Deep sequencing of microRNAs from hickory reveals an extensive degradation and 30 end modification

The degradation and 30 end modification of plantmicroRNAs (miRNAs) may play crucial roles in regulatingmiRNA function and stability. However, the mechanism asto how the degradation and the modification are processedare still poorly characterized. Here, we report a survey ofmiRNA degradation and 30 modification from two hickoryfloral differentiation stages through deep sequencing. Weconstructed two small RNA (sRNA) libraries from twohickory floral differentiation stages and obtained a largenumber of truncated miRNAs and miRNAs with 30 endmodifications. The presence of so many truncated miRNAssuggests a mechanism degrading through both ends simultaneously. Further analysis reveals that the truncation fromthe 30 end has higher probability than from the 50 end. Singleordouble-nucleotide additions to the 30 end have beenobserved in many families. We found that the addition ofadenine base to the 30 end is the most common event,accounting for more than 50 % of all miRNA 30 end modificationin the two sRNA libraries. Uridine addition is thesecond popular modification. These observations suggestthat the 30 end modification of miRNAs preferentially selectsadenine and uridine in the hickory plant. Furthermore, weobserved that expression of either truncated miRNA orisomiR associates with mature miRNAs. Altogether, ourstudy provides more information regarding the degradationand 30 end modification of miRNAs in plants.
Study on Fuzzy Energy Management Strategy of Parallel Hybrid Vehicle Based on Quantum PSO Algorithm

A fuzzy energy management strategy was designed for the single-axle parallel hybrid electric vehicles, and then the quantitative factor of the fuzzy logic controller was optimized by the quantum PSO algorithm under the Matlab platform based on the equivalent fuel economy. Then a comparison test about the energy management before and after the optimization was carried out based on the secondary development of Advisor which proves that the energy management strategy optimized by the quantum PSO algorithm can improve the SOC of the battery pack by 18% when the VAIL2NREL cycle finished while the power of the vehicles nearly remain the same. What’s more, the optimized strategy can make the engine and the motor works in the high efficient area for most of the time which can improve the recycling rate of the energy and reduce the equivalent fuel consumption effectively.

Shiga toxin 2 (Stx2) is a major virulence factor for enterohemorrhagic Escherichia coli (EHEC), which is encoded by λ lysogenic phage integrated into EHEC chromosome. Stx2A1, A1 subunit of Stx2 toxin has gathered extensive concerns due to its potential of being developed into a vaccine candidate. However, the substantial progress is hampered in part for the lack of a suitable in vitro expression system. Here we report use of the prokaryotic system pET-28a::espA-Stx2A1/BL21 to carry out the fusion expression of Stx2A1 which is linked to E. coli secreted protein A (EspA) at its N-terminus. Under the IPTG induction, EspA- Stx2A1 fusion protein in the form of inclusion body was obtained successfully, whose expression level can reach about 40% of total bacterial protein at 25°C, much higher than that at 37°C. Western blot test suggested the refolded fusion protein is of excellent immuno-reactivity with both monoclonal antibodies, which are specific to EspA and Stx2A1, respectively. Anti-sera from Balb/c mice immunized with the EspA-Stx2A1 fusion protein were found to exhibit strong neutralization activity and protection capability in vitro and in vivo. These data have provided a novel feasible method to produce Stx2A1 in large scale in vitro, which is implicated for the development of multivalent subunit vaccines candidate against EHEC O157:H7 infections.

Quorum sensing is a widespread chemical communication in response to fluctuation of bacterial population density, and has been implicated into bacterial biofilm formation and regulation of expression of virulence factors. The luxS gene product, S-ribosylhomocysteinase, catalizes the last committed step in biosynthetic pathway of autoinducer 2 (AI-2), a signaling molecule for inter-species quorum sensing. We found a luxS homologue in 05ZYH33, an epidemic strain of Streptococcus suis serotype 2 (SS2) in China. A luxS null mutant (ΔluxS) of 05ZYH33 strain was obtained using an approach of homologous recombination. LuxS was determined to be required for AI-2 production in 05ZYH33 strain of S. suis 2. Inactivation of luxS gene led to a wide range of phenotypic changes including thinner capsular walls, increased tolerance to H_2O_2, reduced adherence capacity to epithelial cells, etc. In particular, loss of LuxS impaired dramatically its full virulence of SS2 in experimental model of piglets, and functional complementation restored it nearly to the level of parent strain. Genome-wide transcriptome analyses suggested that some known virulence factors such as CPS are down-regulated in the ΔluxS mutant, which might in part explain virulence attenuation by luxS deletion. Similarly, 29 of 71 genes with different expression level were proposed to be targets candidate regulated by LuxS/AI-2-dependent quorum sensing.

Encarsia formosa Gahan is a solitary endoparasitoid that is commercially reared and released for augmentative biological control of whiteflies including Bemisia tabaci (Gennadius). Bemisia tabaci biotypes B and Q are two most invasive species that greatly reduce crop yields in China by feeding on plant sap and by transmitting Tomato Yellow Leaf Curl Virus (TYLCV). The effects of TYLCV infection of tomato on E. formosa foraging on B. tabaci B and Q are unknown. In Y-tube olfactometer assays in the present study, E. formosa significantly preferred TYLCVinfected tomato plants over TYLCV-free plants. The wasp females also significantly preferred TYLCV-infected tomato plants infested with 3rd-instar nymphs of B. tabaci biotype Q over TYLCV-free plants with biotype Q nymphs. However, no significant differences were observed when B. tabaci biotype B was infested on tomato plants. The oviposition bioassays confirmed that TYLCV infection on tomato plants resulted in the recruitment of parasitoids. These results indicate that TYLCV-infection of tomato increase the foraging of E. formosa on B. tabaci, as differs on the B and Q biotypes.
