인간의 치은조직내 CuZn-SOD 와 Mn-SOD 에 관한 연구

The purpose of this study was to ascertain the presence of the scavenging enzyme, superoxide dismutase(SOD) in the human gingival tissue, in addition to the activity of SOㅇ according to age. For this study, a total of 29 patients ranged in age from 20 to 62 years(6 thirties, 7 forties, 12 fifties, and 4 sixties). Gingival tissues were surgically obtained during the periodontal surgery and extraction. The activity of CuZn-SOD and Mn-SOD in the healthy gingival tissues was measured by using UV-spectrophotometer by the same as Crapo et al did. To determine the degree of severity of ginaival inflammation, gingival tissues were stained by hematxylin-eosin stain, and author used leukocyte common antigen for white blood cells, CD3 for T lymphocytes, CD20 for B lymphocytes, CuZn-SOD and Mn-SOD as primary antibody for immunohistolocalization. The results were as follows: 1. There was a slight decreased tendency in Mn-SOD activity only with age. 2. The infiltrated inflammatory cells in excised gingival tissues were primarily CD3-positive T lymphocytes. 3. Immunohistochemistry showed 5∼50% of CuZn-SOD was positive in periphery of matrix collagen fibers in inflamed gingival tissues. 4. Immunohistochemistry showed Mn-SOD was positive less than 5% or negative in periphery of matrix collagen fibers in inflamed gingival tissues. The above results, within the limited experiment, demonstrated that decreased activity of Mn-SOD was related to agin, and the activity of CuZn-SOD increased according to the severity of gingival inflammation, in CD3+ T lymphocyte lesion.
