Cronobacter spp. in powdered infant formula has been etiologically linked to meningitis and necrotizing enterocolitis in certain groups of infants. This study aimed to determine whether C. sakazakii Yrt2a strain experiencing desiccation stress could enter viable but nonculturable (VBNC) state as well as to examine the expression of genes associated with stress and virulence during the above states. Stress and VBNC conditions were determined based on viability and culturability assays. Expression of genes related to stress (rpoS) and virulence (hfq and ompA) was evaluated by real-time PCR. The results showed that C. sakazakii Yrt2a entered VBNC 24 days post exposure to 2 h of desiccation treatment. The expression of rpoS, hfq and ompA genes was up-regulated during stress conditions, suggesting that Cronobacter successfully managed stress to maintain its culturability while maintaining its virulence. The expression of the target genes decreased at VBNC state but remained higher than that of a normal state. These findings reinforce the assumption that C. sakazakii undergoing VBNC state maintains its pathogenicity.

Stability and Growth Characteristics of GFPuv- Labeled Cronobacter sakazakii Isolated from Foods

Stability of ultraviolet green fluorescent protein(GFPuv)-labeled Cronobacter sakazakii and C. muytjensiiisolated from foods and the effects of the plasmid GFPuv(pGFPuv) on growth were analyzed. PCR analysis andmicroscopic observation showed that C. sakazakii and C. muytjensii isolates took up the plasmid into cells andexpressed the GFPuv gene. All Cronobacter transformantsmaintained this plasmid after frozen storage and 2consecutive subcultures. The C. sakazakii FWHd16transformant was the most stable, while the C. muytjensiiFWHd11 transformant was the least stable. All transformantsshowed nearly identical growth curves during lag, log andstationary phases, compared to wild type parental isolates. The maximum bacterial growth rates (μmax) of thetransformants and parents were similar, indicating that thepresence of pGFPuv in transformants did not affect cellgrowth. Stable GFPuv-labeled Cronobacter has potentialfor use in tracking bacterial behavior during food handlingand drying.

This research aimed to evaluate the effect of acidstress on the expression of stress regulator (grxB and rpoS)and virulence (ompA, hfq, and cpa) genes of Cronobactersakazakii Yrt2a. The results showed that C. sakazakii Yrt2aexperienced decrease in number during acid stress and wasno longer culturable 90 min post exposure to pH 3.0. During acid stress, the expression of grxB, rpoS, ompA, cpaand hfq was upregulated by 2.15; 2.19; 1.55; 1.1 and 1.41log, respectively. However, all genes expression wasdownregulated when the bacteria entered the unculturablestate. The expression of gene grxB, rpoS, ompA, cpadecreased to 1.04; 0.37; 0.84 and 1.71 log, respectively;while hfq gene expression reached a level lower than thatof control. This research implies a supposition that duringacid stress, C. sakazakii was capable of maintaining itsculturability and pathogenicity until they are no longerculturable.
Pediatric Quality of Life in Congenital Hypothyroidism: an Indonesian Study

Background and Objectives: Thyroxine is important for brain development. Improper hypothyroid treatment maylead to cognitive and motor impairment, thereby affecting the quality of life. We analyzed the correlation betweenage at first treatment, length of treatment, initial levothyroxine (LT4) dose, and serum levels of free thyroxine(fT4) and thyroid stimulating hormone (TSH) and pediatric quality of life in patients with congenital hypothyroidism(CH). Materials and Methods: This research was a cross-sectional study of 41 children with CH who consumedLT4 for at least 3 months during March 2019-December 2019. The quality of life was assessed from parents’reports using the Pediatric Quality of Life Inventory (PedsQL) generic scale. Spearman correlation analysis wascarried out, and statistical significance was set at p<0.05. Results: A total of 17 of the 41 children were girls. The mean PedsQL scores in physical and psychosocial functioning were 78.12 (68.75-100) and 233.30(215-251.67), respectively. Age at first treatment was correlated with physical functioning (r=−0.501, p<0.05)and psychosocial functioning (r=−0.440, p<0.05). The initial LT4 dose was negatively correlated with physicalfunctioning (r=−0.568, p<0.05) and psychosocial functioning (r=−0.482, p<0.05). The length of treatmentshowed a positive correlation with physical functioning (r=0.776, p<0.05) and psychosocial functioning (r= −0.852, p<0.05). However, the serum fT4 and TSH levels were not correlated with quality of life in children withCH (p>0.05). Conclusion: Age at first treatment, initial dose of LT4, and length of treatment were correlatedwith quality of life in children with CH.
Expression of somatic embryogenesis-related genes in sugarcane (Saccharum officinarum L.)

The low frequency of plantlet regenerates from the somatic embryogenesis (SE) callus in sugarcane becomes a problem to produce its seed. Plant growth regulators were able to increase the regeneration frequency of SE to normal plantlets, such as 2,4-dichlorophenoxyacetic acid (2,4-D) as promoting callus induction. Since molecular mechanisms involved SE in sugarcane have not been reported, expression of Baby Boom (BBM) and Leafy Cotyledon (LEC) genes related to SE had investigated. The effect of diff erence concentration of 2,4-D on callus induction and expression of somatic embryogenesis-related genes (BBM and LEC) in sugarcane were important information for the increasing quantity and quality of seed production. The percentage of callus formation and embryogenic callus in the Murashige and Skoog’s (MS) medium contained 4 mg/L 2,4-D after 6 weeks cultivation were 76% and 86%, respectively. The MS + 4 mg/L 2,4-D medium was recommended for the large-scale embryogenic callus production from sugarcane explant. The high-level expression of BBM and LEC was shown in the embryogenic callus, which suggested that the expressions of both genes were believed to play on the somatic embryogenesis regulation in sugarcane.
Metaverse Tourism: Elements and Consequences on Tourism Experience Journey

Prior research on Extended Reality (XR) has been a starting point for the development of the Metaverse. It is also a concern for researchers in the tourism field since it can enhance the tourist experience. Metaverse Tourism provides a virtual experience of the tourism destination for the users. It is potentially developed and predicted to be the future of tourism, enhancing the tourist experience through communication between humans and avatars and providing a new option for tourists to enjoy leisure activities. Recently, discussion on Metaverse tourism is still emerging, with some open talks from researchers in the intersections between information technology and tourism. This study extends prior studies by proposing six Metaverse tourism elements and categorizing them into core and supporting elements. Then, we analyze the consequences of those elements on the Tourism Experience Journey to enhance each stage with different characteristics. This study can contribute to continuing recent dialogues among scholars on how Metaverse can be applied in the tourism sector by proposing six elements that should be considered in developing and creating Metaverse services.

Indonesian kefir grains are potential sources of lactic acid bacteria (LAB) that may act as probiotics with specific functional properties. In this study we explored the potential of the probiotic and cholesterol-lowering effect of LAB isolated from Indonesian kefir grains obtained from Bogor, Bandung, Jakarta, and Yogyakarta. The results revealed that 10 isolates showed considerable survivability at low pH and bile salt with total cell reduction of ~3 log colony-forming units per milliliter after exposure to pH 2.5 and 0.5% (w/v) bile salt for 1 and 3 h, respectively. All strains exhibited strong antimicrobial activities against pathogenic bacteria and were sensitive to a wide spectrum of antibiotics but exhibited weak bile salt hydrolase activity. Identification based on 16S RNA suggested that nine isolates were Lactobacillus kefiri and one was Lactobacillus rhamnosus. The ability of the isolates to reduce cholesterol from the media varied, ranging from 22.08% to 68.75% with the highest reduction shown by L. kefiri JK17. The ability to remove cholesterol from the media decreased greatly in resting and dead cells, ranging from 14.58% to 22.08% in resting cells and from 7.89% to 18.17% in dead cells. It can be concluded that Indonesian kefir grains contain LAB potentially acting as probiotics capable of reducing cholesterol. The cholesterol-lowering effect especially occurs when the cells are metabolically active.
The Effect of Entrepreneurial Learning towards Entrepreneurial Intention of Indonesian Women

Entrepreneurship has been the center of a worldwide phenomenon as it has contributed a lot in the economy. The researcher is trying to acknowledge the interest of Indonesian women to build their own businesses. The aim of this research is to see how entrepreneurial learning could influence Indonesian women to grow interest in doing entrepreneurial activity, with the help of three mediations: attitude toward entrepreneurship, subjective norms, and perceived behavioral control. The research will use quantitative method and SmartPLS 3.0 to support the statistical data. The respondents are Indonesian women ranging around 17 years old and above who have had the experienced entrepreneurship learning in educational institutions. To show a more accurate result, purposive sampling will be used where it is very objective and selective to help the researcher in providing the justification to make generalizations from the investigated sample from a total of 149 respondents from various regions in Indonesia. The findings on this research are entrepreneurial learning is influencing the entrepreneurial intention of Indonesian women with the help of two mediations, which are attitude toward entrepreneurship and perceived behavioral control. Subjective norms is found has no significant influence on growing the entrepreneurial intention of Indonesian women.
