In the present study, low molecular weight peptides (≤8KDa) were identified using liquid chromatography-electrospray ionization quantitative time-of-flight mass spectrometry from L. reuteri fermented brown rice samples. In L. reuteri fermented brown rice sample, various peptides were identified but six peptides were selected and synthesized based on their abundance in the LC-MS/MS analysis and literature study. The peptides namely P1, P2, P3, P4, P5 and P6 showed strong scavenging IC50 values against DPPH (5.4±0.6-14±1 μg/ml), ABTS (20±1.5-87.5±1.5 μg/ml), FRAP (28±1-87.5±0.5 μg/ml) and PSC (28.1±0.5-148±0.7 μg/ml) radicals respectively. Moreover, the cell antioxidant assay (CAA) also showed significant (81.10±0.9-309.47±2.7 μg/ml) cytoprotective effects against H₂O₂-induced cellular oxidative damage. As per our knowledge, all of the six peptides were not identified before so this study is the first report to identify and confirm their antioxidant ability against different radicals.

In this study, Limosilactobacillus reuteri generated the highest levels of GABA after fermentation. To ascertain whether germination can improve the GABA levels of brown rice, raw brown rice (Raw), germinated brown rice (Germ), fermented brown rice (Ferm) and fermented-germinated brown rice (G+F) were compared. Then, antioxidant activities were investigated for all the samples. In Ferm BR, DPPH (114.40±0.66), ABTS (130.52±0.97) and FRAP (111.16±1.83) mg Trolox equivalent 100g, dry weight (DW), were observed as the highest among all samples. Total phenolic content (97.13±0.59) and total flavonoids contents (79.62±1.33) mg GAE/100 g and catechin equivalent/100 g, DW, were also found to be highest in fermented BR. Further, phytochemical profiling using UHPLC-Q-TOF MS/MS and cell antioxidant assay revealed L. reuteri fermented BR as a strong antioxidant with the abundance of bioactive compounds, such as GABA, tryptophan, coumaric acid, L-ascorbic acid, linoleic acid, β-carotenol, eugenol, 6-gingerol, etc., as well as bioactive peptides which could contribute to the health-promoting properties of L. reuteri fermented brown rice.

In the present study, the Fructooligosaccharide was obtained by enzymatic treatment of banana peel extract and purified. In addition, the growth enhancement of probiotics strains was analyzed. Further, the targeting the microbiota-gut-brain axis, and reverse the impact of chronic Stress in Mice. Based on the in-vitro results, the banana peel crude extract and standard FOS showed similar growth enhancement was observed. Further, the crude BPE were purified using HPLC–ELSD indicating 24.9 mg/g of crude BPE showed similar structural characterization (HPLC, FTIR) with commercial FOS. Anxiety stress reduction was confirmed in Mice fed with BPE and FOS product, based on behavior pattern study (Open field, elevated position, and elevated position test), blood chemistry, and microbiota analysis using metagenome sequencing. The optimal mixing percentage of BPE and oligosaccharide cookies and muffins determined to be 5.00% and 46% had significant effects on color, taste, texture, flavor, and overall quality and sensory scores (p<0.01). In conclusion, the in-vitro and in-vivo results indicate that the extracted prebiotics reduced the stress.

However, the weakening of proteolysis is one downside of natural peptide drugs. But, peptidomimetics may help resolve this issue. In this review, peptide and peptide-based drug discovery were summarized to target one key entry mechanism of severe coronavirus pulmonary emboli syndrome (SARS-CoV-2), which encompasses the association of the host angiotensin-converting enzyme-2 (ACE2) receptor and viral spike (S) protein. Furthermore, the benefits of proteins, peptides and other possible actions that have been studied for COVID-19 through new peptide-based treatments are discussed in the review. Lastly, an overview of the peptide-based drug therapy environment is comprised of an evolutionary viewpoint, structural properties, operational thresholds, and an explanation of the therapeutic area.
