      • Nasal-type NK/T-cell lymphomas are more frequently T rather than NK lineage based on T-cell receptor gene, RNA, and protein studies: lineage does not predict clinical behavior

        <P>Extranodal natural killer (NK)/T-cell lymphoma (ENKTL), nasal type, comprises NK or cytotoxic T cells. We evaluated the clinical impact of cell type and the usefulness of T-cell receptor (TCR) gene transcripts in distinguishing cell lineage. One hundred and eight cases of ENKTL were analyzed for TCR gene rearrangements using the BIOMED-2 protocol and for TCR gene expression using immunohistochemistry for TCR-beta F1 and TCR-c gamma M1, and RNA in situ hybridization for TCR gene transcripts. Prognostic factors were analyzed. Among the 108 cases, 44 were monoclonal for a TCR rearrangement (40%) while 64 (60%) were undefinable. The monoclonal cases expressed TCR-beta F1 in 14 out of 40 cases (35%) and TCR-c gamma M1 in 1 out of 44 cases (2%). The 64 undetermined cases expressed TCR-beta F1 in 15 cases (23%) and TCR-c gamma M1 in 1 (2%). Thirteen of 40 TCR-beta constant gene transcript-positive cases (33%) expressed TCR-beta F1 and one of nine TCR-gamma constant gene transcript-positive cases (11%) expressed TCR-c gamma M1. TCR gene transcripts were not useful in the distinction of cell lineages. TCR gene transcripts were positive in ENKTLs as well as in normal B cells and aggressive NK-cell leukemia. Based on gene rearrangements and immunohistochemistry for TCR, there were 60 T-cell type cases (56%), 32 NK-cell type cases (30%), and 16 cases with an undetermined cell type (14%). TCR protein was expressed in 30/60 T-ENKTLs (50%) in a variable fraction of tumor cells. There were no significant differences in clinical findings or overall patient survival between T-or NK-cell types of ENKTL, although those with a T-cell type tended to show a better prognosis for those with localized nasal lymphomas. Univariate and multivariate analysis showed that a non-nasal ENKTL, age 460 years, high level of lactate dehydrogenase, bone marrow involvement, and the absence of radiotherapy were independent prognostic factors.</P>

        The critical discriminatory event in the activation of T lymphocytes bearing αβT cell receptors (TCRs) is their interaction with a molecular complex consisting of a peptide bound to a major histocompatibility complex (MHC)-encoded classⅠor class Ⅱ molecule on the surface of an antigen-presenting cell. The kinetics of binding were measured of a purified TCR to molecular complexes of a purified soluble analog of the murine MHC classⅠ molecule H-2L^d (sH-2L^d) and a synthetic octamer peptide p2CL in a direct, real-time assay based on surface plasmon resonance. The kinetic dissociation rate of the MHC-peptide complex from the TCR was rapid (2.6×10^-2) second^-1, corresponding to a half-time for dissociation of approximately 27 seconds), and the kinetic association rate was 2.1×10^ 5 M^-1 second^-1. The equilibrium constant for dissociation was approximately 10^-7M These values indicate that TCRs must interact with a multivalent array of MHC-peptide complexes to trigger T cell signaling.

      • Petrophysical property modification in a volcanic sequence on the Naturaliste Plateau, offshore SW Australia (IODP Site U1513): Implications of volcanic stratigraphy, paleoenvironment, and hydrothermal activity

        This study investigates the linkages between physical properties, primary lithology, and alteration processes of a volcanic sequence at Site U1513 on the Naturaliste Plateau, offshore southwest Australia. The volcanic eruption and emplacement occurred in subaerial to shallow water setting on the rifted continental margin during the Early Cretaceous. The volcanic sequence consists of alternating basalt flows and volcaniclastic deposits, intruded by multiple dolerite dikes. We compile a variety of petrophysical data and correlate these with lithologic, mineralogical, and geochemical analyses (e.g., XRD, XRF), acquired during and after the International Ocean Discovery Program (IODP) Expedition 369. Variable alteration intensities from fresh-slight to strong are evaluated using Chemical Index of Alteration, alteration mineralogy, and textural changes. Intervals of fresh-slightly altered dikes exhibit low porosity (~13%) and high values of bulk density (~2.7 g/cm³), P-wave velocity (~4900 m/s) and thermal conductivity (~1.7 W/(m·K)). The increase of alteration intensity corresponds to decreases in bulk density to ~2 g/cm³, P-wave velocity to ~2000 m/s, thermal conductivity to ~1.2 W/(m·K) and an increase in porosity up to 50%. Natural Gamma Ray and magnetic susceptibility vary downhole with rock composition and at lithologic boundaries. The distinct physical variations exhibit a good correlation with primary lithologic characteristics as well as secondary mineralogical and textural changes attributed to alteration processes by subaerial exposure, seawater interaction and hydrothermal activities. Synthesis models of petrophysical variation with alteration intensity are provided for future studies. Although limitations and uncertainties are introduced due to differences in primary lithology, alteration type, and petrophysical methodology, there is a reasonable applicability of our results to rapidly characterize the alteration intensity and volcanic stratigraphy in volcanic sequences. Our results can be used to develop strategies better for exploration, drilling, and geophysical research of volcanic terrains.

        부산시 기장군 연안의 해조상 및 군집 특성

        기장 연안 해조류의 해조상 및 군집 구조를 분석하기 위해서 2006년 8월과 2009년 8월에 연구를 수행하였다. 해조류는 녹조류 6종, 갈조류 10종, 홍조류 38종 등 총 54종의 서식이 확인되었다. 이 중 35종의 해조류가 각각의 조사 시기에 공통적으로 출현하였다. 평균 현존량은 2006년에 $616.0\;g\;m^{-2}$에서 $1,462.4\;g\;m^{-2}$였고, 2009년에는 $354.8\;g\;m^{-2}$에서 $965.6\;g\;m^{-2}$이었다. 현존량이 가장 높은 지역은 문동이었고, 가장 낮은 곳은 서암(2006년)과 동백(2009년)이었다. 출현 해조류를 6개의 기능형으로 구분하면, 성긴분지형이 58.7%와 58.1%, 다육질형이 10.9%와 11.6%, 사상형이 13.0%와 9.3%, 각상형이 6.5%와 9.3%, 엽상형이 6.5%와 7.0%, 유절산호말형이 4.3%와 4.7%로 나타났다. 생태지수인 R/P, C/P, (R+C)/P 값은 2006년에 4.00, 0.75, 4.75와 2009년에 5.17, 1.00, 6.17로 나타났다. 2006년과 2009년의 자료를 분석했을 때, 기장 연안의 해조류 출현 종 수와 현존량은 유사하였다. 이러한 자료로 볼 때, 기장 연안에서 시행된 소규모 공사 전, 후인 2006년과 2009년의 해조류 종 다양성에는 변화가 나타나지 않은 것으로 분석되었다. Marine algal flora and community structure were investigated at four sites in Kijang area on the southern east coast of Korea in August 2006 and August 2009, respectively. A total of 54 seaweeds including 6 green, 10 brown, 38 red were collected and identified. Of 54 seaweeds, 35 species were found throughout the survey period. Mean biomass in wet weight was from $616.0\;g\;m^{-2}$ to $1,462.4\;g\;m^{-2}$2 in 2006, and $354.8\;g\;m^{-2}$ to $965.6\;g\;m^{-2}$ in 2009. Maximum biomass was recorded at Mundong site, and minimum was recorded at Seoam (2006) and Dongbaek (2009) sites. The flora investigated (2006, 2009) could be classified into six functional groups such as coarsely branched form (58.7%, 58.1%), thick leathery form (10.9%, 11.6%), filamentous form (13.0%, 9.3%), crustose form (6.5%, 9.3%), sheet form (6.5%, 7.0%) and jointed calcareous form (4.3%, 4.7%) during survey period. The R/P, C/P and (R+C)/P values reflecting flora characteristics were 4.00, 0.75 and 4.75 at 2006, and 5.17, 1.00 and 6.17 at 2009, respectively. Therefore, the number of marine algae species and biomass in Kijang area were similar when they were comparing with the previous data. It suggest that any changes of seaweed diversity have not been observed in Kijang coastal area before and after the anthropogenic construction between 2006 and 2009.

        Highly Porous Silica Nanoaerogels for Ultrafast Nonlinear Optical Applications

        <P>Highly porous silica nanoaerogels with low apparent density of ~0.1 g/cm3 and ~0.07 g/cm3 were synthesized through two-step sol-gel processing and low temperature supercritical fluid drying. The nonlinear refraction (γ) of silica nanoaerogels was estimated to be ~ -3.4 x 10-16 m2/W for ~0.1 g/cm3 and ~0.07 g/cm3 apparent densities with a signal-beam femtosecond z-scan spectroscopy. The third-order nonlinear refraction coefficient of nanostructure silica nanoaerogels was almost four orders larger than that of bulk silica materials. The large nonlinearrefraction with high nonlinear figure of merit (γ/βλ, β~2×10-10 m/W for 0.07 g/cm3 apparent density, β~6×10-10 m/W for 0.1 g/cm3 apparent density, λ~0.775 µm) is an ideal optical property for nonlinear applications of homeland security, battlefield enhancement, and industrial uses.</P>

        본 증례는 산모의 혈청 속에 존재하는 항-C+e 항체 중 IgG type의 항-C 항체만 태반을 통과하여 발생된 신생아 용혈성 질환으로 산모 혈청 속에 존재하는 항-C의 항체역가는 1:256, 신생아는 1:32를 보였으며 IgM type의 항-e는 산모의 혈청에서만 1:2048의 높은 항체역가를 보였다. 따라서 신생아의 혈청 혈액형 검사가 AHG phase에서 A 혹은 B 혈구에 응집 반응이 일어났다면 동종항체에 의한 응집 반응인지 IgG type의 항-A 혹은 B 항체에 의한 것인지를 구별해야 할 것이며, 이를 위해서는 산모 및 신생아의 항체 선별검사 및 신생아의 ABO 혈구형 검사와 혈청 혈액형 검사가 AHG phase까지 반드시 실시되어야 할 것이다. We describe a case of hemolytic disease of newborn due to anti-C derived from mother who have anti-C+e The titer of anti-C was 1:256 in mother`s serum and 1:32 in serum of nenonate suffering hemolytic disease of newborn. The titer of anti-e was 1:2048 found only in mother`s serum. Because it was IgM antibody, it could not cross the placenta. In order to confirm causative antibody and make a plan for safe transfusion in case of hemolytic disease of newborn, we must investigate the blood group antibody status of both mother and neonate including immunoglobulin types of antibody such as IgG and IgM

        수혈 시 실시하는 교차시험 및 불규칙 항체선별 검사에서 환자가 불규칙 항체를 가지고 있는 경우 항체의 종류를 정확히 동정하여 환자에게 가장 적합한 혈액을 공급하는 것은 중요한 일이다. 이러한 목적으로 국민건강보험공단 일산병원 혈액은행에서 개원 후 지난 1년간 검출된 불규칙 항체에 대해 각 항체의 종류 및 빈도 등에 대해 분석하여 보았다. Gel card를 이용한 항체선별 검사에서 항-E 항체가 6건, 항- E 항체 및 항-c항체까지 검출된 경우가 4건, 항-Ce 항체가 2건, 항체선별 검사에서 양성이나 특이 항체로 검출이 되지 않은 복합 항체의 경우가 4건 등이었다. 불규칙 항체의 빈도(prevalence)는 0.64%이 었으며 대부분이 Rh-Hr 계열의 항체이며. 특히 Rh-Hr 표현형이 DCe인 경우의 환자에서 항-E항체 및 항-c 항체가 동시에 검출된 경우가 전체의 40%로써 국내에서 보고된 항체 검출률 보다 높은 수치였다. 본원에서 사용하는 불규칙 항체 검사법은 gel card를 이용 37도에서 시행하고 동정검사도 동일하여 이제까지 보고들과는 달리 Lewis 계열의 항체나 한냉항체는 드물었는데 이러한 항체는 그 임상적인 의미를 무시할 수 있는 항체이므로 본원과 같이 gel card를 사용 시 항체선별검사 및 동정검사에 효율적인 업무수행을 할 수 있으리라 생각된다. Irregular antibodies is all kind of antibodies except regular antibodies like anti-A or anti-B. The detection of irregular antibodies is very important to the patients with this antibodies for transfusion. Especially, Rh-Hr antibodies(anti-E, anti-c, anti-C, anti-e etc) have clinical significance in developing a delayed hemolytic transfusion reaction and hemolytic disease of the newborn especially. We reviewed identified irregular antibody in National Health Insurance Corporation Ilsan Hospital for one year(2000. 3-2001. 2). Antibody screening test using gel card identified irregular antibodies in 16 of 2482 cases. The frequency of irregular antibodies : anti-E, 0.24% anti-E+c, 0.16%, anti-Ce 2, 0.08%, specificity not determined, 0.16% in our study. Cold antibodies was not detected in gel card but identified in cross matching of saline phase. We use enzyme technique to detect some antibody like anti-c, anti-e, which could be missed in LISS Coombs phase. We detected 4 cases of anti-E+c and 6 cases of anti-E. The R1R1 patients(DCe) with anti-E were transfused by E+c antigen negative blood to prevent presentation of anti-c.

      • Heterochiral Assembly of Amphiphilic Peptides Inside the Mitochondria for Supramolecular Cancer Therapeutics

        <P>Self-assembly of peptides containing both <SMALL>L</SMALL>- and <SMALL>D</SMALL>-isomers often results in nanostructures with enhanced properties compared to their enantiomeric analogues, such as faster kinetics of formation, higher mechanical strength, and enzymatic stability. However, occurrence and consequences of the heterochiral assembly in the cellular microenvironment are unknown. In this study, we monitored heterochiral assembly of amphiphilic peptides inside the cell, specifically mitochondria of cancer cells, resulting in nanostructures with refined morphological and biological properties owing to the superior interaction between the backbones of opposite chirality. We have designed a mitochondria penetrating tripeptide containing a diphenyl alanine building unit, named as Mito-FF due to their mitochondria targeting ability. The short peptide amphiphile, Mito-FF co-assembled with its mirror pair, Mito-ff, induced superfibrils of around 100 nm in diameter and 0.5-1 μm in length, while enantiomers formed only narrow fibers of 10 nm in diameter. The co-administration of Mito-FF and Mito-ff in the cell induced drastic mitochondrial disruption both <I>in vitro</I> and <I>in vivo</I>. The experimental and theoretical analyses revealed that pyrene capping played a major role in inducing superfibril morphology upon the co-assembly of racemic peptides. This work shows the impact of chirality control over the peptide self-assembly inside the biological system, thus showing a potent strategy for fabricating promising peptide biomaterials by considering chirality as a design modality.</P> [FIG OMISSION]</BR>

        A medium duty hydrogen-natural gas fueled engine using hydrogen enriched CNG fuel can obtain stable operation at ultra lean condition and reduce emissions extremely. Reduction of CO₂in its engine is the one of most benefit. In this study, compression ratio were investigated including performance of this engine. As results, it was found that phenomenon of pressure oscillation it means occurring knock was increased due to large specific volume of hydrogen when R_(dH2) was increased. Even if same compression ratio, pressure oscillation was remarkable according to increasing R_(dH2).
