      • Improving Classification Accuracy Using Missing Data Filling Algorithms for the Criminal Dataset

        Predicting crime types by using classification algorithms can help to find factors affecting crimes and prevent crimes. Due to various reasons in the process of data collection, there are often a large number of missing values in actual criminal dataset, which seriously affects the classification accuracy. Therefore, based on mutual KNNI (K nearest neighbor imputation) algorithm and combined with GRA (Grey Relational Analysis) theory, a novel data filling algorithm called GMKNN is proposed in order to improve the classification accuracy. The algorithm replaces the Euclidean distance formula used in mutual KNNI algorithm with the Grey relational grade formula to eliminate the effect of noise from the nearest neighbors and effectively deal with the discrete attributes. By comparing with several popular data filling algorithms based on a real criminal dataset with lots of missing values, higher classification accuracy can be obtained by using GMKNN algorithm, which is up to 77.837%.

        Imaging Manifestations and Misdiagnosis Analysis of Six Cases of Bone Hydatid Disease

        We retrospectively evaluated the clinical and imaging features of 6 patients with bone hydatid disease confirmed by surgery and pathological examination. Among the 6 patients, 2 were infected with Echinococcosis granulosus metacestode and 4 were infected with E. multilocularis metacestode. The 2 cases with cystic echinococcosis were diagnosed by computed tomographic (CT) examination, and other 4 cases were diagnosed by magnetic resonance (MR) imaging. On the initial evaluation, 1 case each was misdiagnosed as a giant cell tumor or neurogenic tumor, and 2 were misdiagnosed as tuberculosis. The imaging manifestations of bone hydatid disease are complex, but most common findings include expansive osteolytic bone destruction, which may be associated with sclerosing edges or dead bone formation, localized soft tissue masses, and vertebral lesions with wedge-shaped changes and spinal stenosis. Combining imaging findings with the patient’s epidemiological history and immunological examinations is of great help in improving the diagnosis and differential diagnosis of bone hydatid disease.

        Recombinant human β-defensin130 inhibited the growth of foodborne bacteria through membrane disruption and exerted anti-inflammatory activity

        Foodborne pathogens causing food poisoning and infections are detrimental to human health, and the abuse of antibiotics induced severe antibiotic resistance in past decades. Thus, it is urgent to develop new antimicrobial agents. In the current study, human β-defensin 130 (hBD130), which is an antimicrobial peptide identified in human macrophages in 2017, was initially produced in Pichia pastoris. The purified hBD130 demonstrated broad bactericidal spectrum against foodborne pathogens through a membrane disruption, with concentrations ranging from 10 to 45 μg/mL. Moreover, hBD130 showed a low hemolytic effect and nearly no cytotoxicity to mammalian cells with a dosage of 400 μg/mL. In addition, the secretion amounts and mRNA levels of NO, IL-6, IL-1β, and TNF-α in LPS-induced mouse macrophage were significantly decreased with 1 mg/mL of hBD130. Taken together, these results showed that hBD130 is a promising antimicrobial agent to treat foodborne bacterial infections and inflammation.

        Antibacterial activity of antimicrobial peptide gcDefb1 against foodborne pathogenic bacteria and its application in pork storage

        Pork and its products are preferably contaminated by bacteria; thus, it is essential to develop low-cost, high-efficiency and biologically safe preservatives to prevent the growth of bacteria during storage. In the current study, grass carp β-defensin 1 (gcDefb1) was produced and purified from Pichia pastoris through the heterologous expression method. The in vitro antimicrobial assay demonstrated that yeast-derived gcDefb1 possesses a broad antibacterial spectrum, including both Gram-positive and -negative bacteria, and the MIC values against Escherichia coli ATCC 25,922 were as low as 30 μg/mL and showed no cytotoxicity or hemolytic activity. The bactericidal rate of gcDefb1 was less than 60 min by disrupting the cell membranes, and it inhibited the formation of bacterial biofilms. Moreover, gcDefb1 was used as a biopreservative for pork storage, indicating that the physicochemical and sensory qualities were improved. This study provides an efficient method to prepare and utilize gcDefb1 as a novel biopreservative.

        Antimicrobial property of recombinant Lactolisterin BU in vitro and its initial application in pork refrigerated storage

        Lactolisterin BU is a novel bacteriocin identified from Lactococcus lactis in 2017. It exhibits antimicrobial activity against food spoilage and foodborne pathogens. In this study, Lactolisterin BU was expressed in Pichia pastoris (P. pastoris) and isolated from the supernatant of yeast culture for the first time. It was found to exhibit a broad antimicrobial spectrum and rapid bactericidal activity against foodborne bacterial pathogens, both gram-positive and gramnegative ones, with minimum inhibition concentrations ranging within 10–60 μg/mL. The recombinant Lactolisterin BU (rLactolisterin BU) also had an antioxidant effect and was resistant to heating, acid–base, and high-dose-saline treatments and barely had any hemolytic activity or cytotoxicity. Moreover, rLactolisterin BU effectively suppressed the growth of bacterial pathogens; suppressed the increases in pH, total volatile basic nitrogen (TVB-N), and thiobarbituric acid reactive substances (TBARS) of pork samples; and maintained a high quality of fresh pork during storage at 4 ℃. Furthermore, rLactolisterin BU effectively inhibited the growth of three kinds of bacteria in a pork-spoilage model. Taken together, rLactolisterin BU could be a promising preservative for food storage.
