        핵산분석법에 의한 한우의 판별

        Genetic differentiation between Korean Native Cattle (Hanwoo) and European cattle breeds was compared by the analysis of DNA polymorphisms. Genomic DNA of Hanwoo or Holstein breeds was digested using 7 restriction endonucleases. BamHI, BglI. EcoRI, HinfIII, MspI PstI, and TagI. All the enzymes produced clear stained bands. The cleavage patterns of all the enzymes were identical between Hanwoo and Holstein breeds. DNA polymotphisms among cattle breeds were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerise chain reaction. Less than 10 polymorphic bands were detected. The RAPD patterns were identical among the Holstein, Hereford and Angus breeds, but the above pattern was different from that of Hanwoo. All bands detected in the Hanwoo breed were observed in European cattle breeds, but one of the common bands found in European cattle breeds was not detected in the Hanwoo breed. Tbis band was independent of sex and individuals, and may be useful as a DNA marker for identifying the Hanwoo breed.
