      • Oridonin Suppresses Proliferation of Human Ovarian Cancer Cells via Blockage of mTOR Signaling

        Oridonin, an ent-kaurane diterpenoid compound isolated from the traditional Chinese herb Rabdosia rubescens, has shown various pharmacological and physiological effects such as anti-tumor, anti-bacterial, and anti-inflammatory properties. However, the effect of oridonin on human ovarian cancer cell lines has not been determined. In this study, we demonstrated that oridonin inhibited ovarian cancer cell proliferation, migration and invasion in a dose-dependent manner. Furthermore, we showed oridonin inhibited tumor growth of ovarian cancer cells (SKOV3) in vivo. We then assessed mechanisms and found that oridonin specifically abrogated the phosphorylation/activation of mTOR signaling. In summary, our results indicate that oridonin is a potential inhibitor of ovarian cancer by blocking the mTOR signaling pathway.

      • Establishment of Paclitaxel-resistant Breast Cancer Cell Line and Nude Mice Models, and Underlying Multidrug Resistance Mechanisms in Vitro and in Vivo

        Background: Breast cancer is a common malignant tumor which affects health of women and multidrug resistance (MDR) is one of the main factors leading to failure of chemotherapy. This study was conducted to establish paclitaxel-resistant breast cancer cell line and nude mice models to explore underlying mechanisms of MDR. Methods: The breast cancer drug-sensitive cell line MCF-7 (MCF-7/S) was exposed in stepwise escalating paclitaxel (TAX) to induce a resistant cell line MCF-7/TAX. Cell sensitivity to drugs and growth curves were measured by MTT assay. Changes of cell morphology and ultrastructure were examined by optical and electron microscopy. The cell cycle distribution was determined by flow cytometry. Furthermore, expression of proteins related to breast cancer occurrence and MDR was tested by immunocytochemistry. In Vivo, nude mice were injected with MCF-7/S and MCF-7/TAX cells and weights and tumor sizes were observed after paclitaxel treatment. In addition, proteins involved breast cancer and MDR were detected by immunohistochemistry. Results: Compared to MCF-7/S, MCF-7/TAX cells had a higher resistance to paclitaxel, cross-resistance and prolonged doubling time. Moreover, MCF-7/TAX showed obvious alterations of ultrastructure. Estrogen receptor (ER) expression was low in drug resistant cells and tumors while expression of human epidermal growth factor receptor 2 (HER2) and Ki-67 was up-regulated. P-glycoprotein (P-gp), lung resistance-related protein (LRP) and glutathione-S-transferase-${\pi}$ (GST-${\pi}$) involved in the MDR phenotype of resistant cells and tumors were all overexpressed. Conclusion: The underlying MDR mechanism of breast cancer may involve increased expression of P-gp, LRP and GST-${\pi}$.

      • The Feasibility Analysis of Single Curved Screw Drill through the Mouth of the Well Casing

        With the development of drilling technology and the needs of the larger Bulit-up, the screw drill with larger structure corner is used more widely in drilling field. Because the existence of single curved screw drill tool with larger structure corner, it have larger friction during the bend PDM in the process of internal casing, so we often need to use the wellhead adjuvant into the well. In order to understand the single bent screw drill into the state in the wellhead diameter of casing, and how the angle of screw structure impact the feasibility, This paper analysis the stress of the single curved screw drilling tools in the well, and the condition it can safely through the mouth of the well conditions, establish the single bend PDM in the calculation wellhead down model. and the finite element model is established combined with the specific parameters of a well which is in the western of SiChuan depression. Using ansys analysis a particular casing inside diameter, under wellhead vertical segment into different structure when the turn of the screw drilling tools down into characteristics and stress distribution characteristics, it is concluded how large of the structure corners and how depth that the bend PDM can rely on gravity well down into the inside diameter of 250.19 mm casing, concluded the relationship between reaction force, friction force, and maximum stress with the well depth and the change of structure corners from 1.5°to 2.75°,provide a basis for drilling site selection of the appropriate combinations of screw drill and how to regulate the radial force of the device while into the well.

        Deformation Feature of Sheet Metals During Inclined Hole-Flanging by Two-Point Incremental Forming

        Incremental hole-flanging is an applicable extension of incremental sheet forming (ISF), but the knowledge on the deformation of metallic sheets during the course is lack, especially when the axes of the flanged necks are not perpendicular to the sheet plane. The current study used Al 6061 and SUS 304 stainless steel sheets with the thickness of 1 mm and 0.3 mm to examine the inclined hole-flanging processes on the flat plates by two-point ISF processes. It was found that, although as a whole the thickness variation is thinning, the decreasing degrees within upper and lower domain are not equal, and the difference increases with the inclination angle. Crack, buckling and local deformity are typical defects found in the experiments. The maximum springback increases with tool diameter, inclination angle and feeding pitch. Due to the material properties and a thinner thickness, local deformity is easier to take place on SUS 304 sheets during forming. Utilizing the tool feeding path along the oblique plane can obtain even force and consequently, more uniform deformation. Moreover, the equations for preliminarily determining the dimension of precut holes were derived according to the radial developed length of the flanged neck.

        Genome-wide association analysis of nine reproduction and morphological traits in three goat breeds from Southern China

        Objective: This study aimed to investigate the significant single nucleotide polymorphisms (SNPs) and genes associated with nine reproduction and morphological traits in three breed populations of Chinese goats. Methods: The genome-wide association of nine reproduction and morphological traits (litter size, nipple number, wattle, skin color, coat color, black dorsal line, beard, beard length, and hind leg hair) were analyzed in three Chinese native goat breeds (n = 336) using an Illumina Goat SNP50 Beadchip. Results: A total of 17 genome-wide or chromosome-wide significant SNPs associated with one reproduction trait (litter size) and six morphological traits (wattle, coat color, black dorsal line, beard, beard length, and hind leg hair) were identified in three Chinese native goat breeds, and the candidate genes were annotated. The significant SNPs and corresponding putative candidate genes for each trait are as follows: two SNPs located on chromosomes 6 (CSN3) and 24 (TCF4) for litter size trait; two SNPs located on chromosome 9 (KATNA1) and 1 (UBASH3A) for wattle trait; three SNPs located on chromosome 26 (SORCS3), 24 (DYM), and 20 (PDE4D) for coat color trait; two SNPs located on chromosome 18 (TCF25) and 15 (CLMP) for black dorsal line trait; four SNPs located on chromosome 8, 2 (PAX3), 5 (PIK3C2G), and 28 (PLA2G12B and OIT3) for beard trait; one SNP located on chromosome 18 (KCNG4) for beard length trait; three SNPs located on chromosome 17 (GLRB and GRIA2), 28 (PGBD5), and 4 for hind leg hair trait. In contrast, there were no SNPs identified for nipple number and skin color. Conclusion: The significant SNPs or genes identified in this study provided novel insights into the genetic mechanism underlying important reproduction and morphological traits of three local goat breeds in Southern China as well as further potential applications for breeding goats.

        Combined Effects of Blood Plasma Powder, Agar, and Microbial Transglutaminase on Physicochemical and Textural Properties of Pork Muscle Gels

        The effects of blood plasma (BP, 0-2.5%), agar (0-1.0%), and microbial transglutaminase (MTG, 0-0.5%)on water-binding capacity, color (L*, a*, and b* values)and textural properties of pork muscle gels (PMG) were investigated with response surface methodology. The potential interaction among these ingredients was evaluated by DSC, SEM, and FT-IR. The results shown that the addition of BP into PMG could result in decreased cooking loss (CL) and L* (p<0.01), as well as increased a*, springiness,and chewiness of the gels (p<0.05), while the treatment with MTG led to increased CL (p<0.01). Agar could obviously improve CL of PMG, but lead to inferior springiness and cohesiveness (p<0.05). Moreover, the synergism among muscle proteins, BP, MTG, and agar implied that there were some kinds of interactions among these ingredients, thus influencing the properties of PMG.

      • Biological and Chemical Microsensors for Health Care and Environment Monitoring

        This paper presents several biological and chemical microsensors developed based on micro and nano technologies for the applications of health care and environment monitoring, including micro amperometric biosensor, micro potentiometric immunosensor and micro gas sensor and so on. The electroactivities of the micro amperometric biosensor with three-dimensional microelectrodes was enhanced obviously with the help of the nano- and microstructures. By introducing nanoparticles with seed mediated growth method, the sensitivity of the micro potentiometric immunosensor for %HbA1c has been increased. A micro pellistor with nanoporous alumina catalyst carrier have been constructed for detecting combustible gas below the lower explosive limit, which shows advantages of low power comsumption, good thermal performance and easy fabrication.

        Prognostic Perspectives of STING and PD-L1 Expression and Correlation with the Prognosis of Epstein-Barr Virus-Associated Gastric Cancers

        Background/Aims: Epstein-Barr virus-associated gastric cancers (EBVaGCs) have unique molecular and clinicopathological characteristics. The cyclic GMP-AMP synthase-stimulator of interferon genes (STING) pathway is recently recognized as the critical innate immunity against pathogens and tumors. STING is also a master regulator in the cancer-immunity cycle and targeting STING could synergize with existing immune-checkpoint therapies. However, the role of STING in GC, especially in EBVaGC, and its correlation with programmed death-ligand 1 (PD-L1) remain largely unclear. Methods: We collected 78 cases of EBVaGCs and 210 cases of EBV-negative GC (EBVnGC) from a total of 1,443 cases of GC analyzed by EBV-encoded small RNA in situ hybridization. We investigated STING and PD-L1 expression and their concomitant prognostic value in EBVaGCs and EBVnGCs using tissue microarray and immunohistochemistry. The effects of STING and PD-L1 expression on the overall survival of patients with EBVaGC or EBVnGC were assessed by univariate and multivariate analysis. Results: We found that both STING and PD-L1 exhibited significantly higher expression in the EBVaGCs than that in the EBVnGCs. The expression of STING was positively correlated with that of PD-L1 in EBVaGCs. Simultaneous negative expression of STING and PD-L1, and positive expression of STING were independent prognostic risk factors for EBVaGC and EBVnGC, respectively. Conclusions: This is the first prognostic retrospective study of STING and PD-L1 expression and the prognosis among EBVaGC and EBVnGC. The expression and prognostic value of STING and PD-L1 are different in the two types of GCs. STING and PD-L1 are promising prognostic biomarkers and therapeutic targets for EBVaGC and EBVnGC.
