        Fabrication of Photopolymer Hierarchical Micronanostructures by Coupling Electrospinning and Photolithography for SERS Substrates

        Reported here is the fabrication of photopolymer hierarchical micronanostructures through a combinative process of electrospinning and subsequent photolithography. Electrospun SU-8 (epoxy-based negative photoresist)nanofiber films have been patterned into gratings with periods of 100, 200, 300, and 400 μm, respectively. Deposition of a silver nanolayer on these interlaced nanofiber films would lead to the formation of various plasmonic nanostructures,and therefore, giving rise to abundant surface-enhanced Raman scattering (SERS) “hot spots”. In the detection of Rhodamine 6G (R6G), probing molecule, the resultant SERS substrates show both high sensitivity and good reproducibility. The SERS enhancement factor could reach as high as ~108, indicating high efficiency. The fabrication of patterned, highly efficient SERS substrates may hold a great promise for the integration of SERS substrates in various microdevices such as microfluidic chips.

        Study on Joint Simulation of Very Fast Transient Overvoltage and Transient Enclosure Voltage in 1000 kV GIS

        Disconnector operation in GIS generates VFTO and TEV, which threaten device insulation and personnel safety. In order to calculate VFTO and TEV more accurately, this paper proposes a VFTO-TEV joint simulation model. Among them, dynamic arc model is used to reflect the dynamic characteristics of high frequency AC arc. In order to reflect the electromagnetic leakage phenomenon caused by the impedance mismatch of high-frequency electromagnetic waves, and the influence of the transient potential difference formed by the transient electromagnetic field on grounding grid on TEV, the geometric structures of high-voltage bushing, grounding pillars and grounding grid are modeled to obtain their respective frequency-dependent admittance matrices, which are converted into corresponding broadband equivalent circuit models by combining vector fitting method and impedance synthesis method. The above modeling method is applied to the 1000 kV GIS test circuit, and the simulation model is built in the ATP-EMTP electromagnetic transient program, and the joint simulation research of VFTO and TEV is carried out. The results show that at each measurement point of the test circuit, the amplitude error of the VFTO obtained by simulation and actual measurement is mostly less than 7%, and the frequency spectrum both contains the main frequency components of 18 MHz and 25 MHz; Compared with the traditional calculation model of obtaining VFTO/TEV using CIGRE model/Petersen's law, the amplitude of VFTO and TEV obtained by joint simulation model is lower and contains richer high-frequency components. In addition, the opening speed of 0.8 m/s also corresponds to the maximum point of the VFTO amplitude of the test circuit. Finally, based on the above modeling method, the effectiveness of adding ferrite beads to suppress VFTO is verified by simulation. © 2017 Elsevier Inc. All rights reserved.

        Oxygen Uptake Rate Controlling Strategy Balanced with Oxygen Supply for Improving Coenzyme Q10 Production by Rhodobacter sphaeroides

        The effects of different oxygen uptake rates (OUR) on the physiological metabolism of Rhodobacter sphaeroides were investigated systematically in 50 L fermenters due to the significant influence on industrial coenzyme Q10 production under oxygen supply limitation. Meanwhile, the seriously decreased oxygen transfer rate caused by the increased broth viscosity was successfully prevented with OUR-directed continuous ammonium sulfate feeding in the late fermentation phase. The statistical analysis results showed that controlling OUR constantly at 45 ± 2.2 mmol/L/h by the oxygen supply level adjustment and the continuous ammonium sulfate feeding could greatly enhance Q10 production. This OUR-Stat controlling strategy successfully achieved the maximal coenzyme Q10 production (2584 ± 82 mg/L), which was 15.4% higher than that of the control. The highest specific CoQ10 content was 25.9 mg/(g DCW)), and the yield of CoQ10 to glucose consumption was up to 19.37 mg/g. These results demonstrated that the optimal OUR-Stat controlling strategy would be effective and economical for improving the industrial CoQ10 production.

        Cloning, characterization, and RNAi effect of the chitin synthase B gene in Cnaphalocrocis medinalis

        The rice leaf folder (RLF), Cnaphalocrocis medinalis, is one of the major pests of rice, and chitin synthase is a key enzyme for the chitin synthesis pathway in insects. In this study, the chitin synthase B gene from C. medinalis (CmCHSB) was cloned and characterized. The cDNA of CmCHSB is 4824 bp in length, containing an open reading frame of 4578 nucleotides that encodes 1525 amino acids. The CmCHSB zymoprotein consists of 10 trans membrane domains (TMDs) in the N-terminus, a middle conserved catalytic domain, and 7 TMDs in the C-ter minus. Homology and phylogenetic analyses revealed that CmCHSB possesses the closest relationship with its homolog in Ostrinia furnacalis. CmCHSB was expressed throughout development and in all of the adult tissues tested, with the highest expression level in the adult and in the midgut. Silencing of CmCHSB through RNA interference (RNAi) severely affected RLF larval growth and caused larval lethality. Our results revealed that CmCHSB is essential for RLF growth and development, which sheds new light on the characteristics and functions of this gene. These findings will be helpful for green control of RLF, by targeting the CmCHSB gene using RNAi technology.

        Cross-immunizing potential of tumor MAGE-A epitopes recognized by HLA-A*02:01-restricted cytotoxic T Lymphocytes

        Almost all melanoma cells express at least one member of the MAGE-A antigen family, making the cytotoxic T cells (CTLs) epitopes with cross-immunizing potential in this family attractive candidates for the broad spectrum of anti-melanoma immunotherapy. In this study, four highly homologous peptides (P264: FLWGPRALA, P264I9: FLWGPRALI, P264V9: FLWGPRALV, and P264H8: FLWGPRAHA) from the MAGE-A antigens were selected by homologous alignment. All four peptides showed high binding affinity and stability to HLA-A*02:01 molecules, and could prime CTL immune responses in human PBMCs and in HLA-A*02:01/Kb transgenic mice. CTLs elicited by the four epitope peptides could cross-lyse tumor cells expressing the mutual target antigens, except MAGE-A11 which was not tested. However, CTLs induced by P264V9 and P264I9 showed the strongest target cell lysis capabilities, suggesting both peptides may represent the common CTL epitopes shared by the eight MAGE-A antigens, which could induce more potent and broad-spectrum antitumor responses in immunotherapy.       • Low Microsomal Epoxide Hydrolase Expression is Associated with Bladder Carcinogenesis and Recurrence

        Microsomal epoxide hydrolase (mEH) plays a significant role in the metabolism of numerous xenobiotics and is associated with several forms of cancer. Here, we investigated the role of mEH expression in bladder carcinogenesis, subsequent progression and recurrence. The expression of mEH was analyzed by Western blot in 50 bladder urothelial carcinoma and 20 normal epithelial tissues. There was a significantly higher mEH expression in the normal epithelium (P<0.05) and mEH expression was lower in high stage than in low stage tumors (P<0.05). Further, immunohistochmistry in 106 bladder urothelial carcinoma demonstrated mEH expression to be negatively correlated with histological grade, pT stage and recurrence (P<0.05). These findings suggest the important role of mEH in bladder carcinogenesis, cancer development and recurrence, providing support for efforts to develop mEH-based gene therapy.

      • Comprehensive Expression Analysis Suggests Functional Overlapping of Human FOX Transcription Factors in Cancer

        Forkhead-box (FOX) transcription factors comprise a large gene family that contains more than 50 members in man. Extensive studies have revealed that they not only have functions in control of growth and development, but also play important roles in different diseases, especially in cancer. However, biological functions for most of the members in the FOX family remain unknown. In the present study, the expression of 39 FOX genes in 48 kinds of cancer was mined from the Gene Expression Atlas database of European Bioinformatics Institute. The analysis results showed that some FOX genes demonstrate overlapping expression in various cancers, which suggests particular biological functions. The pleiotropic features of the FOX genes make them excellent candidates in efforts aimed to give medical treatment for cancers at the genetic level. The results also indicated that different FOX genes may have the synergy or antagonistics effects in the same cancers. The study provides clues for further functional analysis of FOX genes, especially for the pleiotropic biological functions and crosstalk of FOX genes in human cancers.

        Effects of adaptation time and inclusion level of sugar beet pulp on nutrient digestibility and evaluation of ileal amino acid digestibility in pigs

        Objective: Two experiments were conducted to determine the effects of adaptation time and inclusion level of sugar beet pulp (SBP) on nutrient digestibility and to evaluate the ileal amino acid digestibility of SBP fed to pigs. Methods: In Exp. 1, thirty-six crossbred barrows (85.0±2.1 kg) were allotted to 6 diets in a completely randomized design with six replicates per diet. Diets included a corn-soybean meal diet and 5 test diets containing 14.6%, 24.4%, 34.2%, 43.9%, or 53.7% SBP, respectively. The adaptation time consisted 7, 14, 21, or 28 d consecutively for each pig followed by 5 d for fecal collection. Feces were collected from d 8 to 13, d 15 to 20, d 22 to 27, and d 29 to 34, respectively. In Exp. 2, six pigs (35.1±1.7 kg) with T-cannulas at the terminal ileum were fed to 3 diets in a replicated 3×3 Latin square design with 3 periods and 2 replicate pigs per diet. Each period consisted 5 d for diet adaptation followed by 2 d for digesta collection. Results: The digestible energy (DE) value and the apparent total tract digestibility (ATTD) of gross energy (GE), dry matter (DM), ash, and organic matter in diets linearly decreased (p< 0.05) as the adaptation time increased or as the dietary SBP increased, while the ATTD of neutral detergent fibre and acid detergent fibre in diets linearly increased (p<0.01) as the dietary SBP increased. The DE value and the ATTD of GE and crude protein (CP) in SBP linearly increased (p<0.05) as the adaptation time increased, while the ATTD of CP in SBP linearly decreased (p<0.01) as the inclusion level increased. The standardized ileal digestibility of Lys, Met, Thr, and Trp in SBP was 37.03%, 51.62%, 40.68%, and 46.22%, respectively. Conclusion: The results of this study indicated that the ATTD of energy and nutrients were decreased as inclusion rate of SBP increased.

        한국기업의 중국 내수시장 진입을 위한 물류거점 매력도분석에 관한 연구

        본 연구는 중국 내수 물류시장에 진입하고자 하는 한국 물류기업을 위한 거점 대안을 제시하고자 하는 목적으로 수행되었다. 이를 위한 연구방법론으로는, 중국정부의 물류관련 정책 상의 물류거점 분석, 5개 물류업종을 대표하는 중국 물류기업의 통합물류 네트워크 거점 분석, 거점선택 요인의 중요도 평가를 위한 AHP 분석, 후보 거점의 매력도를 평가하기 위한 SGP 분석, 그리고 글로벌물류기업 및 한국물류기업의 거점과 본 매력도 분석 결과 거점과의 비교분석 등이 사용 되었다. 본 연구는 상해, 청도, 천진, 광주, 심천, 중경, 제남, 소주, 심천, 대련, 무석 등을 매력적인 거점도시로 파악하였으며, 중경, 제남, 장사, 정주, 서안, 장춘, 무한, 남경, 호화호특, 태원, 합비 등을 한국기업을 위한 거점도시로 제안하였다. This research attempts to provide logistics hub location options for Korean Logistics firms which are interested in China domestic market. Research methodologies in the paper include analyses on logistics hub locations of logistics-related Chinese government policies and Chinese logistics firms representing five different types of logistics industries; AHP analysis to identify importance of factors in hub location selection decision; and SGP analysis to analyze attractiveness of potential locations. In this research Shanghai, Qingdao, Tianjin, Guangzhou, Shenzhen, Chongqing, Jinan, Suzhou, Shenyang, Dalian, and Wuxi were identified as attractive hub alternative locations and Chongqing, Jinan, Changsha, Zhengzhou, Xian, Changchun, Wuhan, Nanning, Huhehaote, Taiyuan, and Hefeias were suggested as hub locations for Korean logistics firms.
