        Multi-scale Local Difference Directional Number Pattern for Group-housed Pigs Recognition

        In this paper, a multi-scale local difference directional number (MLDDN) pattern is proposed for pig identification. Firstly, the color images of individual pig are converted into grey images by the most significant bits (MSB) quantization, which makes the grey values have better discrimination. Then, Gabor amplitude and phase responses on different scales are obtained by convoluting the grey images with Gabor masks. Next, by calculating the main difference of local edge directions instead of traditionally edge information, the directional numbers of Gabor amplitude and phase responses are encoded. Finally, the block histograms of the encoded images are concatenated on each scale, and the maximum pooling is adopted on different scales to avoid the high feature dimension. Experimental results on two pigsties show that MLDDN impressively outperforms the other widely used local descriptors.

        Concrete pavement monitoring with PPP-BOTDA distributed strain and crack sensors

        In this study, the feasibility of using telecommunication single-mode optical fiber (SMF) as a distributed fiber optic strain and crack sensor was evaluated in concrete pavement monitoring. Tensile tests on various sensors indicated that the SMF-28e+ fiber revealed linear elastic behavior to rupture at approximately 26 N load and 2.6% strain. Six full-scale concrete panels were prepared and tested under truck and three-point loads to quantify the performance of sensors with pulse pre-pump Brillouin optical time domain analysis (PPP-BOTDA). The sensors were protected by precast mortar from brutal action during concrete casting. Once air-cured for 2 hours after initial setting, half a mortar cylinder of 12 mm in diameter ensured that the protected sensors remained functional during and after concrete casting. The strains measured from PPP-BOTDA with a sensitivity coefficient of 5.43 10-5 GHz/ were validated locally by commercial fiber Bragg grating (FBG) sensors. Unlike the point FBG sensors, the distributed PPP-BOTDA sensors can be utilized to effectively locate multiple cracks. Depending on their layout, the distributed sensors can provide one- or two-dimensional strain fields in pavement panels. The width of both micro and major cracks can be linearly related to the peak strain directly measured with the distributed fiber optic sensor.

        The Cancer/Testis Antigen CT45A1 Promotes Transcription of Oncogenic Sulfatase-2 Gene in Breast Cancer Cells and Is Sensible Targets for Cancer Therapy

        Purpose: Invasive breast carcinomas (BRCAs) are highly lethal. The molecular mechanisms underlying progression of invasive BRCAs are unclear, and effective therapies are highly desired. The cancer-testis antigen CT45A1 promotes overexpression of pro-metastatic sulfatase-2 (SULF2) and breast cancer metastasis to the lungs, but its mechanisms are largely unknown. In this study, we aimed to elucidate the mechanism of CT45A1-induced SULF2 overexpression and provide evidence for targeting CT45A1 and SULF2 for breast cancer therapy. Methods: The effect of CT45A1 on SULF2 expression was assessed using reverse transcription polymerase chain reaction and western blot. The mechanism of CT45A1-induced SULF2 gene transcription was studied using protein-DNA binding assay and a luciferase activity reporter system. The interaction between CT45A1 and SP1 proteins was assessed using immunoprecipitation and western blot. Additionally, the suppression of breast cancer cell motility by SP1 and SULF2 inhibitors was measured using cell migration and invasion assays. Results: CT45A1 and SULF2 are aberrantly overexpressed in patients with BRCA; importantly, overexpression of CT45A1 is closely associated with poor prognosis. Mechanistically, gene promoter demethylation results in overexpression of both CT45A1 and SULF2. CT45A1 binds directly to the core sequence GCCCCC in the promoter region of SULF2 gene and activates the promoter. Additionally, CT45A1 interacts with the oncogenic master transcription factor SP1 to drive SULF2 gene transcription. Interestingly, SP1 and SULF2 inhibitors suppress breast cancer cell migration, invasion, and tumorigenicity. Conclusion: Overexpression of CT45A1 is associated with poor prognosis in patients with BRCA. CT45A1 promotes SULF2 overexpression by activating the promoter and interacting with SP1. Additionally, SP1 and SULF2 inhibitors suppress breast cancer cell migration, invasion, and tumorigenesis. Our findings provide new insight into the mechanisms of breast cancer metastasis and highlight CT45A1 and SULF2 as sensible targets for developing novel therapeutics against metastatic breast cancer.

        Enhancement of Immunotherapeutic Effects of HPV16E7 on Cervical Cancer by Fusion with CTLA4 Extracellular Region

        Cervical cancer is caused by infection by high-risk human papillomavirus (HPV), especially HPV16. Limitations in current treatments of cervical cancers call for the development of new and improved immunotherapies. This study aims at investigating the efficacy of a novel vaccine consisting of modified HPV 16E7 fused with human cytotoxic T-lymphocyte antigen 4 (CTLA4). The regions in HPV16 E7 gene associated with its transformation and CTL-enhanced response were modified; the resultant HPV16mE7 was fused with extracellular region of CTLA4 to generate HPVm16E7-eCTLA4 fusion protein. Binding of this fusion protein to B7 molecules expressed on antigen presenting-cells (APCs) was demonstrated. C57BL/6 (H-2b) mice immunized with low dose of the fusion protein (10 μg) produced higher titer antibody and stronger specific CTL response, and expressed higher levels of IFN-γ and IL-12, compared with those immunized with HPVm16E7 only or admixture of HPVm16E7 and CTLA4, or PBS; and were protected from lethal dose tumor challenge. Tumor growth was retarded and survival prolonged in mouse models with the fusion protein treatment. Our results demonstrate that fusion of HPV16 E7 with eCTLA4 targeting APCs resulted in enhanced immunity, and that this fusion protein may be useful for improving the efficacy of immunotherapeutic treatments of cervical cancer and other HPV16 infection-associated tumors.
