        Preservation of the periodontal ligament cell viability on an avulsed tooth is very important to obtain the successful outcome of replantation. The purpose of this study was to evaluate the effect of EGCG as a storage media on the viability of human PDL cell from avulsed tooth. Human PDL cells were collected from the healthy premolar teeth extracted for orthodontic treatment. PDL cells were stored in the various concentration of EGCG in HBSS. Time interval was 0, 1, 3, 6, 12, 24hours. The viability of PDL cells were determined using MTS assay and Nucleocounter?? system. 1. The MTS assay analysis indicated that the viability of PDL cells in less than 5μM EGCG was similar with the HBSS Group in every time (P<0.05). As time increases, viabilities of PDL cells in 10, 20 and 50μM EGCG were decreased compared with the HBSS Group (P<0.05). Viabilities of PDL cells in 100μM EGCG and tap water were remarkably decreased (P<0.05). 2. The Nucleocounter?? system analysis showed that the survival rate of PDL cells in less than 20μM EGCG was similar with HBSS Group (P<0.05). The survival rate of PDL cells in 50μM EGCG was decreased as time passes more than 3 hours (P<0.05). Survival rates of PDL cells in 100μM EGCG and tap water were significantly decreased (P<0.05). 3. Results of the Nucleocounter?? system and MTS assay analysis were showed similar results and the viability and the survival rate of PDL cells in more than 50μM EGCG were decreased when stored more than 12 hours (P<0.05). In this study, it is evaluated that the administration of EGCG could influence on the cell viability and the proper concentration of the EGCG in the storage of PDL cells was predicted. Thus, further studies about the storage media using the HBSS containing less than 20 μM EGCG are needed to determine the maintenance of cell vitality, the anti-oxidant property, the anti-bacterial effect and the anti-inflammatory action.
