        먹장어(Eptatretus burgeri)의 혈액성상과 산소소비에 미치는 수온 및 염분의 영향

        Hagfish Eptatretus burgeri is classified as a agnathans and has many different physiological properties compared with vertebrates. In this study, we examined effects of water temperature and salinity on blood properties and oxygen consumption in hagfish. In the experiment of water temperature change, hematocrit (Ht), red blood cell (RBC) and glucose of hagfish blood revealed the lowest values at 15℃. Oxygen consumption of hagfish had significantly increased with rising water temperature, and the increasing rate was twice as much when the temperature was manipulated every 5℃. Also, oxygen consumption during the night time (a short photoperiod) was significantly higher than that of the daytime. Q10 level was 3.50 in the light period and 3.92 in the dark period. No significant change in plasma glucose level was showned in changing salinity from 30 psu to 22 psu, while it had rapidly increased at 20 psu (13.7±4.0 mg/dL) and thereafter all hagfish were dead at 18 psu. However, osmolarity, Na+, K+ and Cl- levels had significantly decreased when salinity decreased. This results are expected to develop the artificial rearing techniques of natural hagfish.

        고수온에서 조피볼락(Sebastes schlegeli)의 산소소비 및 혈액성상 변화

        Aquaculture production of Korean rockfish Sebastes schlegeli has been continuously increased from 2000s and the fish has become the second most important mariculture fish in Korea. However, there are some environmental problems in aquaculture of Korean rockfish recently. In this regards, stress responses to high water temperature was examined via oxygen consumption, blood physiological parameter and endocrinological method. Oxygen consumption of Korean rockfish had significantly increased with rising water temperature. And oxygen consumption during the light time was no different with the dark time. The levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) showed no difference until 27℃, but it had rapidly increased at 30℃. Moreover high water temperature affected to increase in plasma glucose and cortisol levels.

        담수순화 감성돔 Acanthopagrus schlegelii 동결보존 정자의 수정능력

        Fertilization rates were over 90% when sperm from black porgy Acanthopagrus schlegelii acclimated and raised in freshwater and black porgy raised in seawater were fertilized with eggs from female black porgy raised in seawater, although cryopreserved sperm were significantly lower in fertilization rates than non-cryopreserved sperm. From the observations of embryos development at different salinities (0, 10, 20 and 32 psu) within 3 hours upon fertilization (16 cell stage), all were dead at 0 psu. However, the development process and speed at 10 and 20 psu were the same as at 32 psu. But, many developing embryos were egg envelope injury at low-salinities of 10 and 20 psu unlike at 32 psu. This is attributable to osmotic shock. Hatching rate of embryos at 32 psu was 60% or so, whereas low as 5% or less were successfully hatched at 10 and 20 psu, implying that osmotic shock could be responsible for the failure of embryo development.

        국내 개체굴 양식산업 발전 방향

        Pacific oyster, Crassostrea gigas is very important species that account for more than 70% of shellfish aquaculture production in Korea, but it has variety problems from the prolonged aquaculture industry. We are paying attention to the production of individual oyster to solve these problems, and we will discuss the development direction of individual oyster aquaculture industry in Korea. Individual oyster is a high-value product that is designed to grow individual objects and sold separately without removing the shell. The key success factor in individual oyster aquaculture industry in Korea is: 1) securing good seed, 2) efficient middle rearing, 3) periodic screening and removal of attached organisms. Although individual efforts and cooperation are important industrializing the aquaculture species, mutual cooperation and efforts among the combined communities, such as aquafarm, commodity suppliers and research institutes, are the success factors. Aquafarm shall secure reproducibility through cooperation with research institute. Research institute and policy organizations should play a big role in the development of individual oyster aquaculture industries. Research institute should have a research system that can benefit aqua-farmer by performing basic research based on the site. Policy organizations need to establish a long-term system for the ongoing aquaculture industry.

        실내 사육한 줄가자미(Clidoderma asperrimum)의 성 성숙과 생식주기

        The gonadosomatic index (GSI), sex steroid hormones and gonadal development of roughscale sole Clidoderma asperrimum cultured in indoor tank were investigated to evaluate its sexual maturation and reproductive cycle. The highest GSI values of female and male were 6.91±4.03 (May) and 0.16±0.08 (August), respectively. The reproductive cycle would be classified into four successive developmental stages: growing stage (December to February), maturation stage (March to April), ripe and spawning stage (May to June), recovery and resting stage (July to November). The highest plasma testosterone (T) and estradiol-17β (E2) levels of female were 259.4±76.8 and 633.3±182.5 pg/mL, respectively in May. Also 17α, 20β-dihydroxy-4-pregen-3-one (17α, 20β-OHP) levels of female peaked in April before spawning season (244.2±42.5 pg/mL). The highest plasma testosterone (T) and 11-ketotestosterone levels of male were 231.0±46.0 and 273.9±54.5 pg/mL, respectively in April. But there was no significant difference in 17α, 20β-OHP.

        암모니아 노출에 따른 조피볼락(Sebastes schlegeli)의 생리학적 반응

        본 연구에서는 암모니아 노출에 따른 조피볼락의 생리학적 반응을 조사하고자 아가미 Na+/K+-ATPase (NKA)활성을 비롯한 혈장 parameters를 분석하였다. 실험구의 암모니아 농도는 대조구(자연해수), 1, 2, 4, 8 mg L-1였으며, 조피볼락을 각각의 실험구에 3시간동안 노출한 다음혈액 및 아가미 조직을 샘플링하였다. 실험구의 암모니아 농도가 높아질수록 혈장 암모니아 농도가 증가하였으며, 아가미 NKA 활성 또한 증가하는 경향을 보였다. 혈장 Cl-를 제외한 외부의 암모니아 영향을 받지 않았으나, 4, 8 mg L-1구의 Na+, K+ 및 삼투질농도는 대조구및 1, 2 mg L-1구보다 유의하게 높았다. 암모니아 노출에 따른 조피볼락의 혈장 코티졸은 암모니아 농도와 선형관계를 보였으며, 혈장 글루코스 또한 코티졸과 동반상승하는 것으로 나타났다. 1, 2 mg L-1구의 hematocrit는 대조구와 차이를 보이지 않았으나, 4, 8 mg L-1구는 나머지 실험구보다 유의하게 높았다. 암모니아 농도가 높을수록 아가미 조직 손상은 심하였으며, 특히 4, 8 mg L-1구에서는 상피세포의 과증식, 분리, 괴사 및 2차새변의 곤봉화(club-shaped lamella) 현상이 관찰되었다. The aim of the present study was to assess the effects of ammonia on physiological responses in Korean rockfish (Sebastes schlegeli). Interestingly, no mortality were observed when the specimens (301.1±8.0 g) were exposed to five levels of un-ionized ammonia (NH3) (control, 1, 2, 4,8 mg L-1) for 3 hours. Furthermore, a significantly higher increase in gill Na+/K+-ATPase (NKA) pump activity with was detected due to the ammonia exposure. The activity of the fishes were found to be 4 and 8 mg L-1 NH3, which was significantly high compared to normal 1 mg L-1 NH3. Although ammonia exposure had no effect on plasma Cl-, exposure to both 4 and 8 mg L-1 NH3, however it led to increase in the plasma Na+, K+ and osmolality levels. Also, prolong ammonia exposure cause increase of plasma cortisol and glucose levels. The increase in glucose was accompanied by an increase in cortisol. The fish exposed to 4 and 8 mg L-1 NH3 showed significantly higher hematocrit than control group than those exposed to 1 and 2 mg L-1 NH3. The intensity of cell damage increased with the increase concentration and exposure to ammonia. Furthermore, hyperplasia, separation and epithelial necrosis were also observed in gill tissues. Taken together, the results showed that direction of changes to the investigated parameters can be used to determine the physiological responses of Korean rockfish to ammonia.
