N/A Backgroud : Stimulation of iNOS with subsequent release of NO may play a critical role in pathophysiology or host defense reaction. NO can be produced by several cell types that reside in the skin. Increasing data indicate that NO serves as an autocrine and paracrine mediator in diverse and complex cellular processes throughout the skin. Objective : The aim of the present study was to elucidate whether dendritic epidermal T cell can express iNOS and generate NO. In addition to that, this study was to evaluate whether NO may play a certain role in IL-3 production of activated dendritic epidmal T cell. Methods : iNOS was isolated by RT-PCR at 12, 24, 48, 72 hours after stimulation and NO was measured by the Griess method. IL-3 levels were assessed with a basic cell proliferation bioassay system using supernatants from T cells cultured in 96 well plates. Units of IL-3 in the culture supernatants were measured using the cytokine-dipedent DA-1 cell-line. Results : 1. NO generation was increased by UVB l00mJ/㎠ irradiation, concanavalin-A and lipopolysaccharide stimulation in dendritic epedermal T cell. The amount of NO highest in LPS. 2. The expression of iNOS was increased by UVB 100mJ/㎠ irradiation, concanavalin-A and lipopolysaccharide stimulation in dendritic epidermal T cell. Induction of iNOS revealed peak at 72 hours after concanavalin-A and lipopolysaccharide stimulation, but in UVB irradiation that of iNOS was not observed except at 48 hours. 3. NO may play a role in IL-3 production in activated dindritic epidermal T cell. Conclusion : The results strongly suggest that the expression of iNOS and NO production in dendritic epidermal T cell are upregulated by UVB, concanavalin-A and lipopolysaccharide, and NO may play a role as mediator in IL-3 production of activated of activated dendritic epidermal T Cell as autocrine manner.

Background:Dendritic epidermal T cells(DETC) are skin specific members of the epithelial γδ-T cell family that reside normally in mouse epidermis. Although the mechanisms of antigen recognition by αβ-T cells have become better defined, the physiological role of the γδ-T cells and ligands they recognize are still poorly understood. Object:In the present study we sought to elucidate the antigen specificity and whether the DETC recognize transformed keratinocyte derived antigen. Method:We used a DETC and TCR(-) DETC which were obtained by 1600R γ-irradiation and sorting, in addition to transformed keratinocyte cell lines. We performed coculture experiments of DETC/transformed keratinocyte cell lines to detect direct evidence that DETC recognize the keratinocyte-derived antigen. Result:1. TCR negative variants of DETC do not respond to concanavalin(Con)-A, but respond to phobol myristate acetate(PMA)/ionomycin. 2. PAM 212, UV-irradiated PAM 212 and heat shocked PAM 212 cells stimulate DETC. PAM 212 cells could perform as stimulator of DETC even in the absence of stress signal. 3. UV irradiated XB2 cells stimulate DETC, but XB2 and heat shocked XB2 cells could not stimulate DETC. 4. DETC do not respond to fibroblast, UV-irradiated fibroblast and heat shocked fibroblast 5. TCR negative variants of DETC are no longer stimulated by PAM 212 cells, suggests that PAM 212 cells mediate their effects through the TCR Conclusion:The above results strongly suggest that DETC recognize specific, transformed and stressed keratinocyte-derived antigens and may play a role as an immune surveillant for cellular damage. Therfore, DETC may play critical roles during the induction of immune reaction in the skin. (Korean J Dermatol 2000;38(11):1444~1452)

Relapsing polychondritis is a rare systemic disorder manifested by recurring episodes of inflammation in cartilagenous tissues including the eye, the audiovascular systems, We describe a 53-year-old oriental woman with a one month history of painful swelling of both accompanied by intermittent hoarseness, dyspnea, impaired visual acuity, and swelling of hands. Both ear were typically uninvolved. Auricle including cartilaginous tissues were histologically examined, which revea1ed loss of basophilic staining, degenerative changes, inflammatory cells infiltrations with eosinophils and neutrophils, and perichon4rial inflammation of the cartilage. Oral prednisolone and colchicine were prescribed and the succid resolulion of both ears swelling were observed. As the disease progressed optic neuritis and chewing difficu1ty by non-erosive motion limit of temporo-mandibular joints developed, (Korean J Dermatol 2002;40(9) : 1111~1115)
