The glycosylation pattern of Erythropoietin (EPO), produced by recombinant CHO cells, was studied using the simple and rapid technique of 'Lectin-blotting'. In this experiment we used three different kinds of lectins, MAA(Maackia amurensis agglutinine), RCA(Ricinus communis agglutinine), and DSA(Datura stramonium agglutinine), which bind to the terminal sialic acid, galactose, and the N-acetyllactosamine chain respectively. The lectin-blotting technique was used to analyze the carbohydrate structure of EPO produced in the presence of two physiologically active chemical compounds, ammonium and chloroquine. The effect of the ammonium ion on the glycosylation of EPO was studied because it accumulated in the medium mainly as a by-product of glutamine matabolism. Ammonium chloride significantly inhibited the sialylation of the terminal galactose residue at concentrations of 8mM or more. Chloroquine, a potent inhibitor of glycosylation, inhibited terminal sialylation at concentrations of 100 and 200 $\mu$M, and at a concentration of 300 $\mu$M, also inhibited Nacetyllactosamine chain synthesis.

We have previously reported that N-acetylcysteine (NAC) not only delayed apoptosis but also enhanced the production of recombinant erythropoietin (EPO) in Chinese hamster ovary (CHO) cell culture. To investigate the production enhancement mechanism, the effects of similar thiolreducing agents were studied. Intriguingly, all mild reducing agents examined including mercaptoethanesulfonic acid (MESNA), thiolactic acid (TLA), and thioglycolate (TG) were shown to block apoptosis and increase EPO production. A pulse-chase study of EPO secretion revealed that all four thiol-reducing agents increased the EPO secretion rate; among them TLA showed the highest rate. In terms of product quality, the sialic acid content of the glycoprotein is one of the most important factors. It was reported that a number of glycoproteins produced by CHO cells often have incomplete sialylation, particularly under high-producing conditions. Human α2,3-sialyltransferase (α2,3-ST) was introduced into EPO-producing CHO cells in order to compensate for the reduced sialylation during supplementation with NAC. When α2,3-ST was expressed in the presence of NAC, reduced sialylation was restored and an even more sialylated EPO was produced. Thus, our study is significant in that it offers increased EPO production while still allowing the prevention of decreased sialylation of EPO.

휴전일정은 매월 월간단위로 처리된다. 요청건수는 월간 500여건에 이르며, 그 중 휴전시 계통의 안정적 운영에 크게 영향을 미칠 수 있는 765㎸, 345㎸급 설비의 휴전 건수도 200여건에 이른다. 요청된 설비의 휴전일정을 계통의 안정도에 미치는 영향을 최소화하여 일정을 조정해야 한다. 본 논문에서는 자동화되고 체계적인 휴전일정 검토를 위한 프로그램개발에 대해서 기술한다. 먼저 휴전일정 검토절차를 분석하여 요구사항을 분석하고, 이러한 정보를 효과적으로 저장하기 위한 DB설계방법을 기술 하였으며, 일정 검토자가 편리하고 효율적으로 일정을 검토할 수 있도록 사용자 화면을 개발하였다.

휴전검토는 계통의 안정적인 운영과 신뢰도에 직접적으로 영향을 줄 수 있어 매우 중요한 업무라 할 수 있다. 본 논문에서는 이러한 휴전검토 업무를 체계적이고 효율적으로 수행할 수 있는 휴전검토 종합시스템의 역할을 간단히 소개하고, 구성요소 중 하나인 휴전일정 관리 및 검토 프로그램 설계에 대해 설명하고자 한다.
