        3T3-L1 세포에서 칡잎 에탄올 추출물의 지방합성 저해 효과

        In this study, we investigated the effects of Pueraria lobata leaf ethanol extract (PLLEE) on lipogenesis in 3T3-L1cells during their differentiation from preadipocytes to adipocytes by measuring lipid accumulation and lipogenic-relatedfactors. Treatment with PLLEE reduced the lipid content in Oil-red O staining in a dose-dependent manner. In addition,PLLEE reduced the activity of glycerol-3-phosphate dehydrogenase. Moreover, PLLEE down-regulated acetyl-CoAcarboxylase (ACC) and fatty acid synthase mRNA expression. In conclusion, PLLEE has an anti-lipogenic effect in 3T3-L1 cells.

        Hairless 마우스에서 LED 광 조사의 피부장벽 손상 및 염증 경감 효과

        This study intended to evaluate the alleviating effect of light emitting diode (LED) irradiation on the skin barrier damage and inflammation in hairless mice skin. LED (wavelength: 635 nm, intensity: 3.5 mW/cm2) was irradiated to the region of skin barrier damage and inflammation (E1 group: 30 minutes/day, E2 group: 60 minutes/day). Skin barrier damage and inflammation were artificially induced by UVB irradiation and squalene monohydroperoxide (Sq-OOH) application, 6 days a week for 4 weeks. On the 28th day of experiment, E1 and E2 groups had significantly (p/0.05) lower values of both skin erythema index and transepidermal water loss (TEWL) than the control (C) group. The appearance and degranulation of mast cells in dermal and subcutaneous layers remarkably reduced in E1 and E2 groups compared to the C group. Therefore, LED irradiation seems to be effective in skin barrier damage and inflammation induced by sunlight.

        인체피부섬유아세포에서 녹차,백차,홍차 열수추출물의 MMP-2 활성 및 MMP-1 유전자 발현 저해능

        This study was conducted to evaluate the inhibitory efficacy of water-soluble extracts from green tea (GT), white tea (WT) and black tea (BT) on MMP-2 activity and MMP-1 gene expression in human dermal fibroblasts. In the experiment of MMP-2 activity using a zymography, at the concentrations of 12.5, 25, and 50μg/mL, EGCG decreased it by 15.4, 24.8 (p?0.05), and 36.1% (p?0.01), GT decreased it by 17.1, 27.2 (p?0.01), and 38.7% (p?0.01), WT decreased it by 1.7, 17.6 (p?0.05), and 21.2% (p?0.05), and BT decreased it by 37.8 (p?0.01), 41.5 (p?0.001), and 47.5% (p?0.001), respectively, compared to the control group. The order of high to low sequence for the inhibitory efficacy on MMP-2 activity was BT¤GT¤EGCG¤WT. In the experiment of MMP-1 gene expression using a RT-PCR, at the concentrations of 12.5, 25, and 50 μg/mL, EGCG significantly (p?0.001) decreased it by 71.1, 80.6, and 86.9%, GT significantly (p?0.001) decreased it by 35.4, 77.6, and 84.6%, WT significantly (p?0.001) decreased it by 32.7, 33.1, and 58.4%, and BT significantly (p?0.001) decreased it by 49.2, 81.4, and 88.2%, respectively, compared to the control group. The order of high to low sequence for the inhibitory efficacy on MMP-1 gene expression was BT¤EGCG¤GT¤ WT. These results showed that water-soluble extracts from GT, WT and BT had a high inhibitory efficacy on the collagen degradation in human dermal fibroblasts. Especially black tea, which is fermented tea, showed the highest efficacy among the teas tested in this experiment. even better efficacy than EGCG.

        인체피부섬유아세포에서 녹차,백차,홍차 열수추출물의 콜라겐 합성능 비교 연구

        This study was conducted to evaluate the collagen synthetic abilities of water extracts of green tea (GT), white tea (WT) and black tea (BT) in human dermal fibroblasts. In MTT assay, the maximum permissible level for GT, WT and BT was over 200μg/mL, and that for EGCG was 50μg/mL. In the experiment of type-I procollagen synthetic ability, TGF-β1 (positive control) increased it by 179% (p/0.001) at the concentration of 5 ng/mL, compared to the control group. At the concentrations of 12.5, 25, and 50μg/mL, EGCG increased it by 49.3, 80.1 (p/0.01), and 141.6% (p/0.001), GT increased it by 8.3, 48.1, and 98.5% (p/0.01), WT increased it by 10.3, 38.6, 110.4% (p/0.05), and BT increased it by 89.5 (p/ 0.01), 96.9 (p/0.01), and 114.5% (p/0.01), respectively. The order of high to low sequence for the collagen synthetic ability was EGCG¤BT¤WT and GT. It was conformed that GT, WT and BT had high collagen synthetic abilities, and they could be a useful functional natural source for the anti-wrinkle cosmetic materials.

        인체피부섬유아세포에서 초석잠 에탄올 추출물의 주름형성 억제 효능

        The objective of this study was to evaluate the anti-wrinkle efficacy of Stachys riederi var. japonica ethanol extract (SREE) in human dermal fibroblasts by analyzing collagen synthesis and collagen degradation. The total polyphenolic and flavonoid contents of SREE were 41.2 and 25.4 mg/g, respectively. At concentrations of 1,000 μg/mL, the electron- donating ability of SREE was 82.0%. In vitro SREE treatment (10 mg/mL) inhibited collagenase and elastase activity by 60.6% and 39.8%, respectively. The maximum permissible level for treating SREE to human dermal fibroblasts was shown to be over 200 μg/mL. In comparison with the vehicle, SREE treatment at 200 μg/mL significantly (p<0.05) increased type I procollagen synthesis by 76.6%. Compared with the control, SREE treatment at 200 μg/mL significantly (p<0.05) decreased matrix metalloproteinase (MMP)-1 mRNA and protein expression by 20.4% and 9.7%, respectively. From these results, we confirmed that SREE had low toxicity and had high anti-wrinkle efficacy in human dermal fibroblasts. Therefore, SREE could be used as a functional cosmetic material for the prevention or alleviation of skin wrinkles induced by ultraviolet rays.

        3T3-L1 세포에서 태광콩과 청국장 에탄올 추출물의 항산화능과 지질 축적 저해 효능 비교

        In this study, we investigated the effects of Glycine max Merrill bean (Bean) and cheongkukjang powder ethanol extracts (CP) on the antioxidant activity and anti-lipid accumulation efficacy in 3T3-L1 cells during their differentiation from preadipocytes into adipocytes. Total Bean and CP polyphenolics were 5.7 and 5.9 mg/g, respectively, and flavonoids in the Bean and CP were 5.5 and 5.6 mg/g, respectively. The electron donating abilities of the Bean and CP were 19.1% and 46.1%, respectively, and their 2,2′-azino-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities were 71.1% and 85.1%, respectively. The Bean and CP superoxide dismutase-like activities were 19.3% and 20.2%, respectively. Treatment with Bean and CP reduced the cellular lipid content in a dose-dependent manner. Collectively, Bean and CP had anti-lipogenic effects in 3T3-L1 cells.

        녹차,백차?홍차 열수추출물의 항산화능 비교

        This study was conducted to evaluate the antioxidant activities of water extracts of green tea (GT), white tea (WT) and black tea (BT). The polyphenol contents of EGCG, GT, WT, and BT were 436.0, 105.5, 108.7, and 106.2 mg/g, respectively. The electron-donating abilities of EGCG, GT, WT, and BT at 100 μg/mL were 91.7, 90.2, 90.2, and 89.6%, respectively. The ABTS radical abilities of EGCG, GT, WT, and BT at 100 μg/mL were 99.5, 85.5, 92.2, and 83.7%, respectively. The SOD-like activities of EGCG, GT, WT, and BT at 1,000 μg/mL were 28.9, 26.3, 28.7, and 31.1%, respectively. The nitrite scavenging ability of EGCG, GT, WT, and BT at 3,200 μg/mL were 99.1, 99.5, 99.4, and 99.6%, respectively. It was conformed that GT, WT and BT had high antioxidant activity, and they could be a useful functional source for the cosmetic materials.

        인체피부섬유아세포에서 애기달맞이꽃 메탄올추출물의 항 주름 효능

        The objective of this study was to evaluate the anti-wrinkle efficacy of Oenothera laciniata methanol extract (OLME) in vitro using human dermal fibroblasts. Specifically, we tested the type-I procollagen synthetic ability of OLME at concentrations of 6.25, 12.5, 25, and 50 μg/mL and noted significantly significant (p<0.05) increases versus the control of 38.7, 74.3, 111.4, and 113.0%, respectively. Using RT-PCR, the expression of MMP-1 mRNA in human dermal fibroblasts treated with 6.25, 12.5, 25, and 50 μg/mL OLME was shown to have decreased when compared to the control by 67.7, 71.7, 75.6, and 84.3%, respectively (p<0.05). When measuring MMP-2 activity in human dermal fibroblasts treated with 6.25, 12.5, 25, and 50 μg/mL OLME using zymography, it was shown to have decreased versus the control by 27.6, 37.1, 76.3, and 93.9%, respectively (p<0.05). OLME was less cytotoxic to human dermal fibroblasts than ascorbic acid. From these results, we found that OLME is highly efficacious in its ability to promote collagen synthesis and inhibit its degradation in human dermal fibroblasts, similar or even superior to those of ascorbic acid, a well-known anti-skin aging agent. Therefore, OLME could be used as a functional cosmetic material for the prevention or alleviation of skin wrinkles induced by ultraviolet rays.

        돌단풍 잎 에탄올 추출물의 피부미백 효능

        We evaluated the skin whitening effects of Aceriphyllum rossii leaf ethanol extract (ARLEE) in vitro using melan-a cells. The maximum permissible level for ARLEE application to melan-a cells was 50 μg/mL. In gross observation, ARLEE reduced melanin deposition and melanocyte dendricity in a dose-dependent manner. When compared to treatment with vehicle, ARLEE treatment (50 μg/mL) significantly decreased melanin contents by 33.0% (p<0.001), an even greater decrease than arbutin (positive control) treatment (27.2%). ARLEE treatment (50 μg/mL) significantly decreased intracellular tyrosinase activity by 48.8% (p<0.001), an even greater decrease than seen after arbutin treatment (36.0%). ARLEE treatment (50 μg/mL) significantly decreased tyrosinase protein and mRNA expression by 30.5% and 37.7% (p<0.01), respectively. These results suggest that ARLEE has better skin whitening activity than arbutin in melan-a cells, and this activity is likely mediated at the level of tyrosinase transcription. These results suggest that ARLEE could be used in functional cosmetic materials to prevent or alleviate skin pigmentation.
