Developmentally Regulated Genes in Brassica

Partial cDNA sequencing was conducted on approximately 150 randomly selected cDNA clones of Brassica napus. The cDNA library of root organ of Brassica were constructed in a plasmid vector, to perform double stranded DNA sequencing protocol. The sequencing were resolved on a conventional acrylamide sequencing gel and 150-300 base pair readings were obtained from each reactions. The generated sequences are being analyzed for similarites in PIR 29 protein database and GenBank nucleic acid database. Of the 40 clones analyzed so far, 15 clones represent new genes;especially one clone has significant similarities to calreticulin, an animal Ca^(++)-binding protein. Based on these results, we are developing better experimental procedures for random cDNA sequencing in plants. This approach will facilitate the isolation of novel or functionally related genes in Brassica, providing a powerful genetic resource in plant molecular biology.; Ras- and protein kinase-related genes from Brassica napus are being identified and isolated. First, genomic southern analysis was performed to reveal the presence of ras- and protein kinase- related genes in Brassica napus, using the corresponding Arabidopsis clones as probes. Next, using the synthetic oligonucleotides designed to hybridize to highly conserved domain of protein kinases from animal and Arabidopsis thaliana, polymerase chain reation product was obtained from cDNA and was confirmed, by southern analysis, that the fragment has homology to the Arabidopsis clone we have isolated previousely. We are also isolating ras- and protein kinase-related clones by screening the cDNA library of B. napus with the probe of genomic clone and cDNA clone from Arabidopsis thaliana.
