      • No association Between Calcium Channel Blockers and Survival in Patients with Cancer: A systematic Review and Meta-analysis

        Background: The association between calcium channel blockers (CCBs) and survival in cancer patients remains unclear and the results of related studies are conflicting. The objective of the study was to investigate the association between calcium channel blockers (CCBs) use and survival in cancer patients. Materials and Methods: We searched PubMed, EMBASE, Web of Science and Cochrane Library for studies published before January 2016 with the terms related to CCBs and survival in cancer patients. The information was reviewed and extracted by two evaluators independently. Data of publications was extracted and calculated into hazard ratios (HRs) for overall survival (OS). Statistical analysis was performed by using Review Manager 5.3. Results: There were 11 studies included in our meta-analysis. Analysis of all studies showed that CCBs use was not associated with survival in cancer patients (HR=1.07; 95% CI: 0.91-1.25; P=0.42). No association between CCBs use and overall survival in cancer patients was existed whether in Asian (HR=1.18, 95% CI: 0.72-1.93; P=0.52) or Caucasian population (HR=1.03, 95% CI: 0.89-1.20; P=0.66). Conclusions: There is no evidence that CCBs use is associated with a better or worse outcome of survival in cancer patients.

      • Aberrant Expression of Pim-3 Promotes Proliferation and Migration of Ovarian Cancer Cells

        Pim kinase-3(Pim-3), a member of serine/threonine protein kinases, has been implicated in multiple human cancers and involved in Myc-induced tumorigenesis. However, little is known regarding its expression and biological function in human ovarian cancer. In this study we showed that the clinical significance and biological functions of Pim-3 in ovarian cancer and found that higher Pim-3 mRNA level are detected in ovarian cancer tissues than those in normal ovarian tissues. There are significant correlations between higher Pim-3 expression levels with the FIGO stage, histopathological subtypes, and distant metastasis in ovarian cancer patients. Lentivirus-mediated gene overexpression of Pim-3 significantly promotes the proliferation and migration of SKOV3 cell lines. Furthermore, MACC1 and Pim-3 expression were significantly correlated in human ovarian cancer cells, and overexpression of Pim-3 in ovary cancer cells increased MACC1 mRNA and protein expression. The data indicate that Pim-3 acts as a putative oncogene in ovary cancer and could be a viable diagnostic and therapeutic target for ovarian cancer.

        Review : Review on Research and Development of Shale Gas in China

        Shale gas reserves in China are large, although shale gas production is still in its infancy. This paper reviews Chinese national policies and guidelines related to shale gas development, in particular those related to recent progress in the development of shale gas reserves in Sichuan Province and Chongqing since February 2015. In addition, three large-scale R&D projects related to shale gas development, funded by the Government of China, are introduced, and the scope of the work in each project is described.

      • Recombinant Growth Differentiation Factor 9 and Bone Morphogenetic Protein 15 Decrease Kit Ligand Gene Expression in Canine Ovarian Granulosa Cells

        Prolonged communication between oocytes and the surrounding somatic cells is one of the unique reproductive physiology in canine. Paracrine Kit ligand (KITL) signaling is a well-known communication between granulosa cells and the oocyte. KITL is a cytokine growth factor secreted by granulosa cells that signals via the c-kit receptor expressed by oocytes. Paracrine factors, including growth differentiation factor 9 (GDF9) and bone morphogenetic protein 15 (BMP15), exert their effects by binding with the kinase receptors expressed on the granulosa cells. However, the regulations of GDF9 and BMP15 in the canine KITL expression are currently poorly understood. Therefore, we investigated the effects of GDF9 and BMP15 on the expression of KITL in canine ovarian granulosa cells in vitro. In Annexin V assay recombinant GDF9 and BMP15 did not induce apoptosis in the cultured ovarian granulosa cells. When treated, FSH significantly increased KITL expression, and hCG suppressed its expression. When both FSH and hCG were treated, the expression of KITL was affected by GDF9 and BMP15 in dose and time dependent manner in the luteal granulosa cells. GDF9 (10 ng/mL) significantly decreased KITL expression after12 h. BMP15 (10 ng/mL) significantly also decreased KITL expression after 24 h. Western blot and immunochemistry results indicate that GDF9 activated Smad2/3. After blocking ALK 4/5/7 receptors by SB, GDF9 failed to activate Smad2/3, also BMP15 did not activate Smad1/5/8 after blocking ALK 2/3/6 receptors by DM. So GDF9 exerts its effects via using ALK 4/5/7 receptors to activate SMAD2/3 signaling, and BMP15 binds ALK 2/3/6 receptors to activate SMAD1/5/8 signaling. The expression of KITL was not changed by SB or DM treatment. However, the effect of GDF9 and BMP15, which decreased the expression of KITL, was suppressed by SB or DM treatment. In conclusion, this study provides the first evidence that recombinant GDF9 and BMP15 decrease KITL expression in canine ovarian granulosa cells.

        Back-stepping Active Disturbance Rejection Control for Attitude Control of Aircraft Systems Based on Extended State Observer

        Robust flight control laws based on back-stepping technology and ADRC method are designed for attitude control of a non-linear aircraft system. First, non-linear aircraft model is introduced and converted to standard equation of state. Then, the extended state observer is applied to estimate the unknown variables, the homologous ADRC is designed to ensure the state variables of the CLS to astringe to the reference state. Next, the stability of ESO and ADRC are analyzed and proven theoretically. At last, the effectiveness of this method is illustrated by extensive comparative simulations. The results acquired from simulation attest that ADRC can achieve better control performance than PID and SMC method.

        Active Disturbance Rejection Control for Attitude Control of Missile Systems Based on Back-stepping Method

        In this paper, a novel attitude control method for nonlinear missile system is proposed, which combines backstepping technique with linear active disturbance rejection control (LADRC). Firstly, the nonlinear missilemodel is introduced and transformed into the standard state equation. The linear extended state observer (LESO) is used to estimate the unknown variables. Compared with PID control method, the homologous LADRC designensures that the state variables of the closed-loop system converge to the reference state. The stability of LESO and LADRC is theoretically analyzed and proved. A large number of comparative simulations show the effectiveness ofthe method. The simulation results show that LADRC has better control performance than SMC and PID method.

        MEG3 LncRNA from Exosomes Released from Cancer-Associated Fibroblasts Enhances Cisplatin Chemoresistance in SCLC via a MiR-15a-5p/CCNE1 Axis

        Purpose: Long non-coding RNAs (lncRNAs) may act as oncogenes in small-cell lung cancer (SCLC). Exosomes containing lncRNAsreleased from cancer-associated fibroblasts (CAF) accelerate tumorigenesis and confer chemoresistance. This studyaimed to explore the action mechanism of the CAF-derived lncRNA maternally expressed gene 3 (MEG3) on cisplatin (DDP) chemoresistanceand cell processes in SCLC. Materials and Methods: Quantitative real-time PCR was conducted to determine the expression levels of MEG3, miR-15a-5p, andCCNE1. Cell viability and metastasis were measured by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-h-tetrazolium bromide andinvasion assays, respectively. A xenograft tumor model was developed to confirm the effect of MEG3 overexpression on SCLC progressionin vivo. Relationships between miR-15a-5p and MEG3/CCNE1 were predicted using StarBase software and validated bydual luciferase reporter assay. Western blotting was used to determine protein levels. A co-culture model was established to explorethe effects of exosomes on MEG3 expression in SCLC cell lines. Results: MEG3 was overexpressed in SCLC tissues and cells. MEG3 silencing significantly repressed cell viability and metastasis inSCLC. High expression of MEG3 was observed in CAF-derived conditioned medium (CM) and exosomes, and promoted chemoresistanceand cancer progression. Additionally, MEG3 was found to serve as a sponge of miR-15a-5p to mediate CCNE1 expression. Overexpression of miR-15a-5p and knockout of CCNE1 reversed the effects of MEG3 overexpression on cell viability and metastasis. Conclusion: MEG3 lncRNA released from CAF-derived exosomes promotes DDP chemoresistance via regulation of a miR-15a-5p/CCNE1 axis. These findings may provide insight into SCLC therapy.

      • Regulatory Roles of Oocyte Secreted Factors in Canine Cumulus-oocyte Complexes and Granulosa Cells

        Oocyte is the central factor in the bi-directional communication axis in the ovarian follicles. It controls the cumulus or granulosa cells to perform functions which are beneficial for its own development via secreting paracrine growth factors, including GDF9 and BMP15. The aim of this study was to investigate whether the recombinant GDF9 and BMP15 are able to promote meiotic resumption and cumulus expansion of canine COCs during IVM, as well as to demonstrate the actions of GDF9 and BMP15 in regulating the expression of connexin transcripts in the ovarian granulosa cells. As results, GDF9 and BMP15 significantly improved the meiotic resumption rate and cumulus expansion by activating ERK1/2 signaling. Treatments with GDF9 significantly improved the expression of CyclinB1 but inhibited the expression of Cx43 transcripts. In addition, cumulus expansion genes (MAPK1, Ptgs2, Tnfaip6 and Ptx3) were differentially improved by GDF9 and BMP15. In the ovarian granulosa cells, GDF9 suppressed the expression of Cx43 transcripts by binding ALK4/5/7 receptors and activation Smad2/3 signaling, whereas, BMP15 stimulated the expression of Cx43 transcripts by binding ALK2/3/6 receptors and activating Smad1/5/8 signaling. In conclusion, by regulating functions of granulosa/cumulus cells, oocyte has the potential to enhance the growth and maturation of itself.

        Experimental Evaluation of Effect Factors on Seismic Performance of Concrete Columns Reinforced with HTRB630 High-Strength Steel Bars

        HTRB630 steel bar is a new type of high-strength steel bars. To study the seismic performance and promote the application of concrete columns reinforced with HTRB630 high-strength steel bars, the pseudo-static test of 10 concrete columns reinforced with HTRB630 high-strength steel bars and 3 concrete columns reinforced with HRB400 was carried out. Test specimens were divided into five categories according to concrete grade, reinforcement strength, and degree of confinement. The effect of concrete strength (C45 and C60), axial load ratio (0.1 and 0.25), equal strength substitution of longitudinal reinforcements (HTRB630 and HRB400), equal strength and volume substitution of stirrups (HTRB630 and HRB400), equal strength substitution of confined reinforcements (HTRB630 and HRB400) on the seismic performance are analyzed and discussed from the failure mode, hysteresis loops, skeleton curves, lateral strength, ductility, energy dissipation capacity, stiffness and strength degradation. The failure mode of each specimen was bending failure. Reducing of the axial lo ratio or increasing the strength of reinforcements, the seismic performance of the specimen could be improved. C60 concrete can improve the seismic performance of HTRB630 reinforced columns. The lateral strength of the concrete column specimen with HTRB630 reinforcements was slightly increased, and the ductility and energy dissipation capacity were reduced, the stiffness degradation was more gradual, the strength degradation coefficient was greater than 0.94, but still met the requirements of the code for seismic design of buildings (GB50011-2010).
