        흰쥐 분만의 인위적 조절에 관한 연구 3 . Indomethacin 투여에 의한 분만지연 시험

        This experiment was carried out to study the efficacy of the exogenous indomethacin for prolongation of parturition in white rats. Sixty of the pregnant white rats used were injected with indomethacin. The survival rate of newborn, the administration time and the amount of dosage affecting on the prolongation parturition were investigated. The results obtained were as follows: 1. The average survival rates of newborn in groups injected with indomethacin on day 15, 17 and 19 of gestation were 78.2, 82.0 and 85.5%, respectively. There were no differences by the amount of dosage of indomethacin. 2. Mean gestation lengths of rats injected indomethacin on day 15, 17 and 19 of gestation were 23.39±0.18, 23.28±0.22 and 23.19±0.09 days, respectively. There were no significant differences (P$lt;0.05) among experimental groups. 3. For the amounts of dosage of 0.2㎎ × 1time, 0.4㎎ × 1time and 0.6㎎ × 1time per ㎏ per day the average intervals from injection to parturition were 22.98±0.05, 23.48±0.15 and 23.40±0.04 days, respectively. There were no significant differences (P$lt;0.05) among experimental groups.

        흰쥐 분만의 인위적 조절에 관한 연구 4 . Aspirin 투여에 의한 분만지연 시험

        This experiment was carried out to study efficacy of the exogenous Aspirin for prolongation of parturition in white rats. Sixty of the pregnant White rats used were injected with Aspirin. The average survival rate of newborns in groups injected with Aspirin on day 15, 17 and 19 of gestation were 77.1%, 76.0% and 79.7% respectively. There were no differences by the amount of dosage of Aspirin. 2. Mean gestation length of rats injected Aspirin on day 15, 17 and 19 of gestation were 23.08±.22, 22.65±0.12 and 23.18±0.30 days, respectively. There were no significant differences (P$lt;0.05) among experimental groups. 3. For the amount of dosage of 50㎎/㎏ × ltime, 100㎎/㎏ × Mime and 150㎎/㎏ × Mime per ㎏ per day the average intervals from injection to parturition were 22.67±0.17, 22.97±0.25 and 23.28±0.22 days, respectively. There were no significant differences (P$lt;0.05) among experimental groups.

        생쥐 초기배의 분리할구와 분할배의 체외배양

        This experiment was carried out to investigate the developmental potential of bisected morulae and blastocysts, and the isolated blastomeres from 4-and 8-cell embryos in mouse. The results obtained were summarized as follows: l. The rates of blastomeres survived manipulation were 83.4 and 76.7%, in 4-and 8-cell embryos, respectively. 2. The rates of half embryos survived bisection were 77.7 and 75.3%. in morulae and blastocysts, respectively. 3. The rates of development of isolated blastorneres from 4-and 8-cell embryos were 24.7 and 10,4% respectively. 4. The rates of development of the bisected morulae and blastocysts were 67.7 and 71.9% respectively.

        유우정액보존에 대한 난구액과 SEMINAN 액 사용에 관한 연구

        This experiment was conducted to determine the effects of the various levels of egg yolk in egg yolk-citrate extenders and Seminan, S.P. extender on the motility of daily bull sperm ejaculated from two Holstein bulls and one Korean bin! from August 1960 to August 30, 1960. Four egg yolk-citrate extenders used were different in the egg yolk contents added such as 20%, 35%, 50%, 75%, and the Seminan, S.P. extender was a commercial boil semen extender made in Japan. The results obtained were as follows; 1. Observing the sperm motility immediately after collection, there was no significant difference in the motility of sperm from 3 bulls 2. Until the 3 rd day after dilution, there was no significant difference in the sperm motility between 5 diluted semens. 3. It was found that the motility of sperm in semen diluttd with egg yolk 75% solution was lower than 70% in average in the the 4th day and became to be somewhat coherent. Thus the semen diluted with egg yolk 75% could not be available after 3 days. And rating the sperm motility microscopically, too much fat gloubles from egg yolk used made the identification of sperm difficult. 4. Until the 5th day after diluting the semen, the sperm motility of all semen except fur the egg yolk 75% semen was higher than 70% and there was no difference in sperm motility of these 4semens statistically. 5. In the 6th day after diluting the semen, the sperm motility of the egg yolk 50% semen was also lower than 70% and could not be used for insemination. So the semen diluted with egg yolk 50% might be possible to be preserved for 5 days. 6. The sperm motility of only the semen diluted with Seminan, S.P. extender was brought to be higher than 70% until the 7th day after diluting. Both the egg yolk 20% semen and the egg yolk 35%-semen could be available until only the 6th day after diluting the semen. From the above results it was concluded that the best results on the preservation of daily bull semen would be brought from the 30% level of egg yolk added in the egg yolk-citrate extender.

        Silver Nitrate 용액의 정소내 주입에 의한 가토의 거세효과에 관한 연구

        This experiment was carried out to develop a chemical castration with a single intratesticular injection of silver nitrate solution in domestic rabbits. A total of 44 bucks of 4 to 5 months old were alloted at random into the following 4 groups: 9 bucks for controls, 6 bucks for 0.3㎖ injection of 1% AgNO₃, 14 bucks for 0.5㎖ injection of 1% AgNO₃ and 15 bucks for 0.5㎖ injection of 5% AgNO₃. Changes in body weight, testicular width in scrotum. weight and histology of testis at necropsy, blood cell counts and plasma levels of testosterone of the buck were measured for 4 weeks after treatment. The results obtained were summarized as follows: Compared with the controls, the body weight gain of bucks treated with AgNO₃ was not significantly (P$lt;0.05) different during the experimental period for 4 weeks. The white blood cell counts were tended to be increased slightly but insignificantly 1 day after injection of AgNO₃ and then decreased to normal level soon. There was no significant change in red blood cell counts in the bucks treated with AgNO₃. Most of the testis of bucks were swollen significantly (P$lt;0.05) by 6 to 12㎜ in the width of testis in scrotum 1 day after injection of AgNO₃, but began to decrease in sue from 3 days after treatment and to their normal size in a week. The AgNO₃ injection was not sufficient to castrate some bucks successfully. Only 60.0 to 66.7% of the bucks treated with 0.0㎖ of 1 or 5% AgNO₃ solution showed the plasma levels of testosterone of !sower than 0.1 ng/㎖ by 28 days after treatment. This result was considered to be due to the fact that not a whole but a limited part of testis was injured by the present method of injection. The weight of testis of the bucks of which plasma testosterone were less than 0.1 ng/㎖ by 7 to 28 days after treatment was not significantly(P$lt;0.05) different between the treatments or periods after treatment. This result indicates that the period of 4 weeks following treatment might be insufficient for the injured testicular tissue to be necrotized and absorbed fully. The extent of damage of testis by injection of AgNO₃ was much different by the local effect in the parts of distance from injection. The proximal portion of testis injected with AgNO₃ was fully damaged and necrotized, but a distal part of testis was not severely damaged.

        Rat 의 임신기 및 분만직후의 혈액상에 관한 연구

        This experiment was conducted to investigate the change of blood pictures and total serum protein in clinically health rats (body weight: 200-250) during 7, 15, 20 days of gestation and after parturition. The results obtained were as follows : 1. Number of erythrocytes, value of hemoglobin and hematocrit during gestation and after parturition significantly (P$lt;0.05) decreased to after parturition. 2. The mean of corpuscular hemoglobin content, corpuscular hemoglobin concentration and corpuscular volume were not significantly (P$lt;0.05) different between gestation and after parturition. 3. The total leukocyte count was not significantly (P$lt;0.05) different during gestation, but it was significantly (P$lt;0.05) increased to after parturition. 4. Total serum protein was significantly (P$lt;0.05) decreased as the gestation progressed, but it wars slightly increased after parturition.

        완만동결법에 의한 생쥐의 정상배 및 분할배의 생존성에 관한 연구

        The effects of cryoprotectant, equilibration time and cell stage on the survival of intact and bisected mouse embryos were determined following slow freezing. The number of blastomeres during in vitro development was counted by nuclei staining with Hoechst 33342. The post-thaw survival rates of intact 8-cell embryos, morula and blastocysts after slow freezing were 62.8, 76.8 and 60.0% in 1.5 M glycerol, and were 64.4, 75.6 and 57.6% in 1.5 M DMSO, respectively. The survival rates of ZP-free or bisected morula after slow freezing were 60.5 and 48.6% in 1.5 M glycerol, respectively. The number of blastomeres following in vitro culture for 24 to 28 hours of fresh and frozen morula was significantly (p$lt;0.05) different as 53.6±1.3 and 40.0±1.0, respectively. The mean number of blastomeres following in vitro culture for 24 to 28 hours of bisected and bisected frozen morula was 25.0±1.0 and 21.8±0.8, respectively. When 531 fresh, 282 frozen, 130 bisected and 66 bisected frozen embryos were transferred to 46, 23, 15 and 12 recipient mice, 26.9, 21.7, 14.6 and 7.6% of them developed to normal offspring, respectively.
