T nin(Persicae Semen) is the herb medicine that contains amygdalin as a major ingredient. Amygdalin in water is decomposed into benzaldehyde, HCN, and glucose by emulsin, a hydrolysis enzyme in t nin. A useful and practical method for the optimum extraction condition of amygdalin without enzymatic hydrolysis is required. The extraction yield of amygdalin of natural formula t nin was 0.1% from crude powders, 1.4% from small pieces, 3.5% from half pieces and 2.4% from whole pieces. The extraction yield of amygdalin of outer shell-eliminated t nin was 0.3% from crude powders, 1.4% from small pieces, and 3.5% from half pieces and whole pieces respectively. The extraction yield of amygdalin was most high when using the size larger than half.

Background: White ginseng consists of the roots and rhizomes of the Panax species, and red ginseng ismade by steaming and drying white ginseng. While red ginseng has both polar and nonpolar ginsenosides,previous studies showed white ginseng to have only polar ginsenosides. Because nonpolar ginsenosidesare formed through the manufacture of red ginseng from white ginseng, researchers havegenerally thought that nonpolar ginsenosides do not exist in white ginseng. Methods: We developed a simultaneous quantitative method for six nonpolar ginsenosides in whiteginseng using reverse-phase high-performance liquid chromatography coupled with integrated pulsedamperometric detection. The nonpolar ginsenosides of white ginseng were extracted for 4 h under refluxwith 50% methanol. Results: Using the gradient elution system, all target components were completely separated within 50min. Nonpolar ginsenosides were determined in the rhizome head (RH), main root (MR), lateral root, andhairy root (HR) of 6-year-old white ginseng samples obtained from several regions (Geumsan, Punggi,and Kanghwa). The total content in the HR of white ginseng was 37.8e56.8% of that in the HR of redginseng. The total content in the MR of white ginseng was 5.9e24.3% of that in the MR of red ginseng. Inaddition, the total content in the RH of white ginseng was 28.5e35.8% of that in the HR of red ginsengConclusion: It was confirmed that nonpolar ginsenosides known to be specific components of redginseng were present at substantial concentrations in the HR or RH of white ginseng.
