accordance with national technical manual for early detection and treatment of cancer. Results: In three years, 36,154 people were screened, and 16,847 (46.60%) cases were found to have precancerous lesions. A total of 875 cases were found to have cancers (2.42%), and among them 739 cases had early stage with an early diagnosis rate is 84.5%. Some 715 patients underwent prompt treatment and the success rate was 81.8%. Conclusions: In a high-risk population of esophageal and gastric carcinoma, it is feasible to implement early detection and treatment by endoscopic screening. Screening can identify potential invasive carcinoma, early stage carcinoma and precancerous lesions, improving efficacy through early detection and treatment. The exploratory analysis of related influential factors will help broad implementation of early detection and treatment for esophageal and gastric carcinoma.

        The strength analysis and probabilistic design of a bogie frame with incomplete probabilistic information

        A bogie frame is a key component of high-speed rail; higher requirements of the safety and stability of the bogie frame are put forward with the continuous improvement of the train speed. In this study, we first performed a strength analysis of a bogie frame according to the JIS E 4207 standard by using finite element method. Then, we did a reliability analysis of the bogie frame. The reliability problems are defined as strength reliability and resonant frequency reliability, which indicate the structural safety and stability of the bogie frame. The reliability estimation is realized by experimental design, artificial neural network and stochastic perturbation theory. Additionally, reliability-based sensitivity indices were derived to measure the parameter importance of random input variables. An illustrative example of the bogie frame with incomplete probabilistic information was used to demonstrate the applications of the proposed method for reliability and reliability-based sensitivity estimation in terms of strength and resonant frequency reliability. The results indicate that vertical loads and the material density have more of an impact on reliability than other variables of the bogie frame.

        Function of Rhs proteins in porcine extraintestinal pathogenic Escherichia coli PCN033

        Extraintestinal pathogenic Escherichia coli (ExPEC) is an important zoonotic pathogen that places severe burdens on public health and animal husbandry. There are many pathogenic factors in E. coli. The type VI secretion system (T6SS) is a nano-microbial weapon that can assemble quickly and inject toxic effectors into recipient cells when danger is encountered. T6SSs are encoded in the genomes of approximately 25% of sequenced Gram-negative bacteria. When these bacteria come into contact with eukaryotic cells or prokaryotic microbes, the T6SS assembles and secretes associated effectors. In the porcine ExPEC strain PCN033, we identified four classic rearrangement hotspot (Rhs) genes. We determined the functions of the four Rhs proteins through mutant construction and protein expression. Animal infection experiments showed that the Δrhs-1CT, Δrhs-2CT, Δrhs-3CT, and Δrhs-4CT caused a significant decrease in the multiplication ability of PCN033 in vivo. Cell infection experiments showed that the Rhs protein is involved in anti-phagocytosis activities and bacterial adhesion and invasion abilities. The results of this study demonstrated that rhs1, rhs3, and rh4 plays an important role in the interaction between PCN033 and host cell. Rhs2 has contribution to cell and mice infection. This study helps to elucidate the pathogenic mechanism governing PCN033 and may help to establish a foundation for further research seeking to identify potential T6SS effectors.

        Mutations in DZIP1L, which encodes a ciliary-transition-zone protein, cause autosomal recessive polycystic kidney disease

        <P>Autosomal recessive polycystic kidney disease (ARPKD), usually considered to be a genetically homogeneous disease caused by mutations in PKHD1, has been associated with ciliary dysfunction. Here, we describe mutations in DZIP1L, which encodes DAZ interacting protein 1-like, in patients with ARPKD. We further validated these findings through loss-of-function studies in mice and zebrafish. DZIP1L localizes to centrioles and to the distal ends of basal bodies, and interacts with septin2, a protein implicated in maintenance of the periciliary diffusion barrier at the ciliary transition zone. In agreement with a defect in the diffusion barrier, we found that the ciliary-membrane translocation of the PKD proteins polycystin-1 and polycystin-2 is compromised in DZIP1L-mutant cells. Together, these data provide what is, to our knowledge, the first conclusive evidence that ARPKD is not a homogeneous disorder and further establish DZIP1L as a second gene involved in ARPKD pathogenesis.</P>

      • Weight Loss Correlates with Macrophage Inhibitory Cytokine-1 Expression and Might Influence Outcome in Patients with Advanced Esophageal Squamous Cell Carcinoma

        Background: Weight loss during chemotherapy has not been exclusively investigated. Macrophage inhibitory cytokine-1 (MIC-1) might play a role in its etiology. Here, we investigated the prognostic value of weight loss before chemotherapy and its relationship with MIC-1 concentration and its occurrence during chemotherapy in patients with advanced esophageal squamous cell carcinoma (ESCC). Materials and Methods: We analyzed 157 inoperable locally advanced or metastatic ESCC patients receiving first-line chemotherapy. Serum MIC-1 concentrations were assessed before chemotherapy. Patients were assigned into two groups according to their weight loss before or during chemotherapy:>5% weight loss group and ${\leq}5%$ weight loss group. Results: Patients with weight loss>5% before chemotherapy had shorter progression-free survival period (5.8 months vs. 8.7 months; p=0.027) and overall survival (10.8 months vs. 20.0 months; p=0.010). Patients with weight loss >5% during chemotherapy tended to have shorter progression-free survival (6.0 months vs. 8.1 months; p=0.062) and overall survival (8.6 months vs. 18.0 months; p=0.022), and if weight loss was reversed during chemotherapy, survival rates improved. Furthermore, serum MIC-1 concentration was closely related to weight loss before chemotherapy (p=0.001) Conclusions: Weight loss both before and during chemotherapy predicted poor outcome in advanced ESCC patients, and MIC-1 might be involved in the development of weight loss in such patients.

      • Zeolite-templated nanoporous carbon for high-performance supercapacitors

        <P>Hierarchical porous carbon prepared with calcium-containing nanocrystalline beta zeolite as the template and ethylene as the carbon source at a relatively low carbonization temperature (600 °C) displayed excellent electrocapacitive properties. The presence of both micro/mesopores and surface oxygen-containing groups on the ordered porous structure, along with a high specific surface area (2280 m<SUP>2</SUP> g<SUP>−1</SUP>) and pore volume (1.95 cm<SUP>3</SUP> g<SUP>−1</SUP>) coupled with good wettability towards the electrolyte enabled the carbon to perform well as a supercapacitor electrode. This carbon electrode achieved a specific capacitance of 250 F g<SUP>−1</SUP> at a current density of 1 A g<SUP>−1</SUP> with 1 M H2SO4 as the aqueous electrolyte measured in a three-electrode system. A symmetric capacitor fabricated with the carbon as both electrodes exhibited a specific capacitance of 161 F g<SUP>−1</SUP> at a current density of 1 A g<SUP>−1</SUP>, and an excellent cycling stability. After being cycled 17 000 times and shelved for another two months, the electrode exhibited a specific capacitance of 246 F g<SUP>−1</SUP> at 1 A g<SUP>−1</SUP>, 153% of the initial capacitance, and still maintained an excellent cycle stability. Besides, an all-solid-state supercapacitor cell fabricated with this carbon as both electrodes and polyvinyl alcohol/H2SO4 gel as the electrolyte displayed an areal specific capacitance of 413 mF cm<SUP>−2</SUP> at a current density of 0.25 mA cm<SUP>−2</SUP>.</P>

      • Sequence variations in <i>OsAGPase</i> significantly associated with amylose content and viscosity properties in rice (<i>Oryza sativa</i> L.)

        <B>Summary</B><P>Sequence-based variations in starch synthesis-related genes (SSRGs) exert a basic influence on the determination of eating quality in rice (<I>Oryza sativa</I> L.). This study aimed to investigate the relationship between the sequence variations from parts of 10 SSRGs and the amylose content (AC) plus rapid viscosity analysis (RVA) profiles in a heuristic rice core set by association mapping (AM). In total, 86 sequence variations were found in 10 sequenced amplicons, including 79 single-nucleotide polymorphisms (SNPs), six insertion-deletions (InDels) and one polymorphic simple sequence repeat (SSR). Among them, 61 variations were exon-based, of which 41 should lead to amino acid changes. Four subpopulations were revealed by population structure analysis based on 170 genome-wide SSR genotypes. The final AM showed a sum of four significant associations between three phenotypic indices and three sequence variations. An <I>ADP-glucose pyrophosphorylase small unit 1</I> (<I>OsAGPS1</I>) SNP (A-G) was significantly associated with increased AC (<I>P</I><0·001, <I>R</I><SUP>2</SUP>=15·6%), while a 12-bp deletion of <I>AGPase large unit 4</I> (<I>OsAGPL4</I>) was significantly related to the decreased breakdown viscosity (BDV) (<I>P</I><0·001, <I>R</I><SUP>2</SUP>=16·6%) in both general linear model (GLM) and mixed linear model (MLM). This study provides a new perspective of allele mining for breeding strategies based on marker-assisted selection.</P>

      • Where Does Trust and Service Quality Reputation Come From?

        The term ‘business reputation’ has certainly moved into the spotlight recently. However, most studies conducted on business reputation have focused on large or multinational enterprises. Within a potentially very large topic, this research concentrates on the perception of the reputation of Taiwanese small enterprises, namely small Chinese restaurants. The motivation for this study stems from the notions about business reputation from customers’ points of view. In addition, significant frameworks reflect conceptual and empirical work which focuses on servicescape, social responsibility, trust and service quality, all of which are relatively common in Western countries. Existing studies which concentrate on Chinese societies are rather more limited and need to be expanded. The purpose of this paper is to fill in some of this research gap by discussing some of the above models in the Chinese cultural society of Taiwan. The results correspond to previous studies conducted, such as the strong relationship between servicescape and service quality, and servicescape being positively connected to trust. The role of trust is critically important in small Chinese restaurants.
