        Lactobacillus acidophilus Strain Suppresses the Transcription of Proinflammatory-Related Factors in Human HT-29 Cells

        Previous studies have shown that lactic acid bacteria can inhibit inflammatory responses, but the mechanisms are very little known. In this study, transaction and expression of three proinflammatory factors, iNOS, PTGS-2, and IL8, which are closely related to the inflammatory response, were investigated by luciferase reporter assay and RTPCR in HT-29 cells treated by Lactobacillus acidophilus. The results showed that the live L. acidophilus sharply down-regulated the transcription of these three genes. Because there was a NF-κB binding site located at -265 bp, -225 bp, and -95 bp upstream of the iNOS, PTGS-2, and IL8 promoters, respectively, we further addressed the effects of NF-κB on transaction of the three promoters by cotransfection. As was expected, NF-κBs remarkably upregulated the activity of the reporter gene and, no effect of NF-κBs on IL-8 promoter transaction was found after NF-κB binding site mutation of the IL8 promoter in HT- 29 cells. In conclusion, the live L. acidophilus decreased the transcriptional activity of NF-κB and, in turn, inhibited the transaction of NF-κB on the three proinflammatory factors mentioned above.

        NANOSEEDING EFFECT IN ZSM-5CRYSTALLIZATION PROCESS

        ZSM-5 zeolites were hydrothermally synthesized in three different seeding pathways under the direction of tetrapropylammonium bromide (TPABr) template. In order to investigate the seeding effect in ZSM-5 crystallization process, ZSM-5 crystals and pre-fabricated MFI-type nanoseeds were added into the original self-induction system, respectively. The final ZSM-5 zeolites were systematically investigated based on XRD (X-ray diffraction), SEM (scanning electron micrograph), TEM (transmission electron micrograph), nitrogen adsorption characterizations and NH3-TPD (ammonia-temperature programmed desorption). The self-induction system produced ca. 20 μm ZSM-5 zeolite displaying hexagonally uniform prisms. After the addition of ZSM-5 crystal seeds, the crystal sizes were decreased greatly to ca. 5 μm. When MFI-type nanoseeds were adopted, irregular aggregate particles consisting of 20–50 nm primary particles were rapidly synthesized. The varied hydrothermal crystallization kinetics of the three synthesis system was also explored. Adjusting the seed agents alone, ZSM-5 crystals with diverse structural, morphological, textural and hydrothermal behaviors could be fabricated conveniently. The three ZSM-5 zeolites loaded by 0.05 wt.% Pt were assessed for the xylene isomerization reaction to investigate the particle size effect on the catalytic properties.

        Expression of selenium-independent glutathione peroxidase 5 (GPx5) in the epididymis of Small Tail Han sheep

        Objective: Selenium-independent glutathione peroxidase (GPx5) is specifically expressed in the mammalian epididymis and plays an important role in protecting sperm from reactive oxygen species and lipid peroxidation damage. This study investigates GPx5 expression in the epididymis of Small Tail Han sheep. Methods: GPx5 expression was studied in three age groups: lamb (2 to 3 months), young (8 to 10 months), and adult (18 to 24 months). The epididymis of each age group divided into caput, corpus and cauda, respectively. Analysis the expression quantity of GPx5 in epididymis and testis by real-time fluorescent quantitative polymerase chain reaction and Western blot. Finally, GPx5 protein locating in the epididymis by immunohistochemical. Results: The results demonstrate that in the lamb group, the GPx5 mRNA, but not protein, can be detected. GPx5 mRNA and expressed protein were detected in both the young and adult groups. Moreover, both the mRNA and protein levels of GPx5 were significantly higher in the young group than in other two groups. When the different segments of epididymis were investigated, GPx5 mRNA was expressed in each segment of epididymis regardless of age. Additionally, the mRNA level in the caput was significantly higher than that in corpus and cauda within same age group. The GPx5 protein was in the epithelial cells' cytoplasm. However, GPx5 mRNA and protein were not detected in the testis. Conclusion: These results suggest that GPx5 is mainly expressed in the epididymis of Small Tail Han sheep, and that the expression level of GPx5 is associated with age. Additionally, GPx5 was primarily expressed in the epithelial cells of the caput. Taken together, these studies indicate that GPx5 is expressed in the epididymis in all age grades.

        A Novel Recursive Algorithm for Efficient ZF-OSIC Detection in a V-BLAST System

        To reduce the implementation complexity of the Vertical Bell Labs layered space-time (V-BLAST) systems with respect to the zero-forcing (ZF) criterion, a computationally efficient recursive algorithm is proposed. A fast implementation of the proposed algorithm is developed and its complexity is analyzed in detail. The proposed algorithm matches the ZF-OSIC detection well, and its three significant advantages can be demonstrated by analyses and simulations. Firstly, its speedups over the conventional ZF-OSIC with norm-based ordering, the original fast recursive algorithm (FRA) and the fastest known algorithm (FKA) in the number of flops are 1.58, 2.33 and 1.22, respectively. Secondly, a much simpler implementation than FRA and FKA can be expected. Finally, the storage requirements are lower than those of FRA and FKA. These advantages make the proposed algorithm more efficient and practical.

      • hARIP2 is a Putative Growth-promoting Factor Involved in Human Colon Tumorigenesis

        Activin is a multifunctional growth and differentiation factor of the growth factor-beta (TGF-${\beta}$) superfamily, which inhibits the proliferation of colon cancer cells. It induces phosphorylation of intracellular signaling molecules (Smads) by interacting with its type I and type II receptors. Previous studies showed that human activin receptor-interacting protein 2 (hARIP2) can reduce activin signaling by interacting with activin type II receptors; however, the activity of hARIP2 in colon cancer has yet to be detailed. In vitro, overexpression of hARIP2 reduced activin-induced transcriptional activity and enhanced cell proliferation and colony formation in human colon cancer HCT8 cells and SW620 cells. Also, hARIP2 promoted colon cancer cell apoptosis, suggesting that a vital role in the initial stage of colon carcinogenesis. In vivo, immunohistochemistry revealed that hARIP2 was expressed more frequently and much more intensely in malignant colon tissues than in controls. These results indicate that hARIP2 is involved in human colon tumorigenesis and could be a predictive maker for colon carcinoma aggressiveness.
