Upconversion Nanoparticles as a Contrast Agent for In Vivo Photoacoustic Imaging

Lanthanide doped upconversion nanomaterials have proven their great potentiality in modern biological applications, however, tracing UC materials from tissue depths still remains challenging due to visible emission, increased optical extinction in biological tissues and luminescence quenching in aqueous/biological conditions, which further limits their optical resolution in vivo. In this work, we report the use of NaYF4:Yb3+,Er3+ UC materials for in vivo photoacoustic imaging (PAI) by making use of hitherto unexplored luminescence quenching effect of UC materials in water. We observed a significant luminescence quenching effect of UC-α-CD in water upon 980 nm excitation. The observed non-radiative relaxation enhances thermal conductivity and subsequent PA signal of UC-α-CD in water. PAI properties of UC-α-CD were then investigated, and its application for in vivo high resolution PAI at 980 nm was demonstrated in live mice and thus proved as an excellent candidate for in vivo PAI.
