축산폐수 처리를 위한 회전원판 생물막 공정에 관한 연구

고농도 유기물 축산폐수의 유기물 제거효율 및 설계를 위한 동력학인자를 도출하는 연구를 하였다. 유기물 제거율은 RBC 반응기 내에서 수리학적 체류시간이 0.79일, 1.2일 및 1.98일에 대하여 각각 65%, 71%, 85%의 COD 제거율을 나타내었다. RBC 반응기 설계를 위한 동력학 인자 연구에서 P와 Ks는 각각 25,000 mg/㎡-day와 1,023 mg/L로 도출되었다. The organic removal efficiency and the kinetic parameters for design of the livestock wastewater with high organic loading using the RBC was investigated. The COD removal efficiency was 65%, 71%, and 85%, respectively, which was operated 0.79 day, 1.2 day, and 1.98 day of hydraulic retention time. The kinetic parameters for design of RBC found that P and Ks were 25,000 mg/㎡ -day, and 1,023 mg/L, respectively.

Hemoglobin (Hb) is a member of heme-protein that can perform catalytic non-specific chain reaction in the presence of hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>). Catalytic ability of Hb to degrade pyrene was demonstrated using soil contaminated with 14C pyrene and 10 mg pyrene /kg soil. The composition of soil was similar to previously used soil except that it had lower organic carbon content. Bench scale laboratory tests were conducted in the presence of buffer only, H<sub>2</sub>O<sub>2</sub> only, or Hb with H<sub>2</sub>O<sub>2</sub> for 24 h. After 24 h reaction, 0.1 and 1.3% of <sup>14</sup>C pyrene in contaminated soil were mineralized with H<sub>2</sub>O<sub>2</sub> only or Hb plus H<sub>2</sub>O<sub>2</sub>. No mineralization to <sup>14</sup>CO<sub>2</sub> was detected with buffer only. Approximately 12.2% of pyrene was degraded in the presence of H<sub>2</sub>O<sub>2</sub> only while 44.0% of pyrene was degraded in the presence of Hb plus H<sub>2</sub>O<sub>2</sub> during 24 h of catalytic reaction. When degradation intermediate products were examined, two chemicals were observed in the presence of H<sub>2</sub>O<sub>2</sub> only while 25 chemicals were found in the presence of Hb plus H<sub>2</sub>O<sub>2</sub>. While most degradation products were simple hydrocarbons, four of the 27 chemicals had aromatic rings. However, none of these four chemicals was structurally related to pyrene. These results suggest that Hb catalytic system could be used to treat pyrene-contaminated soil as an efficient and speedy remediation technology. In addition, intermediate products generated by this system are not greatly affected by composition change in soil organic matter content.

<P><B>Abstract</B></P> <P>This study investigates the efficiency of hemoglobin (Hb)-catalyzed biocatalytic reactions for removal of polycyclic aromatic hydrocarbons (PAHs) in a historically PAHs-contaminated soil and of benzo(<I>a</I>)pyrene (BaP) in an artificially BaP-contaminated soil. PAHs removal tests at various H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratios (0–3.7) showed that the PAHs removal was greater at H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio of ≥3. This was attributed to the greater removal of high molecular weight PAHs at higher H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratios. The BaP removal increased from 36% to 85% with increasing H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio from 1 to 3, and further increase in H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio decreased the BaP removal. Thus, the optimal H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio for BaP removal was determined to be 3 in the artificially BaP-contaminated soil. The BaP removal in the presence of only Hb can be attributed to the capture of BaP by Hb. The increased BaP removal in the presence of H<SUB>2</SUB>O<SUB>2</SUB> is likely due to BaP mineralization as the BaP removal and the CO<SUB>2</SUB> generated showed a strong positive correlation (R<SUP>2</SUP> = 0.999). Overall, this study shows that the Hb-catalyzed biocatalytic reactions can effectively remove PAHs in soil.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PAHs removal in a contaminated soil was greater at H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio ≥3. </LI> <LI> Greater high molecular weight PAHs were removed at higher H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratios. </LI> <LI> The optimal H<SUB>2</SUB>O<SUB>2</SUB>-to-Hb mass ratio for BaP removal in soil was determined to be 3. </LI> <LI> BaP removal in the presence of only Hb can be attributed to the BaP capture by Hb. </LI> <LI> BaP removal and CO<SUB>2</SUB> generation was positively correlated supporting mineralization. </LI> </UL> </P>
