        Development and Evaluation of Protected Fat in Wheat Straw Based Total Mixed Ration

        Ca salt of soybean oil (PSO) and that of mustard oil plus mahua oil (PMOMO) (50:50) were prepared using double decomposition method, and further tested for their fatty acid composition and degree of saponification. Furthermore, the different levels of protected fat of PSO and PMOMO were evaluated in wheat straw based total mixed ration (TMR) in vitro. Results indicated that capric, lauric, myristic, palmitic, steric, oleic, linoleic, leinolenic acids were traces, traces, traces, 10.00, 2.00, 25.00, 58.50, 5.0% in PSO while the corresponding values in PMOMO were 1.08, 0.28, 0.45, 16.9, 12.95, 44.38, 17.46 and 6.50%, respectively. The degree of saponification of both protected fat supplements was more than 80%. Six treatment combinations were tested I.e., blank without feed and fat supplement (T1); control diet with out fat supplement (T2); control diet plus bypass fat supplement (PSO) so that diet contain 5% fat (T3); control diet plus bypass fat supplement (PSO) so that diet contain 7.5% fat (T4); two more diets viz. T5 and T6 were formulated using bypass fat supplement from PMOMO containing 5 and 7.5% fat respectively. TMR was prepared using 50% concentrate mixture and 50% wheat straw. Result indicated that TVFA, $NH_3-N$,TCA-N, total-N and total gas production were increased in treatment diets at 7.5% level of supplementation, however, fermentation pattern remain similar at 5.0% level of supplementation with respect to control diet. Nevertheless, IVDMD and IVOMD values remained unchanged, rather non-significant at both fat levels and with the both fat sources. On the basis of results it was concluded that Ca-salt of Soybean oil or Mustard plus Mahua oil did not show any negative effect either on digestibility or on microbial protein synthesis in rumen, hence the dietary fat upto 7.5% level in total mixed ration based on wheat straw, could be safely used without any adverse effect on rumen fermentation.

        Synergistic Effect of Urea and Lime Treatment of Wheat Straw on Chemical Composition, In Sacco and In Vitro Digestibility

        Chopped wheat straw (0.5-1.5 cm) was subjected to different treatment combinations in a $5{\times}4$ factorial arrangement involving the five levels of urea (0, 2, 3, 4 and 5%, w/w) and four levels of lime (0, 2, 4 and 6%, w/w) at 50% moisture and kept for 3 wk reaction period at about $35{^{\circ}C}$ in laboratory. Treated wheat straw samples were analyzed to study the associative effect of urea and lime on chemical composition, in sacco and in vitro digestibilities. Results showed that cell wall constituents (CWC) solubilized significantly (p<0.01) due to urea and lime treatment on one hand and substantially increase the crude protein (CP) on the other in wheat straw. The main effect on synergism of both chemicals was noticed on organic matter (OM), neutral detergent fibre (NDF), hemicellulose (HC), acid detergent lignin (ADL) and silica by solubilising their contents as a result of considerable increase in cell contents in treated wheat straw. The respective decreases were 5.45, 13.0, 37.23, 44.95 and 26.16% in different treatment combinations. The most interesting feature of the treatment was evident by increase in ash content on each level of lime application. CP content increase up to 12.78% due to urea treatment in comparison with untreated wheat straw (2.56%). The effect of solubilization of structural carbohydrates and increased crude protein due to synergistic effect of urea and lime were clearly seen on improved digestibility of OM and DM. The increase in ISOMD, ISDMD, and IVDMD were 21.67, 21.67, 16.24, and 17.5 units. The increase in digestibility were relative to additions of both chemicals and digestibility values increased with increasing levels of urea plus lime concentration in different treatment combination. The maximum improvement was noticed at 4% urea and 4% lime levels at 50% moisture for 3 wk reaction period in treated wheat straw.

        CHILD LABOUR, HUMAN CAPITAL ACCUMULATION AND FOREIGN AID

        In a three period overlapping generations model with child labour, exogenous increases in child health endowments increase child labour and fertility. On the other hand, cash transfer and compulsory education funded through foreign aid have a negative effect on child labour. Foreign aid has a positive effects on fertility if cash transfers for child support are unconditional, and no effect if transfers are conditional on time spent in school. This result supports conditional cash transfer programs like the Bolsa Familia.

        Methyl Coenzyme M Reductase (mcrA) Gene Based Phylogenetic Analysis of Methanogens Population in Murrah Buffaloes (Bubalus bubalis)

        The aim of the present study was to decipher the diversity of methanogens in rumen of Murrah buffaloes so that effective strategies can be made in order to mitigate methane emission from these methanogens. In the present study diversity of rumen methanogens in Murrah buffaloes (Bubalus bubalis) from North India was evaluated by using mcr-A gene library obtained from the pooled PCR product from four animals and by using MEGA4 software. A total of 104 clones were examined, revealing 26 different mcr-A gene sequences or phylotypes. Of the 26 phylotypes, 16 (64 of 104 clones) were less than 97% similar to any of the cultured strain of methanogens. Seven clone sequences were clustered with Methanomicrobium mobile and three clone sequences were clustered with Methanobrevibacter gottschalkii during the phylogenetic analysis. Uncultured group of methanogens comes out to be the major component of the methanogens community structure in Murrah buffaloes. Methanomicrobium phylotype comes out to be major phylotype among cultured methanogens followed by Methanobrevibacter phylotype. These results help in making effective strategies to check the growth of dominant methanogenic communities in the rumen of this animal which in turn help in the reduction of methane emission in the environment and ultimately helps us in fighting with the problem of global warming.

        Perioperative Epirubicin, Oxaliplatin, and Capecitabine Chemotherapy in Locally Advanced Gastric Cancer: Safety and Feasibility in an Interim Survival Analysis

        Purpose: Perioperative chemotherapy improves survival outcomes in locally advanced (LA) gastric cancer. Materials and Methods: We retrospectively analyzed patients with LA gastric cancer who were offered perioperative chemotherapy consisting of epirubicin, oxaliplatin, and capecitabine (EOX) from May 2013 to December 2015 at Tata Memorial Hospital in Mumbai. Results: Among the 268 consecutive patients in our study, 260 patients (97.0%) completed neoadjuvant chemotherapy, 200 patients (74.6%) underwent D2 lymphadenectomy, and 178 patients (66.4%) completed adjuvant chemotherapy. The median follow-up period was 17 months. For the entire cohort, the median overall survival (OS), 3-year OS rate, median progression-free survival (PFS), and 3-year PFS rate were 37 months, 64.4%, 31 months, and 40%, respectively. PFS and OS were significantly inferior in patients who presented with features of obstruction than in those who did not (P=0.0001). There was no difference in survival with respect to tumor histology (well to moderately differentiated vs. poorly differentiated, signet ring vs. non-signet ring histology) or location (proximal vs. distal). Survival was prolonged in patients with an early pathological T stage and a pathological node-negative status. In a multivariate analysis, postoperative pathological nodal status and gastric outlet obstruction on presentation significantly correlated with survival. Conclusions: EOX chemotherapy with curative resection and D2 lymphadenectomy is a suggested alternative to the existing perioperative regimens. The acceptable postoperative complication rate and relatively high resections, chemotherapy completion, and survival rates obtained in this study require further evaluation and validation in a clinical trial.

        Effect of Dietary Feeding Regimens on Urea and Protein Concentration of Milk in Murrah Buffaloes

        The present study was planned to examine the effect of different feeding regimens on milk urea concentration and milk protein concentration. The objectives are to describe the diurnal variations of milk urea (MU) concentration and to predict plasma urea (PU) concentration from MU concentration. Six lactating Murrah buffaloes were distributed in two groups and were fed two different diets in a crossover design. The diets consisted of leguminous crops as diet 1 (berseem (Trifolium alexandrinum)+concentrate mixture 1+wheat straw)) and non-leguminous crops as diet 2 (oats (Avana sativa)+concentrate mixture 2+wheat straw). All the diets were isocaloric and isonitrogenous. Each diet was fed to the animals for a period of 28 days, followed by a 10 day gap to obviate the carry over effect of the previous diet and then a switch over to the other diet. Digestibility trials were conducted on the last 7 days of each feeding period. Milk samples were collected on day 3, 7, 10, 14, 17, 21, 24 and 28 of the feeding period and blood samples were collected on the same day at morning within 30 minutes after morning milking. The average milk urea (MU) values (mg/dl) differed significantly (p<0.01) and were 44.83짹0.62 and 42.53짹0.73, respectively, for diets 1 and 2. Milk urea concentrations (mg/dl) also varied (p 0.01) among the days of feeding period, but were stabilized after 10th day of feeding period. In contrast, diets and days of feeding period had no significant effect on percent milk protein. Plasma urea concentration showed a significant (p<0.01) positive correlation (r = 0.93) with MU concentration. To predict the PU from MU the following equation was developed `PU = 10.67짹0.76횞MU (mg/dl) with R2 = 0.87`. A clear diurnal variation of MU was found with lowered morning value (42.04짹0.68 mg/dl) than the evening value (45.32짹0.66 mg/dl). Present findings suggested that MU or PU concentration could be used as an indicator to monitor the feeding strategy. Plasma urea can be predicted from MU, whenever interpretation of milk urea data required consideration of diurnal variation.

        Monitoring Feeding Adequacy in Dairy Cows Using Milk Urea and Milk Protein Contents under Farm Condition

        The experiment was conducted on 264 crossbred Karan-Fries (Holstein Friesian${\times}$Tharparkar) cows, over one year to explore the possibility of using milk urea (MU) concentration and milk protein content to monitor feeding adequacy under farm condition and to investigate the effects of different animal factors and season on MU concentration. Individual noon (1200 to 1300 h) milk samples were collected once in every month and analysed for urea and protein contents. Representative feed samples were also collected on the same day of milk collection and were analysed for CP content. A significant positive association (p<0.01) between MU concentration and milk yield was observed. MU concentrations (mg/dl) were found to be significantly (p<0.01) higher and lower in first lactation (44.8${\pm}$0.7) and in early lactation stage (40.7${\pm}$0.5), respectively. Average MU values were found to be significantly (p<0.01) higher in winter (50.7${\pm}$0.3) and lower in summer (32.9${\pm}$0.6). During the investigation, of the total MU observations, 50.3% were within the range of 30 to 50 mg/dl, 21.4% were <30 mg/dl and only 7.5% were >60 mg/dl. MU concentration was found to be associated significantly (p<0.05) with CP content of forages rather than concentrate. A close positive association (p<0.01) between MU level and daily milk protein (DMP) yield was observed during the investigation. The regression equation, DMP yield (g) = -24.6+33.5 daily milk yield (kg) +0.9 MU (mg/dl) was developed to establish the reference level of DMP yield. The result indicates that the effect of parity and stage of lactation may be ignored while interpreting MU values. However, reference MU values may be standardised separately for high milk yielders as level of milk yield contributes significantly to the variation of MU. The study revealed that the MU values together with DMP yield and milk protein content could be used as a potential non-invasive pointer to monitor feeding adequacy in dairy cows under farm conditions.

        Influence of Milk Yield, Parity, Stage of Lactation and Body Weight on Urea and Protein Concentration in Milk of Murrah Buffaloes

        The present study was carried out to investigate the effect of test day milk yield, test day evening milk yield, parity, stage of lactation and body weight on milk urea and milk protein concentration. A total of 319 milk samples was collected from buffaloes over four month's period and subjected to urea and protein analysis. Milk urea concentration (mg/dl) was significantly (p<0.01) increased with increasing test day milk yield. The lowest value ($57.03{\pm}1.13$) was observed in the milk yield group ${\leq}4.5kg/day$ and the highest value ($64.15{\pm}1.13$) in the group 7.7-10.7 kg/day. However, test day evening milk yield had no significant effect on milk urea concentration. Milk protein did not vary significantly with the test day milk yield as well as test day evening milk yield. A clear decreasing trend of milk urea concentration (mg/dl) was found with the increasing parity. The highest MU concentration ($64.03{\pm}1.14$) was found in the first parity and the lowest ($55.67{\pm}1.22$) was found in the sixth and above parity. Whereas, stage of lactation had no effect on milk urea concentration. Moreover, parity and stage of lactation did not have any significant effect on milk protein concentration. Body weight (kg) was also found negatively (p<0.05) related with urea content (mg/dl) in milk. The highest mean MU concentration ($64.34{\pm}0.88$) was found when body weight was between 532 and 598 kg and lower mean values ($59.24{\pm}0.94$ and $59.33{\pm}1.23$) were observed in 599 to 665 kg and ${\geq}666kg$ group. Body weight also had significant (p<0.05) effect on milk protein content. The highest milk protein content (%) was found in ${\geq}666kg$ group and the lowest in <531 kg group. In conclusion, for proper interpretation of milk urea values to monitor protein nutrition status of the buffaloes parity, milk yield and body weight should be considered.

        Role of epigenetics in crop improvement: Water and heat stress

        Overall transcriptomic activity of an organism is determined not only by its genetic makeup but also by epigenetic regulations. Reprogramming by epigenetic modification triggered by various environmental challenges contributes to the phenotypic diversity and defense against these challenges. Different stresses acts as stimulus and changes genic expression level by various mechanisms namely DNA methylation, histone modifications and miRNA. Epigenetics is a continuously progressing branch since the past two decades. Improvement of crop varieties with traditional breeding methods is tedious, time consuming, expensive and is unable to meet progressive living standards and demand in today’s world whereas epigenetics holds great potential for the improvement of plant varieties with respect to yield and nutritional quality by creation of novel epialleles, transgenic RNAi. RNA interference has been used for the improvement of several crops like barley, wheat, soybean etc by reduction of various anti-nutritive factors or enhancement of flavor. Silencing of Omega–3 fatty acid desaturase gene is achieved using RNAi technology for the reduction of linoleic acid and improvement of flavor. HvTX1 from barley shows increased expression during drought stress indicating its active role in drought tolerance which further can be exploited for the development of improved crop variety with increased drought tolerance. Also multiple HAT genes in rice are reported to be actively expressed under heat and water stress. Studies of epigenetic responses to different stresses can increase our understanding of plant stress adaptation and mechanism underlying them which can be further exploited for the development of improved crop varieties.
