Cytoprotective Role of the Flavonoid Quercetin in Mammalian Cells

The unfolded protein response (UPR) is an intracellular signal transduction mechanism that is activated when unfolded proteins acumulate in the endoplasmactivated UPR induces transcription of ER resident chaperones and folding enzymes, atenuates translation of general proteins, and induces genes that play roles in protein degradation. Depending on the ER stres level, the UPR decides whether cells live or die. In t h i s s t u d y , w e t e s t e d w h e t h e r f l a v o n o i d s a f e c t t h e U P R a n d c e ll viability under stres ltured mamalian cells from death under ethanol stres and doxorubicin stres, probably by activating the UPR. Quercetin treatment increased the viability of both ethanol-treated human HepG2 hepatocytes and doxorubicin-treated rat H9c2 myocardiocytes. Quercetin treatment induced the expresion of Grp78/BiP, one of the UPR target genes, increased splicing of Xexpression of the UPRE reporter gene. Interestingly, ectopic expresion of Grp78/BiP protected cels under ethanol and doxorubicin stres from death, suggesting that the flavonoid quercetin somehow activates the UPR in mamalian cells and plays a protective role under certain stres conditions.
