        Preliminary identification of gut microbes between normal and diseased Dorcus titanus castanicolor (Coleoptera: Lucanidae)

        The popularity of keeping stag beetles (Dorcus titanus castanicolor Motschulsky 1861, Coleoptera: Lucanidae) as pets has increased. Consistent with the rise in the number of insect farms using these beetles, the number of contaminated or diseased D. titanus castanicolor has also increased. This investigation was conducted to analyze the cause of D. titanus castanicolor disease. The contaminated larvae of D. titanus castanicolor showed Allomyrina nudivirus infection symptoms similar to those of Allomyrina nudivirus infection. However, the disease carried by of D. titanus castanicolor is not derived from the virus infecting Allomyrina, as determined by PCR. Our study revealed that the major gut microbes of infectious D. titanus castanicolor belonged to the phylum Proteobacteria, and specifically, Pseudomonas knackmussi (Symptom 1 - 39.62% to Symptom 2 - 41.50% to Symptom 3 - 76.76% as the disease progressed severely) and Citrobacter koseri (Symptom 1 - 1.48% to Symptom 2 - 6.04% to Symptom 3 - 6.16% as the disease progressed severely) were detected. Additionally, a high proportion of larvae from the uninfected group were found to harbor bacteria belonging to the phylum Firmicutes (72%). However, as the disease progressed severely in these beetles, the proportion of Firmicutes decreased (Symptom 1 - 72.03% to Symptom 2 - 44.7% to Symptom 3 - 26.3%). These findings imply that colonization by Firmicutes was inversely proportional to Proteobacteria colonization in the gut. This was found to be true for both the normal and disease conditions of D. titanus castanicolor. In this study, we examined the distribution of intestinal microbial communities in normal and contaminated larvae. We observed a correlation between these contaminated microbes and the overall health of the beetle, and our findings suggest that there may be a link between disease progression and the gut microbiome.

        Analysis of the Inhibitory Effect of two Bacterial Strains on Metarhizium anisopliae Induced Fatality Rates in Protaetia Brevitarsis

        Bacterial species, Bacillus amyloliquefaciens and Lactobacillus species (L. sp.5-1), are known to inhibit the growth of pathogenic bacteria and fungi. Metarhizium anisopliae is a pathogenic fungal species which causes fatal damage to P. brevitarsis populations. Therefore, we investigated the inhibitory effect of B. amyloliquefaciens and L. sp. 5-1 on M. anisopliae induced fatality rates in P. brevitarsis. Samples of M. anisopliae-infected sawdust were treated with strain B. amyloliquefaciens KACC10116, strain L. sp. 5-1 KACC19351, and a combination of the two. P. brevitarsis were fed treated sawdust samples, and their subsequent fatality rate was monitored. The fatality rate fell below 1.5% after 10 days and decreased by approximately 40% after 15 days. On average, the fatality rate decreased by 20%, compared to the control. The difference in the decrease in fatality rate between B. amyloliquefaciens treatment and L. sp. 5-1 treatment was not significant. Results indicate that both strains exhibit high anti-fungal activity, which may be useful in environmental purification efforts. These strains may be used for effective prevention of fungal infection in P. brevitarsis.

      • Prevention of Oryctes rhinoceros Nudivirus in Allomyrina dichotoma using fermented sawdust combined Mulberry leaves and its effects in the outdoor application test

        The purpose of this study was to determine the safe and effective method for preventing the occurrence of Oryctes rhinoceros Nudivirus in Allomyrina dichotoma on agricultural farms. There is a high demand for the use of A. dichotoma larvae in animal feed and as pet for educational purpose. Recently, we reported that OrNV is fatal virus diagnosed in A. dichotoma larvae in local farms in Korea. Mulberry leaves contain 1-deoxynojirimycin that represented anti-inflammatory, anti-virus, and anti-tumor effects. To prevent OrNV, we have fed the sawdust combined with 1% and 5% mulberry leaves powder to OrNV infected 2nd and 3rd stage larvae of A. dichotoma, and identified the mortality rate(%) during ten weeks. As a results, the 2nd stage larvae which were fed the sawdust combined with 5% mulberry leaves treatment recorded 60% mortality rate after ten weeks compared to the 100% mortality rate in the control. And the fatality rate of 3rd stage larvae which were fed 5% mulberry leaves treatment decreased 70% compared to the control. Therefore, application sawdust combined with mulberry leaves might be effective in the prevention and control of OrNV disease in A. dichotoma. Additionally, in the ten insect breading farms application test, OrNV virus disease have not appeared in the mulberry leaf powder treatment group.
