        Relationship of Nitrate Reductase Activity to Leaf Yield, Protein, Sugar and Physiological Attributes in Mulberry (Morus alba L.)

        Ten improved mulberry varieties (Vl, C1730, C2016, C2017, Anantha, RFS-175, Thallaghatapura, Vishala, S1 and S1635) were evaluated through enzyme assay and estimation of soluble protein content followed by regression analysis, grown under irrigated conditions in the alluvial soils of Gangetic plains of West Bengal in India for five successive crops in a year. The nitrate reductase (EC No. 1.6.6.1) activity (NRA, μmol NO2-h-1 g-1 fr, wt.), total soluble protein (mg g-1 fr, wt.) was estimated which showed to vary significantly in the tested varieties. In addition to these, the other parameters like unit leaf fresh and dry weight (g), moisture %, unit leaf area (㎠), specific leaf weight (g ㎝-2), total soluble sugar (mg g-1 fr, wt.), leaf yield/plant (kg), shoot yield/plant (kg) and net photosynthetic rate (NPR, μ㏖ m-2 g-1) were also studied which showed to vary significantly in tested varieties. Among them, S1635, haying higher NRA (13.25 μ㏖ NO2-h-1g-1 fr, wt.), total soluble protein (39.63 mg g-1 fr, wt.), NPR (16.66 μ㏖ m-2 S-1), total soluble sugar (48.44 mg g-1 fr. wt.), leaf yield/plant (0.689 kg) and shoot yield/plant (1.135 kg) showed its superiority over other tested varieties. Regression and correlation coefficients were analysed, and a strong positive correlation was found between NRA & total soluble protein, NRA & NPR, NRA & total soluble sugar, NRA & unit leaf weight, NRA & specific leaf weight, NRA & leaf yield/plant, NRA & shoot yield/plant, NPR & leaf yield and NPR & specific leaf weight.

      • Synthesis and self-assembly of amphiphilic and biocompatible poly(vinyl alcohol)-block-poly(l-lactide) copolymer

        Well-defined amphiphilic and biocompatible poly(vinyl alcohol)-block-poly(l-lactide) copolymers (PVA-b-PLLA) were synthesized by the combination of reversible addition-fragmentation chain transfer (RAFT), ring opening polymerization (ROP), and click chemistry. First, an innovative azide-functionalized xanthate mediated chain transfer agent (CTA) [S-(3-azidopropyl propanamide)-(O-butyl xanthate)] was synthesized and used for vinyl acetate (VAc) polymerization through RAFT. Then, azide-terminated poly(vinyl acetate) (azide-PVAc) was converted to the corresponding azide-terminated poly(vinyl alcohol) (azide-PVA) through hydrolysis. Alkyne-terminated poly (l-lactide) (alkyne-PLLA) was synthesized via ROP using a bifunctional initiator, having an alkyne and a hydroxyl group. Finally, azide-PVA and alkyne-PLLA were coupled by 1,3 cycloaddition click reaction to get PVA-b-PLLA. Synthesized azide-PVAc, azide-PVA, alkyne-PLLA and PVA-b-PLLA were characterized by size exclusion chromatography and <SUP>1</SUP>H NMR spectroscopy. The self-assembly of PVA-b-PLLA in water was investigated by transmission electron microscope, atomic force microscopy, dynamic light scattering and <SUP>1</SUP>H NMR spectroscopy. Spherical micelles in water were clearly observed. The critical micelle concentration of PVA-b-PLLA in water was determined by fluorescence spectroscopy, and it was decreased with increasing the molecular weight of PLLA block. PVA-b-PLLA exhibited low cytotoxicity which could be used in biomedical application.

      • Utilisation of Municipal Waste Water in Aerobic Composting of Solid Organic Waste of Bhubaneswar City

        Approximately 600 tons of solid organic wastes and a huge bulk of sewage water are generated in Bhubaneswar city, Orissa, India daily endangering the urban environment. Solid wastes in windrows with sewage water inoculum decomposed faster compared to cow dung slurry and plain water treated wastes. C:N and C:P ratios declined relatively faster in sewage water treated windrows than cow dung slurry and water treated windrows during the experimental period of 45 days. The results indicated that the Bhubaneswar Municipal sewage water would be successfully utilised in solid waste composting.

        Maximizing Biomass Productivity and CO2 Biofixation of Microalga, Scenedesmus sp. by Using Sodium Hydroxide

        A series of experiments were carried out with three native strains of microalgae to measure growth rates, biomass, and lipid productivities. Scenedesmus sp. IMMTCC-6 had better biomass growth rate and higher lipid production. The growth, lipid accumulation, and carbon dioxide (CO2) consumption rate of Scenedesmus sp. IMMTCC-6 were tested under different NaOH concentrations in modified BBM. The algal strain showed the maximum specific growth rate (0.474/day), biomass productivity (110.9 mg l-1d-1), and CO2 consumption rate (208.4 mg l-1d-1) with an NaOH concentration of 0.005 M on the 8th day of cultivation. These values were 2.03-, 6.89-, and 6.88-fold more than the algal cultures grown in control conditions (having no NaOH and CO2). The CO2 fixing efficiency of the microalga with other alternative carbon sources like Na2CO3 and NaHCO3 was also investigated and compared. The optimized experimental parameters at shake-flask scale were implemented for scaling up the process in a self-engineered photobioreactor. A significant increase in lipid accumulation (14.23% to 31.74%) by the algal strain from the logarithmic to stationary phases was obtained. The algal lipids were mainly composed of C16/C18 fatty acids, and are desirable for biodiesel production. The study suggests that microalga Scenedesmus sp. IMMTCC-6 is an efficient strain for biodiesel production and CO2 biofixation using stripping solution of NaOH in a cyclic process.

        Two new crystalline coordination polymers have been synthesized with Zn<SUP>2+</SUP> and Cd<SUP>2+</SUP> ions using an N-(4-(pyridinyl-4-ethynyl)phenyl)-isonicotinamide (HL) donor ligand. The ligand readily reacts with metal nitrate in DMF to afford 1-dimensional (1D) coordination polymers 1 and 2 comprising a repeating unit of [Zn(HL)(NO<SUB>3</SUB>)<SUB>2</SUB>(DMF)].DMF and [Cd(HL)(NO<SUB>3</SUB>)<SUB>2</SUB>(DMF)].DMF, respectively. Both complexes 1 and 2 were characterized by IR spectroscopy, elemental analysis, thermogravimetric analysis (TGA), and X-ray diffraction. Both complexes contained a 1D chain in which crystallography revealed two metal ions were connected by each ligand. The weak π-π stacking interactions and intermolecular hydrogen bonds resulted in the one-dimensional network structures. The photoluminescent properties for both complexes were also investigated.

        Effect of Sodium Hydroxide plus Hydrogen Peroxide Treated Mustard (Brassica campestris) Straw Based Diets on Rumen Degradation Kinetics (In sacco), Fermentation Pattern and Nutrient Utilization in Sheep

        Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.
