      • Sodium Fluoride(NaF)가 外因性 Glucose에 依해 惹起되는 心房收縮力의 增加에 미치는 影響

        To study the effects of NaF on the positive inotropic action to be induced by the addition of the glucose, the changes of the contractility of the rat atria were observed in the medium of Kreb's ringer bicarbonate (KRB) buffer solution at pH 6.5. The results obtained were as follows: 1. In the pH 6.5, the contractility of the rat atria was increased by the addition of NaF and the increment of the contractility was uprised to the maximum level after 30 minutes and its increment was decreased gradually. Otherwise, in the KRB buffer solution to be added the glucose, the increment of the contractility was durated after 60 minutes. 2. When the contractility to be increased by NaF was gradually decreased in the pH 6.5 of KRB buffer solution, the contractility was recovered by the addition of the glucose. 3. When the pH 7.4 of the glucose-added KRB buffer solution was exchanged to the pH 6.5 of the glucose-free KRB solution, the decreased contractility was recovered very poorly by the addition of glucose, but the contractility was recovered more remarkably by the addition of NaF.

        Fast vital capacity is the volume of gas can be exhaled by maximal voluntary effort following fullest inspiration, expiration being as rapid as complete as possible ("forced"). Forced expiratory volume(FEV, qualified by time internal T: FEV_T or timed vital capacity) is a measurement of a fast vital capacity related to be the time required for the maneuver. Therefore, it is important to have a formula with one compute the normal value for the subject and compare with the measured value. Hence, present study was attempt to derive the formulas for prediction of forced expiratiry volume(FEV_T, ml) of the normal Korean children and adolescents. Forced expiratory volume was measured in healthy Korean children and adolescents (556 male and 390 female) whose age range from 8 to 18 years. A 9 litter spirometer (Collins Inc.) was used for the measurement of FEV. The subject was trained on how to perform a fast vital capacity. Then, with nose clip in place, subject breathed quietly through spirometer for a few breaths, then takes as deep a breath as possible and then exhales all the gas within the lung as rapid as forcefully as possible (drum speed was 32 mm/sec and standing position). Towards the end of the expiration the subject was exhausted to continue the effort until no further gas was expired. During the measurement, investigator stood by the subject to give a constant encouragement, and measurement was repeat two or three times and the best one was subjected to analysis. Two values of FEV were calculated : the volume exhaled during the one-half and the first second(FEV_T, ml) and the percentage of the total vital capacity exhaled during the one-half, the first and the first two second(FEV_T, %). In addition to FEV, maximum mid-expiratory flow(MMF) was obtained from the forced expiratory spirogram and all the measured values were subsequently converted to values at BTPS. The results are summarized as follows: 1. The total vital capacity was increased linearly with ages until 17 in both sexes. The values of male and female at the age of 17 were 4369±470 and 3220±510ml, respectively. 2. The forced expiratory volume(FEV_T, ml) was increased linearly with ages until 17 in both sexes. The FEV_0.5 were 2643±367ml in male and 2089±308ml in female, and the FEV_1.0 were 3815±425ml in male and 2897±373 ml in female at the age of 17. 3. The FEV_0.5 of male and female were 65.6±8.1 and 64.4±10.1% in the age of 8, and this values were not related to the age in both sexes, FEV_1.0 was about 90% in all ages and both sexes. 4. Correlation coefficients between MMF and FEV_1.0 or FEV_0.5(ml) were 0.75 and 0.74 in male, and 0.83 and 0.84 in female subjects. 5. The prediction formula for forced expiratory volume(FEV_T, ml, BTPS) was derived from above results were as follows. For male FEV_0.5={3.55+(0.71×age)}×Ht(cm) FEV_1.0={2.79+(1.13×age)}×Ht(cm) For female FEV_0.5={3.98+(0.53×age)}×Ht(cm) FEV_1.0={5.59+(0.75×age)}×Ht(cm)

      • Vincristine이 家兎 赤血球膜의 Na-K-ATPase 活性度에 미치는 效果

        This study was to investigate the effect of the anticancer drug, vincristine, that is approved for use widely in acute leukemia in children, in Hodgkin's disease, and in 1vmphocytic lymphosarcoma, etc., on the activity of Na-K-ATPase, which is most abundantly present in tissu to have a high capacity for sodium and potassium transport, and is activated by their physiological concentration. And this study had observed the difference of effects between the presence of ginseng water extract or not in the experimental process. The results were summarized as follow 1. Vincristine was significantly inhibited the Na-K-ATPase activity at the dose of 0.5mg per ml. 2. The ginseng water extract was significantly decreased the degree of the inhibited activity of Na-K-ATPase at does of 1. 5%. 3. Vincristine had no effect on the Mg-ATPase activity in this experimental doses.

      • 人蔘이 抗癌劑의 副作用에 미치는 影響

        5-fluorouracil and mitomycin C are used extensively for cancer therapy in the clinical part. But they have been known to possess various the side effects, such as depression of the bone marrow, and of the kindney, etc. These side effects became the limited factors for the cancer therapy. The object of this study was to investigate the route that is reduced the side effects. And, then, 5--fluorouracil and mitomycin C selected were administered with or/without the water extract of Panax ginseng through the ear vein of the rabbits. The period of injection was for a week at first, and then every other week for 35 days. And it was attempted to compare the the effects to inject with or/without the ginseng extract. The results were summarized as follow: 1. The number of r. b. c. was not affected by the administration of anticancer agents with or/without the ginseng extract. 2. The number of w. b. c, was decreased significantly by the administration of anticancer agents with or/without the ginseng extract. But the degree of decrement in the group to be injected with the ginseng extract was more inhibited significantly than that of the group to be injected the agent alone. 3. Urine flow, renal plasma flow, glomerular filtration rate and urinary excretion of sodium were decreased significantly by the administraction of the agents with or/without the ginseng extract, respectively. But the degree of decrement in the group to be injected with the ginseng extract was more inhibited significantly than that of the group to be injected the agent alone. From the above result, it was suggested that the water extract of Panax ginseng may be reduced the side effects of anticancer agents, 5-fluorouracil and mitomycin C.

      • Vinblastine Sulfate가 家兎의 腎機能에 미치는 影響

        This study was to investigate the effect of vinblastine sulfate(VLB) on the renal function of the rabbits. VLB was administered intravenously in a single dose of 0.2 mg/kg and 0.3mg/kg, and in chronic doses. The results were summarized as follows: 1. Urine volume, renal plasma flow and glomerular filtration rate were decreased, respectively, and its decremental ratio was proportion to the dose administered. 2. The absorptive ratio of sodium was markedly increased in comparison to that of potassium. 3. The osmolar clearance and negative free water clearance were decreased, respectively. From the above results, it was suggested that the effect of VLB on the renal function of the rabbits may by caused the changes of renal hemodynamics, the secretion of aldosterone and antidiuretic hormone.

      • 瓦松이 신수질의 Na-K-ATPase 活性度에 미치는 影響

        Sodium-potassium activated adenosine triphosphatase (Na-K-ATPase), which is contained in the cell membrane, have the important roles in the material transport through the cell membrane, especially sodium and potassium. And its activity is inhibited by a kind of cardiac glycoside, ouabain. Orostachys malacophyllus Fischer (Tsume renge) is a plant to grow on the tiled roof of the house, and in the field or mountain. And the compositions of the plant have not been known yet. But it has transmitted to this time as a kind of medical plant from remote antiquity. Sometimes, it is used in the folk remedies for the treatment of the malignant diseases. But it is not clear in the therapeutic effect. This study was attempted to investigate the effect of the water extract of Tsume renge on the Na-K-ATPase activity, which was purified from the outer medulla of pig kidneys. And the cell membrane fraction was incubated with sodium dodecyl sulfate and was purified by the method of Jørgensen. The results obtained were summarized as follows: 1. Na-K-ATPase activity of the control group was 3.6±0.3μM/g protein/10min. 2. Total ATPase activity was not affected by the concentrations between 0.01% and 1.0% of Tsume renge water extract solution. 3. 0.01% of Tsume renge water extract was increased the Na-K-ATPase activity significantly. 4. Na-K-ATPase activity was inhibited in the concentrations of 0.1%, 0.5%, and 1.0% of Tsume renge water extract. From the above results, it was suggested that the water extract of Tsume renge has dual effect in accordance with its concentrations.

        Orostachys malacophyllus Fischer(Tsume renge) is a plant to grow on the tiled roof of the house, and in the field or the mountain. And the compositions of the plant have not been known yet. But it has transmitted to this time as a kind of medical plant from remote antiquity. Sometimes, it is used in the folk remedies for the treatment of the malignant diseases. But it is not clear in the therapeutic effect. This study was attempted to investigate the effect of the water extract of Tsume renge on the renal function of rabbits. Physiological saline was infused into the jugular vein for the hydration of rabbit(30ml/hr), and the urine and blood were collected to take the control values after the steady state. Samples were collected by the regular interval of time during the infusion of 0.5% solution of Tsume renge water extract in the physiological saline. The results obtained were summarized as follows: 1. The urine volume, glomerular filtration rate, and renal plasma flow were 0.095±0.013 ml/min. 5.66±0.39 ml/min. and 21.01±1.52 ml/min, respectively. But their flow rates were decreased significantly during the infusion of the solution of water extract. 2. The water extract of Tsume renge had no any effects on the renal tubular reabsorption of the osmolar substances and the electrolytes. 3. The negative free water clearance was decreased significantly during the infusion of the water extract of Tsume renge. From the above results, it was suggested that the water extract of Tsume renge affected to the renal hemodynamics and the mechanism of water reabsorption in the renal tubules.
