Background: Safflower (Carthamus tinctorius L.) is a useful medicinal and oil crop in Korea. However, when safflower is cultivated, the flowering period overlaps with the rainy season, and seed maturation is poor. Therefore, this study aimed to use basic research data to develop superior varieties using agronomic characteristics and crossing method. Methods and Results: A total of 34 safflower germplasms were sown and their agronomic characteristics were investigated. Based on these investigations, the cultivar ‘ui-san-hong-hwa’ was selected as the mother plant, and ‘Myanmar safflower’ (Hsu Pan) was selected as the father plant. In addition, we developed a floret-protecting cap to cover florets after emasculation during the artificial crossing. When florets were protected by the cap, the seed setting rate increased in comparison to that in the non-covered florets. Conclusions: Agronomic characteristics can contribute to developing suitable varieties. The results suggest that the protection cap will be helpful in breeding without the floral organ drying. This study contributes an efficient breeding method to develop new safflower varieties.

Background: This study was conducted to determine the effect of temperature, light, hormones, and after-ripening treatment on Rehmannia glutinosa seed germination to provide valuable information for its breeding and cultivation. Methods and Results: The seeds used in this study were harvested from the ‘Jihwang-1-ho’ cultivar. The seed length and width were 1.13 ± 0.03 ㎜ and 0.86 ± 0.02 ㎜, respectively, with a seed coat characterized by polygonal cells. The germination efficiency was evaluated at various temperatures (15, 20, 25, and 30℃) and light conditions (light/dark). The optimal germination condition was found to be 25℃ with light (91% germination). Pre-soaking the seeds with gibberellic acid (GA₃) increased germination, and the highest germination (72%) was observed with 1,000 ㎎/ℓ GA₃. Additionally, this also increased the mean germination time , germination uniformity, and germination index. The seeds that matured at 45% and 70% relative humidity showed the highest germination rates at 32 and 24 weeks, respectively. Conclusions: The best method for R. glutinosa seed germination is maintaining 25℃ under light conditions with after-ripening treatment for over 16 weeks. Pre-soaking the seeds in 1,000 ㎎/ℓ GA₃ for 24 h further improved their germination. Rehmannia glutinosa seeds were classified under non-deep physiological dormancy.

Background: Licorice is an important medicinal plant, but climate conditions can hinder seed production. Because it is difficult to produce the seeds, Korea mostly depends on the imported product. In addition, when grown domestically, the standard ingredients do not comply with those of the Korean Pharmacopoeia (glycyrrhizin 2.5% and liquritigenin 0.7%). Therefore, it is necessary to develop licorice variety suitable for the domestic environment. Methods and Results: A new interspecific hybrid cultivar designated ‘Dagam’, was developed from Glycyrrhiza glabra × G. uralensis (G. korshinskyi Grig.) and is regarded as a high-yielding and high-quality licorice by Rural Development Administration. On average the yield for ‘Dagam’ was recorded 431.5 ㎏/10a. The cultivar displayed strong brown spot disease and lodging resistance. Additionally, the glycyrrhizin and liquiritigenin contents of ‘Dagam’ (3.0 and 0.7%, respectively) met, the Korean Pharmacopoeia standard. This cultivar can widely adapt to most cultivation areas in Korea except Jejudo, Gangwondo, and the mountain-alpine regions. Furthermore, ‘Dagam’ was not showed any cytotoxicity and high cell viability compared to ‘G. uralensis’. Conclusions: The interspecific hybrid cultivar ‘Dagam’ will contribute to the stability of domestic cultivation of licorice in Korea.

Background: When the licorice cultivated in Korea, the quality index components are lower than the standard value, and physiological disorders occur frequently. Therefore, we evaluated the growth and yield characteristics of resources to develop licorice varieties suitable for the domestic environment. Methods and Results: Glycyrrhiza resources were collected from five nations. Leaf margin has been identified as a species-specific characteristic that distinguishes G. uralensis and G. glabra. Glycyrrhiza resources were observed in flowers and seeds for 2 years. Leaf abscission began to fall from July and lasted until November according to the characteristics of the resources. As the number of cultivation years increased, root length, rhizome length, and rhizome number were not significantly related, however, root diameter, root weight and rhizome weight were significantly related. The skin root color became dark after a year. Yield (dry weight of underground part) increased mostly from the first to second year, whereas the yield changed depending on resource characteristics in three years. The yield was positively correlated with dry rhizome weight, plant height, stem diameter, and root diameter. Cluster analysis indicated that, they were classified into three clusters. Among them, the resources in group Ⅲ were confirmed to be of high quantity and quality. Conclusions: GLY13-05, GLY14-02, and GLY14-06 of Glycyrrhiza resources were evaluated as having excellent growth and yield characteristics. Moreover, they can be cultivated in domestic environments.
