        Enhanced accumulation of triterpenoid saponins in in vitro plantlets and dedifferentiated cultures of Eryngium planum L.: a medicinal plant

        We investigated the conditions leading to the accumulation of saponins in plants using in vitro cultures of Eryngium planumL. The aim was to establish a system that produced saponins of the same quality as those found in fi eld-grown plants and toenhance their accumulation in the laboratory. We observed that the content of saponins in organs from in vitro plantlets andplants grown in an experimental plot after micropropagation was higher (up to 8.7 times) compared with plants from naturalsites. Untreated callus accumulated saponins in amounts similar to those of the roots from fi eld-grown plants. Applyingsucrose to the growth medium at higher concentrations (40, 50, or 60 g L −1 ) resulted in enhanced saponin production in callus(1.2–1.7-fold). The elicitation with 100 μmol L −1 methyl jasmonate of callus grown on Murashige and Skoog medium supplementedwith 30 g L −1 sucrose resulted in a 1.2-fold increased saponin accumulation compared with callus maintained onthe same medium (30 g L −1 sucrose). This study provides the fi rst evidence that triterpenoid saponins can be biosynthesizedin in vitro plantlets and dediff erentiated cultures of E. planum .
