        Antioxidant and free radical scavenging activities of Cleome rutidosperma

        The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro nonenzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.         In vivo evaluation of anti-diarrheal activity of the rhizome of Nymphaea alba (Nymphaeaceae)

        Ethanol extract of the of rhizome of Nymphaea alba, at graded doses was investigated for anti-diarrhoeal activity in term of reduction in the rate of defecation in castor oil induced diarrhoea and anti-inflammatory activity in term of reduction in inflammation of rat paw. To understand the mechanism of its anti-diarrhoeal activity, its effect was further evaluated on intestinal transit and castor oil induced intestinal fluid accumulation (enteropooling). At various doses (100 & 200 mg/kg body weight) the extract showed a remarkable anti-diarrhoeal activity evidenced by the reduction in the rate of defecation. Results are comparable to that of standard drug diphenoxylate (5 mg/kg body weight). A single oral dose of N. alba extract of 100 mg/kg body weight produced a significant decrease in the severity of diarrhoea. Extract produced profound decrease in intestinal transit (25.73 and 37.29%) also significantly inhibited castor oil induced enteropooling comparable to that of intraperitoneal injection of standard drug atropine sulphate at doses of 0.1 mg/kg body weight and loperamide at 5 mg/kg body weight respectively. Results indicate N. alba possess significant anti-diarrhoeal activity and may be a potent source of anti-diarrhoeal drug in future. But the extract did not show anti-inflammatory activity.

        Development of standardization parameters of crude drugs used in Ayurvedic Balarista formulation

        ‘Balarista’ is an important Ayurvedic arista formulation containing twelve crude drugs among which Sida cordifolia and Withania somnifera are the two major drugs. The Ayurvedic Formulary of India mentions various usages of Balarista, but it is mainly used in the treatment of rheumatoid arthritis. This study deals with the evaluation of pharmacognostic and physico-chemical properties of all incorporated drugs along with the HPLC and GC–MS analysis of the two main drugs. The macroscopic, microscopic, powder microscopy and physico-chemical parameters of diferent drugs were assessed according to the Ayurvedic Pharmacopoea of India. The scanning electron microscopic study, fuorescence analysis and powder behavior studies of various drugs in powder form were also carried out. The quantifcation of vasicine and total withanolides were analyzed in chloroform extract of Sida cordifolia and Withania somnifera root by HPLC method. Further, presences of other phytoconstituents were also confrmed by GC–MS analysis. The macroscopical, organoleptic, histological, powder characteristics and scanning electron microscopic images of various drugs revealed the important diagnostic features. The physico-chemical, fuorescence analysis and powder behavior characteristics provided useful tools to diferentiate them from the adulterants. The HPLC analysis detected percentage of vasicine and total withanolide (withanoside IV, withanoside V, withaferin A, 12-Deoxy withastramonolide, withanolide A and withanolide B) in Sida cordifolia and Withania somnifera root respectively. GC–MS analysis identifed ffty bioactive compounds in both the drugs. The pharmacognostical, physicochemical and chromatographic assessment of these medicinally important plant drugs could be helpful for their authentication and in ensuring the quality of Balarista formulation.

        A study on wound healing activity of Bacopa monnieri Linn. aerial parts

        Bacopa monnieri is being used in the traditional system of medicine for a variety of ailments. In the present study, the ethanolic extract of Bacopa monnieri aerial parts has been studied for its wound healing activity using various models in rats. Significant increase in wound contraction and skin breaking strength were observed in the excision and incision wound models respectively. There was also significant increase in hydroxyproline content, DNA content, superoxide dismutase activity, catalase activity, glutathione level and decrease in the level of thiobarbituric acid reactive substances in the 12th day post wounding tissue of experimental rats in the ethanol extract treated groups with respect to the control group. The effect of the extract was found to be comparable with the standard drug nitrofurazone. From the results it may be concluded that the plant Bacopa monnieri is endowed with significant wound healing activity, thereby justifying its use in the traditional medicine. Saponins may be responsible for the observed wound healing activity.

        Effect of various fractions of Bacopa monnieri Linn. aerial parts on ethanolinduced hepatotoxicity in rats

        The ethyl acetate fraction (EAF) and n-butanol fraction (NBF) of ethanolic extract of Bacopa monnieri aerial parts were screened for hepatoprotective activity and in vivo antioxidant activity on ethanolinduced hepatotoxic rats. Ethyl acetate fraction was found to be more potent even though both the fractions were endowed with significant hepatoprotective activity. EAF and NBF were investigated for hepatoprotective activity in albino rats at 300 mg/kg, p.o. dose and compared with standard drug Silymarin (25 mg/kg, p.o.). Results show that both the fractions were effective in blunting ethanolinduced enhanced activities of serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, alkaline phosphatase, level of serum bilirubin (both total and direct), liver weight loss and was also effective in reducing ethanol–induced lipid peroxidation both in vitro and in vivo. Furthermore, the fractions could also enhance ethanol-induced suppressed activities of superoxide dismutase, catalase and decreased level of reduced glutathione. Results of hepatocellular damage caused by ethanol and its recovery by EAF and NBF, suggest that they might be considered as a potential source of natural hepatoprotective agents, which could be related to the free radical scavenging properties of saponins present in high concentration in the fractions.

        Hepatoprotective Activity of Ethanolic Extract of Bacopa monnieri Linn. AerialParts Against CCl4-induced Hepatotoxicity in Rats

        ethanolic extract of Bacopa monnieri Linn. aerial parts were studied for its hepatoprotective efecton CCl4-induced hepatotoxic rats. The extract was found to decrease significantly CCl4-induced elevation ofSGOT, SGPT, ALP, bilirubin and total cholesterol. But, it increased HDL-cholesterol level and liver weight withrespect to CCl4 toxic rats. The extract was also found to decrease significantly the CCl4-induced elevation of lipidperoxidation and increase the activity of antioxidant enzymes (SOD and CAT) and GSH level in the liver ofextract treated rats when compared with CCl4 induced rats. Histopathological profiles showed that the extract hadsignificant protective effect against CCl4may be possible that the mechanism of hepatoprotection of the extract is due to its antioxidant effect.KeywordsBacopa monnieri L., ethanolic extract, hepatoprotective activity, antioxidant activity, histopathology.
