Modulation of the Self-Assembly of Collagen by Phytic Acid: An In Vitro Study

Phytic acid, containing a myoinositol ring coupled with six phosphate groups, can react with the amino groups of collagen to regulate their self-assembly behavior. The aim of this research is to evaluate the effects of phytic acid on the selfassembly behavior of collagen, the structures and properties of the resulting fibrils and hydrogels. Turbidity and chloramine T assay suggested that phytic acid could improve the self-assembly kinetics and degree of collagen, and the optimal ratio of phytic acid/collagen was 1/1 (w/w). Scanning electron microscopy (SEM) analysis indicated that co-fibrils of collagen with phytic acid are more slender than that of pure collagen, and transmission electron microscopy (TEM) reveals that the characteristic D-periodicity of collagen fibrils is not affected by phytic acid. Besides, differential scanning calorimetry (DSC) and rheology revealed that the thermal stability of collagen fibrils and the viscoelasticity of collagen hydrogels could be improved by phytic acid and the optimal ratio of phytic acid/collagen is 1/1 (w/w).

Four-bar linkage has been used in motion generation synthesis for a long time, but there are inevitably some shortcomings such as order defects and poor design flexibility. In this work, a planetary gear train with noncircular gears is proposed to realize planar motion generation synthesis. Compared with the four-bar linkage, it has the advantages of a compact structure, full rotatability, and flexible design. The planetary gear train with noncircular gears is composed of two interrelated parts: a two-stage noncircular gear pair and the RR dyad. The synthesis method is divided mainly into three steps: First, the constraint equation of RR dyad for planar motion generation is established, and a solution region of the double revolute joint (RR) dyads is calculated by kinematic mapping theory. Second, after the full rotatability identification of RR dyads and the determination of each transmission ratio, the centrodes of two-stage noncircular gear pairs, which correspond to the specific RR dyad, are obtained. Finally, the planetary gear train with noncir-cular gears is established to pass the given poses. This method has a universal significance and can be applied to many practical cases.

Serine protease inhibitor Kazal type 1 (SPINK1) plays a role in protecting the pancreas against premature activation of trypsinogen and is involved in cancer progression. SPINK1 promoted LAC cells growth, migration, and invasion. Mechanistically, we found that SPINK1 promoted LAC cells migration and invasion via up-regulating matrix metalloproteinase 12 (MMP12). We observed that SPINK1 expression was only up-regulated in lung adenocarcinoma (LAC) tissues, and was an independent prognostic factor for poor survival. Our results indicate that SPINK1 might be a potential biomarker for LAC that promotes progression by MMP12. [BMB Reports 2018; 51(12): 648-653]
