Activities of Natural Plant Extracts against HIV-1
Eum, Jin-Seong,Park, Young-Doo,Hong, Seong-Karp The Korea Institute of Information and Commucation 2009 Journal of information and communication convergen Vol.7 No.4
Anti-HIV-1 activities for the extracts (buthanol, hexane, chloroform, and water) of medicinal plants widely used in the folk medicine were evaluated for screening of anti-AIDS agents. The activities of the extracts to inhibit HIV-1 replication were also analyzed. The 50% effective concentration (EC50) of inhibition activity of the p24 production for chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph showed 5.8, 29.9, and 37.3 2g/ml, respectively, as good activities. Hexane extract of Sophora flavescens, buthanol extract of Tulipa edulis, hexane extracts of Tulipa edulis, Herba ephedra, and Pachyma hoelen Rumph in the 50% cytotoxic concentration ($CC_{50}$) in inhibition activities of recombinant HIV-1 RT showed 12.9, 19.5, 11.6, 12.0, and 36.8 % at concentration of 200 ${\mu}g$/ml, respectively, as good activities. From these results, chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph were very effective against HIV-1 among all extracts tested. Therefore, we expect these plants will be a useful for anti- HIV-1 therapeutics in future.
The Mutagenesis in the virE Promoter of the octopine Ti Plasmid from Agrobacterium tumefaciens A348
Eum, Jin-seong 목원대학교 자연과학연구소 1999 自然科學 硏究論文集 Vol.8 No.-
The virE promoter of octopine plasmid pTiA6 has a vir box and an inverted repeat sequence. To investigate how the inverted repeat sequence of the virE promoter plays a role in virE gene expression, two virE recombinant plasmids were constructed by base substituted and deleted mutagenesis in the inverted repeat sequence of virE promoter. The base substituted mutant showed the level of β-galactosidase activity about 3% of the wild type virE promoter activity. And the activity of deleted mutant displayed β-galactosidase activity at 6% of the wild type virE promoter activity. These results indicated that the inverted repeat sequence in the virE promoter is necessary for the expression of virE.
Expression of the mutated virE promoter in Agrobacterium tumefaciens A348 and KU12
Eum, Jin Seong,Park, Young Doo 목원대학교 자연과학연구소 1997 自然科學 硏究論文集 Vol.6 No.-
After the transformation of pSM358cd into Agrobacterium tumefaciens KU12 which was isolated in Korea, vir expression was induced with acetosyringone and β-galactosidase activity was measured. Its activity was 48.7% of that of A. tumefaciens A348. Thus it is thought that vir expression with acetosyringone in A. tumefaciens KU12 is lower than that of A. tumefaciens A348. In order to identify the important regulatory sites of virE located in A. tumefaciens A348, virE was partially deleted and recombinated. It's expression was induced by acetosyringone and the activities of the mutated virE recombinant plasmids were measured. The β-galactosidase activity of the mutated virE recombinant plasmids, pMW20 and pMW30, showed at 65.2% and 1.3% of the native virE promoter activity, respectively. According to the analysis of the mutated virE recombinant plasmids, about 120-240 bps upstream of native virE promoter is thought to be related with an important regulatory site.
