Purpose: To report a case of combined laparoscopic and carbon dioxide partial cystectomy and foreign body removal and to review the existing literature on the topic. Materials and Methods: A 43-year-old Asian woman was found to have an intrauterine device embedded in the bladder wall during evaluation for chronic pelvic pain and urinary tract infection. She underwent cystoscopic-laparoscopic partial cystectomy, with an uncomplicated postoperative course. She had normal renal function during the follow-up period. This case demonstrates the possibility and safety of performing cystoscopic-laparoscopic partial cystectomy for the removal of a partially implanted intravesical foreign body. Results: The patient recovered without incident and was discharged 7 days after surgery. No abnormalities were noted in the urine output or renal function in the postoperative follow-up period. No complications due to retrograde flow of carbon dioxide up the ureters or air embolism were noted during the procedure or postoperatively. Conclusions: The combination of laparoscopy and air cystoscopy has been shown to be an optimal method for retracting foreign bodies embedded in the bladder wall. Also, air cystoscopy can be used to give doctors a better view in cases in which vision is compromised under water-contrast cystoscopy.

Gardenia jasminoides Ellis, a representative for “homology of medicine and food”, can be used to produce pigment and edible oil. Here, aqueous enzymatic extraction (AEE) combined with puffing pre-treatment was explored to prepare oil from gardenia seeds. Both wet-heating puffing (WP) at 90 °C and dry-heating puffing (DP) at 1.0 MPa facilitated the release of free oil by AEE, resulting in the highest free oil yields (FOY) of 21.8% and 23.2% within 3 h, much higher than that of un-puffed group. Additionally, active crocin and geniposide were also completely released. The FOY obtained was much higher than mechanical pressing method (10.44%), and close to solvent extraction (25.45%). Microstructure analysis indicated that gardenia seeds expanded by dry-heating puffing (1.0 MPa) had a larger, rougher surface and porous structure than other groups. Overall, AEE coupled with puffing pre-treatment developed is an eco-friendly extraction technology with high efficiency that can be employed to oil preparation.

<P>To investigate the role of histone H3K27 demethylase UTX in embryonic stem (ES) cell differentiation, we have generated <I>UTX</I> knockout (KO) and enzyme-dead knock-in male ES cells. Deletion of the X-chromosome-encoded <I>UTX</I> gene in male ES cells markedly decreases expression of the paralogous <I>UTY</I> gene encoded by Y chromosome, but has no effect on global H3K27me3 level, <I>Hox</I> gene expression, or ES cell self-renewal. However, <I>UTX</I> KO cells show severe defects in mesoderm differentiation and induction of <I>Brachyury</I>, a transcription factor essential for mesoderm development. Surprisingly, UTX regulates mesoderm differentiation and <I>Brachyury</I> expression independent of its enzymatic activity. UTY, which lacks detectable demethylase activity, compensates for the loss of UTX in regulating <I>Brachyury</I> expression. UTX and UTY bind directly to <I>Brachyury</I> promoter and are required for Wnt/β-catenin signaling-induced <I>Brachyury</I> expression in ES cells. Interestingly, male <I>UTX</I> KO embryos express normal levels of <I>UTY</I> and survive until birth. In contrast, female <I>UTX</I> KO mice, which lack the <I>UTY</I> gene, show embryonic lethality before embryonic day 11.5. Female <I>UTX</I> KO embryos show severe defects in both <I>Brachyury</I> expression and embryonic development of mesoderm-derived posterior notochord, cardiac, and hematopoietic tissues. These results indicate that UTX controls mesoderm differentiation and <I>Brachyury</I> expression independent of H3K27 demethylase activity, and suggest that UTX and UTY are functionally redundant in ES cell differentiation and early embryonic development.</P>
