        뱀독 Arginine - Esterase 가 혈액응고 및 섬유소 용해력에 미치는 영향

        N/A Objectives: Arginine esterase in snake venom cleave peptide bonds in the fibrinogen molecule, thereby causing either coagulation or incoagulability of fibrinogen. There has been a controversy over whether arginine esterase of agkistrodon halys venom has any fibrinolytic activity in the thrombin-clotted fibrin. The aim of this study was to see whether the arginine esterase of agkistrodon halys venom itself has direct fibrinolytic activity and to compare the sentitivity of fibrin made of both thrombin and arginine esterase to the fibrinolytic activity of t-PA and urokinase. Methods: Fibrinolytic activity of arginine esterase was measured both by the fibrin plate method, and by the chromogenic method(S-2251). Coagulation activities of thrombin and arginine esterase were compared by the thrombin time(automatic coagulometer), based on the equivalent unit of each. Sensitivity of fibrin made of both thrombin and arginine esterase to the fibrinolytic activity of t-PA and urokinase was measured on fibrin film. To see whether arginine esterase degrade fibrinogen chain directly, fibrinogen was incubated with plasmin, thrombin, and arginine esterase and loaded on SDS-PAGE. Results: Fibrinolytic activity of arginine esterase was detected neither by the fibrin plate method nor using the S-2251 in NPP. Arginine esterase has no effect of the fibrinolytic activity of the euglobulin fraction of NPP, urokinase and t-PA. Arginine esterase does not degrade fibrinogen chain directly. Arginine esterase was found to be as strong a coagulant as thrombin. But the fibrin made of arginine esterase was more susceptible to both urolkinase and t-PA than that made of thrombin. Conclusion: Arginine esterase from Agkistrodod halys may be an useful anti-thrombotic drug.

        N/A Pseudomembranous colitis is a potentially severe farm of colitis that occurs almost exclusively in the presence of antibiotic exposure. Less commonly implicated are antineoplastic agents and very few other conditions. We report a case who had diarrhea due to pseudomembranous colitis after receiving chemotherapy for cancer. A 38-year-old woman with metastatic adrenocortical carcinoma was admitted with diarrhea of 12 days duration. Twenty-four days before onset of diarrhea, she had received etoposide, pepleomycin, and cisplatin by intravenous infusion. On admission, she complained of abdominal pain, diarrhea, and fever. She was dehydrated, hypotensive, and tachypneic. Her abdomen was distended with absent bowel sounds. She was not neutropenic. Diagnosis of pseudomembranous colitis was confirmed by endoscopic examination, positive biopsy specimen, and culture of C. difficile in the stool. She recovered after receiving vancomycin and metronidazole orally. Patients with severe diarrhea after receiving anticancer chemotherapy should be promptly tested for pseudomembranous colitis even in the absence of a history of antibiotic administration.

        Lumbricus Rubellus 가 인체의 섬유소 용해력에 미치는 영향

        N/A To evaluate the lumbrikinase effect on plasmin (ogen) related physiologic fibrinolytic activity, we measured euglobulin fibrinolytic activity, t-PA activity and t-PA antigen during lumbrikinase therapy and the results were compared with the data of control day of before and after lumbrikinase ingestion. Serial dilution of lumbrikinase showed the fibrinolytic activity on fibrin plate by dose dependent pattern between 375㎍/ml and 1㎍/ml concentration. In regard to the fibrinolytic area, 300㎍ of lunbrikinase equivalent to 25 unit of t-PA and 75㎍ of lumbrikinase equivalent to 5 unit of t-PA. Euglobulin fibrinolytic activity increased in all cases by two or three days after lumbrikinase ingestion but decreased to basal level 36 hours after discontinuation of lunbrikinase. The lumbrikinase added to plasma did not influence to the euglobulin fibrinolytic activity. t-PA ag was increased in 5 out of 6 cases and t-PA activity was increased all of the cases after lumbrikinase ingestion. There was direct correlation between euglobulin fibrinolytic activity and t-PA activity. This finding suggest that the increased fibrinolytic activity of euglobulin after lumbrikinase ingestion is due to increased t-PA rather than direct effect of lumbrikinase itself.

        N/A Defective fibrinolysis, which has been associated with an increased risk of thromboembolic disease, may be relevant to the vascular complications of diabetes. The euglobulin lysis area was measured in 56 NIDDM patients and 116 healthy controls before and after a venous occlusion, and the following results were obtained: 1) Compared to the control groups, there were significantly lower basal and stimulated euglobulin lysis areas in diabetics. The serum fibrinogen Are you malang a garacl stctement lee which wrtrasts with the net og the sentane, on it gyyoset to be tendel, as pant of you ieult? tends be lower in diabetes, but there was no statistical significance. 2) No relation of fibrinolytic activity to age, duration of diabetes, sugar, serum lipid, or obesity was found. 3) Both basal and stimulated fibrinolytic activity were significantly lower in diabeics who developed retinopathy than in diabetics who were fuee of retinopathy, In diabetics, serum fibrinolytic activity is lower than in healthy controls, a finding which could play a role in the development of diabetic retinopathy.
