      • Incorporation of Eukaryotic Translation Initiation Factor eIF4E into Viral Nucleocapsids via Interaction with Hepatitis B Virus Polymerase

        <B>ABSTRACT</B><P>The DNA genome of hepatitis B virus (HBV) replicates via reverse transcription within capsids following the encapsidation of an RNA template, the pregenomic RNA (pgRNA). We previously demonstrated that the 5′ cap proximity of the stem-loop structure (ε or epsilon), an encapsidation signal, is critically important for the encapsidation of the pgRNA (J. K. Jeong, G. S. Yoon, and W. S. Ryu, J. Virol. 74:5502-5508, 2000). Therefore, we speculated that the viral polymerase (Pol), while bound to the 5′ ε stem-loop structure, could recognize the cap via its interaction with eIF4E, a eukaryotic translation initiation factor. Our data showed the direct interaction between HBV Pol and eIF4E, as measured by coimmunoprecipitation. Further, we demonstrated that eIF4E interacts with the Pol-ε ribonucleoprotein complex (RNP) rather than Pol alone, resulting in eIF4E-Pol-ε RNP complex formation. In addition, we asked whether eIF4E remains engaged to the Pol-ε RNP complex during nucleocapsid assembly. Density gradient analysis revealed that eIF4E indeed was incorporated into nucleocapsids. It is of great importance to uncover whether the incorporated eIF4E contributes to viral reverse transcription or other steps in the HBV life cycle.</P>

        가야산국립공원의 관속식물상

        The study on vascular plants of Gayasan National Park was conducted in 2016 over a period of 34 days from March to October 2016. As the study, a total of 768 taxa were recorded, including 118 families, 396 genera, 691 species, 15 subspecies, 51 varieties, and 11 forms. In the vascular plants, four endangered wild plants were recorded, Ponerorchis cucullata (L.) X.H.Jin, Trientalis europaea var. arctica (Fisch.) Ledeb., Cypripedium macranthos Sw., and Pedicularis hallaisanensis Hurus. Taxa of Least Concern (LC) or higher level on National Red List published by the Ministry of Environment of Korea were 21 taxa for example Tricyrtis macropoda Miq., and Lilium cernuum Kom. A total of 37 Korean endemic plants were investigated for example Abies koreana E.H.Wilson and Impatiens atrosanguinea (Nakai) B.U.Oh & Y.P.Hong. A total of 162 taxa of floristic regional indicators (3rd to 5th degree) were investigated including 9 taxa of V, 15 taxa of IV, 38 taxa of III, 45 taxa of II, and 53 taxa of I. Naturalized plants were recorded a total of 46 taxa with a Naturalization Index of 6.0%, the Urbanization Index was calculated to be 14.3% and 5 taxa of ecosystem disturbance plants were investigated such as Sicyos angulatus L., Rumex acetosella L., and Humulus scandens (Lour.) Merr. As the result of comparison with previous studies, in this study a total of 206 taxa were newly identified including 62 families, 144 genera, 184 species, 2 subspecies, 15 varieties, and 5 forms.

        Four Conserved Cysteine Residues of the Hepatitis B Virus Polymerase Are Critical for RNA Pregenome Encapsidation

        <B>ABSTRACT</B><P>Hepadnaviruses replicate via reverse transcription of an RNA template, the pregenomic RNA (pgRNA). Although hepadnaviral polymerase (Pol) and retroviral reverse transcriptase are distantly related, some of their features are distinct. In particular, Pol contains two additional N-terminal subdomains, the terminal protein and spacer subdomains. Since much of the spacer subdomain can be deleted without detrimental effects to hepatitis B virus (HBV) replication, this subdomain was previously thought to serve only as a spacer that links the terminal protein and reverse transcriptase subdomains. Unexpectedly, we found that the C terminus of the spacer subdomain is indispensable for the encapsidation of pgRNA. Alanine-scanning mutagenesis revealed that four conserved cysteine residues, three at the C terminus of the spacer subdomain and one at the N terminus of the reverse transcriptase subdomain, are critical for encapsidation. The inability of the mutant Pol proteins to incorporate into nucleocapsid particles, together with other evidence, argued that the four conserved cysteine residues are critical for RNA binding. One implication is that these four cysteine residues might form a putative zinc finger motif. Based on these findings, we speculate that the RNA binding activity of HBV Pol may be mediated by this newly identified putative zinc finger motif.</P>

        DDX3 DEAD-Box RNA Helicase Inhibits Hepatitis B Virus Reverse Transcription by Incorporation into Nucleocapsids

        <B>ABSTRACT</B><P>Viruses utilize host factors in many steps of their life cycles. Yet, little is known about host factors that contribute to the life cycle of hepatitis B virus (HBV), which replicates its genome by reverse transcription. To identify host factors that contribute to viral reverse transcription, we sought to identify cellular proteins that interact with HBV polymerase (Pol) by using affinity purification coupled with mass spectrometry. One of the HBV Pol-interacting host factors identified was DDX3 DEAD-box RNA helicase, which unwinds RNA in an ATPase-dependent manner. Recently, it was shown that DDX3 is essential for both human immunodeficiency virus and hepatitis C virus infection. In contrast, we found that the ectopic expression of DDX3 led to significantly reduced viral DNA synthesis. The DDX3-mediated inhibition of viral DNA synthesis did not affect RNA encapsidation, a step prior to reverse transcription, and indicated that DDX3 inhibits HBV reverse transcription. Mutational analysis revealed that mutant DDX3 with an inactive ATPase motif, but not that with an inactive RNA helicase motif, failed to inhibit viral DNA synthesis. Our interpretation is that DDX3 inhibits viral DNA synthesis at a step following ATP hydrolysis but prior to RNA unwinding. Finally, OptiPrep density gradient analysis revealed that DDX3 was incorporated into nucleocapsids, suggesting that DDX3 inhibits viral reverse transcription following nucleocapsid assembly. Thus, DDX3 represents a novel host restriction factor that limits HBV infection.</P>

        Depressive symptoms among dance artists in South Korea: balance between self- and social identity on job value

        Background: The balance between self- and social identity on job value of Korean dance artists, considered as performing artists, was measured to investigate the relationship between balance and depressive symptoms. Methods: Data of 139 dance artists were analyzed for depressive symptoms assessed by their responses to questionnaires from the Center for Epidemiologic Studies Depression (CESD) translated into Korean. The balance score of each aspect of social identity compared to that of self-identity on job value was measured and classified as lower, equal, or higher. Following a graphical approach, we performed locally weighted scatterplot smoothing algorithm, simple linear regression, and quadric equation regression. Results: The mean of CESD was highest in the higher balance group and lowest in the lower balance group. There was a linear relationship between balance score and CESD regarding specific audience, unspecified audience, and unspecified public. Balance score showed no association with other aspects of social identity. In the balance score range of 1 or higher, it was associated with CESD with respect to artist colleague, unspecified audience, and unspecified public. Conclusions: Dance artists should maintain a balance between self- and social identity in job value to prevent depressive symptoms.

        A multifunctional mesoporous nanocontainer with an iron oxide core and a cyclodextrin gatekeeper for an efficient theranostic platform

        <P>We report on the dual function of a silica–iron oxide hybrid nanoparticle combined with a stimulus responsive gatekeeper on the surface. Multifunctional hybrid nanoparticles (Fe@Si–DOX–CD–PEG) composed of an Fe<SUB>3</SUB>O<SUB>4</SUB> core, a mesoporous silica shell, and GSH-responsive CD gatekeepers are an excellent platform for anticancer drug delivery and MR imaging. In particular, the CD gatekeepers on the surface of the hybrid nanoparticle play a key role in accommodating anticancer drug molecules in the pore of the silica shell without premature release until CD gatekeepers are cleaved by GSH. We confirmed, from an <I>in vitro</I> study with the A549 cell line, that DOX was released from the internalized carriers due to GSH-mediated cleavage of the CD gatekeeper. Consequently, apoptotic and clonogenic death occurred in the cells treated with Fe@Si–DOX–CD–PEG. The accumulation of Fe@Si–DOX–CD–PEG in the tumors was detected by <I>in vivo</I> MR imaging. The growth of the tumor <I>in vivo</I> was effectively suppressed by the intravenously injected Fe@Si–DOX–CD–PEG.         Evaluation of Genetic Diversity and Population Structure Analysis among Germplasm of Agaricus bisporus by SSR Markers

        Agaricus bisporus is a popular edible mushroom that is cultivated worldwide. Due to its secondary homothallic nature, cultivated A. bisporus strains have low genetic diversity, and breeding novel strains is challenging. The aim of this study was to investigate the genetic diversity and population structure of globally collected A. bisporus strains using simple sequence repeat (SSR) markers. Agaricus bisporus strains were divided based on genetic distance- based groups and model-based subpopulations. The major allele frequency (MAF), number of genotypes (NG), number of alleles (NA), observed heterozygosity (HO), expected heterozygosity (HE), and polymorphic information content (PIC) were calculated, and genetic distance, population structure, genetic differentiation, and Hardy-Weinberg equilibrium (HWE) were assessed. Strains were divided into two groups by distance-based analysis and into three subpopulations by model-based analysis. Strains in subpopulations POP A and POP B were included in Group I, and strains in subpopulation POP C were included in Group II. Genetic differentiation between strains was 99%. Marker AB-gSSR-1057 in Group II and subpopulation POP C was confirmed to be in HWE. These results will enhance A. bisporus breeding programs and support the protection of genetic resources.

        기기별 전력 모니터링 시스템 개발에 관한 연구

        본 연구에서는 일반 가정에서 Smart Grid를 통하여 누릴 수 있는 서비스를 발굴하고, 이러한 서비스를 구현할 수 있는 기기별 전력 모니터링 시스템을 구현하였으며, 모니터링한 데이터를 분석하였다. 기기별 전력 사용현황을 수집하고 이를 분석함으로써, 모니터링 시스템에 표시되고 있는 전력사용현황이 어떤 기기의 전력사용현황인지 사전에 정의된 프로토콜 없이 알 수 있다. 나아가서 전력 절감 방법 제시, 지능적인 대기전력 차단 및 복구, 기기의 이상여부 감시와 같은 서비스를 구현할 수 있을 것이다. 본 연구에서는 이러한 시나리오의 실현 가능성을 점검하기 위해 기기별 전력 모니터링 시스템을 개발하여 가정 내 전력소비의 대부분을 차지하는 냉장고, 전기밥솥, TV, 세탁기 샘플에 대하여 전력소비패턴을 수집하여 database화 하였다. 또한 이 database를 분석하여 제시한 서비스들을 구현하는데 필요한 기본 기능인 기기식별이 전력사용패턴분석을 통하여 가능함을 보였다. This paper presents ideas of service scenarios for home residents using electric power monitoring system per appliance, the implementation of the monitoring system, and analysis of acquired electric power usage pattern. By acquiring and analyzing electric power usage pattern, home residents can get information of power usage pattern of every legacy (non-Demand Response-ready) appliance. Further they can get pieces of recommendation how to reduce energy consumption, intelligent standby power blocking service, and alarming service to abnormality of appliances. In order to check the feasibility of the ideas, a system that can acquire electric power pattern per appliance is implemented, and electric power pattern of some appliances are stored to a database and it was analyzed to show if auto-identification of a type of a device is possible, which is a basic required function for the scenarios presented.
