구강 악성 흑색종에서 PCNA 발현에 관한 면역조직화학적 연구

Malignant melanoma arising from the mucosa of head and neck region was rare and showed poor prognosis. Some of malignant melanoma were transformed from benign melanotic lesion. Malignant melanoma had high cellular proliferation and rapid growth. The percentage of PCNA-positive cell (labeling index) is high in many malignant tumor. So we compared the pattern of PCNA expression in the melanotic lesion. We performed the immunohistochemical study in malignant melanoma(19 cases), benign melanotic macule(24 cases) and normal mucosa(20 cases) were diagnosed in Seoul National University Dental Hospital between 1980 and 2000. Positive PCNA staining was found mainly in malignant melanoma. The mean PCNA expression in malignant melanoma, melanotic macule, normal tissue were 29.2%, 1.4%, 0%, respectively. Significant differences in PCNA expression were noted between malignant melanoma and melanotic macule(p<0.01), normal mucosa(p<0.01). These result suggested that the PCNA expression seems to be used as a diagnostic indicator for malignancy in malignant melanoma and melanotic lesion.
동종골과 다공성 하이드록시 아파타이트의 골 유합능 비교

목적: 동종골과 다공성 하이드록시 아파타이트(본그로스-HA<SUP>®</SUP>)를 이용하여 백색 가토의 후외방 유합술을 시행함으로써 유합 능력의 차이를 비교하고자 하였다. 대상 및 방법: 3-3.5 kg의 뉴질랜드산 수컷 백색 가토의 양측 제4-5 요추 횡돌기를 후외방 도달법으로 노출한 후 각각 3 cc의 동종골 또는 다공성 하이드록시 아파타이트를 이식하였다. 15마리씩 두 군으로 나누었고, 술 후 12주에 전체를 희생하여 육안 및 도수 촉진 검사, 방사선학적 검사, 인장 검사, 조직학적 검사 등을 통해 골형성 및 유합 능력을 평가하였다. 동종골을 이식한 군에서 생육 도중 6마리가 사망하여 부검을 시행하였으나 특이 소견은 관찰되지 않았다. 결과: 술후 12주에 동종골 이식 군에서는 9마리 중 5마리(55.6%)가 완전 유합, 4마리가 부분 유합으로 판정되었고, HA 이식 군에서는 15마리 중 10마리(66.7%)가 완전 유합, 4마리가 부분 유합, 1마리에서 불유합의 소견을 보였다. 인장 검사 상 유합체의 평균 파단점은 동종골 이식 군에서 140.7 N, HA 이식 군에서 189.6 N이었다. 각 군 당 2마리씩 시행한 조직학적 검사에서는 HA 이식 군의 골전도 양상이 다소 좋은 결과를 나타내었다. 결론: 골 유합능 비교 실험에서 통계적으로 유의한 차이가 없었으나 HA 이식군에서 다소 우수한 결과를 보였다. Purpose: An experimental animal study was performed to compare the bone fusion capacity of an allograft and porous hydroxyapatite. Materials and Methods: Three milliliters of allograft or porous hydroxyapatite particles were inserted between the 4<SUP>th</SUP> and 5<SUP>th</SUP> lumbar transverse processes of New Zealand white rabbits weighing 3-3.5 kg. The total number of rabbits was 30, which were divided randomly into 2 groups. The bone formation and fusion capacity were evaluated 12 weeks after surgery through the gross findings and manual palpation, as well as radiological, biomechanical, and histological studies. Six rabbits in the allograft group died during breeding but the autopsy finding did not show any evidence suggesting an infection or graft rejection. The allograft was harvested from the iliac crest of the rabbits of the same species aseptically and was preserved at - 80°C for at least 7 days before implantation. Results: The fusion rates were 55.6% (5/9) and 66.7% (10/15) in the allograft and porous hydroxy-apatite groups, respectively. The mean values of the tensile strengths were 140.7 N in the allograft group and 189.6 N in the porous hydroxyapatite group. Histological analysis of 2 specimens from each group revealed theporous hydroxyapatite group to show a slightly better osteoconduction capacity. Conclusion: The porous hydroxyapatite group showed better bony union capacity even though there was no significant difference between the 2 groups.

<P>CD109 gene encodes a glycosylphosphatidylinositol-linked glycoprotein found in a subset of platelets and endothelial cell, and human platelet antigen (HPA) 15 is found on CD109. We evaluated the HPA genotype and/or the CD109 mRNA expression on two peripheral blood stem cells (PBSC), two peripheral bloods (PB), 12 granulocyte products, natural killer (NK)-92, B-lymphocyte (CO88BV59-1), K-562 leukemia cell line, human embryonic stem cell (hESC), and human fibroblasts (HF). HPA genotyping was performed by SNaPshot assay and CD109 mRNA expression was evaluated by real-time PCR with SYBR green and melting curve analysis. Genotype HPA-15a/-15a was found in PBSC#1 and two granulocyte products, and HPA-15a/-15b was found in PBSC#2, eight granulocyte products, NK-92, K-562, hESC, and HF, and HPA-15b/-15b was found in two granulocyte products. CD109 mRNA expression was highly increased in HF and increased in CD34+ and CD34− PBSCs and some granulocyte products, compared to the PB. However, the increase of expression level varied among the PBSC and granulocyte products. The CD109 mRNA expression of NK-92, K-562, hESC, and CO 88BV59-1 was not detected. HPA genotype was evaluated in various cells and the expression of CD109, which contains HPA 15, was different among cell lines and high in HF and PBSCs.</P>

<P>Prediction of malignancy or invasiveness of branch duct type intraductal papillary mucinous neoplasm (Br-IPMN) is difficult, and proper treatment strategy has not been well established. The authors investigated the characteristics of Br-IPMN and explored its malignancy or invasiveness predicting factors to suggest a scoring formula for predicting pathologic results. From 1994 to 2008, 237 patients who were diagnosed as Br-IPMN at 11 tertiary referral centers in Korea were retrospectively reviewed. The patients' mean age was 63.1 ± 9.2 yr. One hundred ninty-eight (83.5%) patients had nonmalignant IPMN (81 adenoma, 117 borderline atypia), and 39 (16.5%) had malignant IPMN (13 carcinoma in situ, 26 invasive carcinoma). Cyst size and mural nodule were malignancy determining factors by multivariate analysis. Elevated CEA, cyst size and mural nodule were factors determining invasiveness by multivariate analysis. Using the regression coefficient for significant predictors on multivariate analysis, we constructed a malignancy-predicting scoring formula: 22.4 (mural nodule [0 or 1]) + 0.5 (cyst size [mm]). In invasive IPMN, the formula was expressed as invasiveness-predicting score = 36.6 (mural nodule [0 or 1]) + 32.2 (elevated serum CEA [0 or 1]) + 0.6 (cyst size [mm]). Here we present a scoring formula for prediction of malignancy or invasiveness of Br-IPMN which can be used to determine a proper treatment strategy.</P>

This research investigated the effect of mechanical pretreatment and ozone/catalytic oxidation on the reduction of sewage sludge. The pretreatment process was carried out to enhance sludge solubilization. The degree of solubilization was evaluated based on the DR. TS 1∼4% sludge was treated by ball mil using glass beads of 1 mm in diameter with the 50% (v/v) zircon bead loading. TS 4% showed 4 times higher solubilization efficiency than that of untreated sludge. The combination of the mechanical pretreatment and ozone/catalytic oxidation(10 mg Mn/g TS) showed higher efficiency than ozone/catalytic oxidation. At the 40% solubilization efficiency of sludge achieved by using ball mill, combination treatment showed 3 times higher than control. Ozone/catalytic oxidation improved the solid reduction effectively in the lower pH, presumably due to a high production of OH radical.
Complementary p- and n-Type Polymer Doping for Ambient Stable Graphene Inverter

<P>Graphene offers great promise to complement the inherent limitations of silicon electronics. To date, considerable research efforts have been devoted to complementary p- and n-type doping of graphene as a fundamental requirement for graphene-based electronics. Unfortunately, previous efforts suffer from undesired defect formation, poor controllability of doping level, and subtle environmental sensitivity. Here we present that graphene can be complementary p- and n-doped by simple polymer coating with different dipolar characteristics. Significantly, spontaneous vertical ordering of dipolar pyridine side groups of poly(4-vinylpyridine) at graphene surface can stabilize n-type doping at room-temperature ambient condition. The dipole field also enhances and balances the charge mobility by screening the impurity charge effect from the bottom substrate. We successfully demonstrate ambient stable inverters by integrating p- and n-type graphene transistors, which demonstrated clear voltage inversion with a gain of 0.17 at a 3.3 V input voltage.
